A simple and highly efficient protocol for 13 C-labeling of plant cell wall for structural and quantitative analyses via solid-state nuclear magnetic resonance

Plant cell walls are made of a complex network of interacting polymers that play a critical role in plant development and responses to environmental changes. Thus, improving plant biomass and fitness requires the elucidation of the structural organization of plant cell walls in their native environment. The 13 C-based multi-dimensional solid-state nuclear magnetic resonance (ssNMR) has been instrumental in revealing the structural information of plant cell walls through 2D and 3D correlation spectral analyses. However, the requirement of enriching plants with 13 C limits the applicability of this method. To our knowledge, there is only a very limited set of methods currently available that achieve high levels of 13 C-labeling of plant materials using 13 CO 2 , and most of them require large amounts of 13 CO 2 in larger growth chambers. In this study, a simplified protocol for 13C-labeling of plant materials is introduced that allows ca 60% labeling of the cell walls, as quantified by comparison with commercially labeled samples. This level of 13 C-enrichment is sufficient for all conventional 2D and 3D correlation ssNMR experiments for detailed analysis of plant cell wall structure. The protocol is based on a convenient and easy setup to supply both 13 C-labeled glucose and 13 CO 2 using a vacuum-desiccator. The protocol does not require large amounts of 13 CO 2 . This study shows that our 13 C-labeling of plant materials can make the accessibility to ssNMR technique easy and affordable. The derived high-resolution 2D and 3D correlation spectra are used to extract structural information of plant cell walls. This helps to better understand the influence of polysaccharide-polysaccharide interaction on plant performance and allows for a more precise parametrization of plant cell wall models.

Bull Trout and Westslope Cutthroat Trout movement in a dam tailrace

Populations of Bull Trout Salvelinus confluentus and Westslope Cutthroat Trout Oncorhynchus clarkii lewisi in the Pend Oreille Basin have declined, partly due to fragmentation caused by hydropower dams. This study aimed to analyze the movements and behavior of these species downstream of Albeni Falls Dam over two years to inform fishway design. Radiotracking investigations were conducted to monitor 10 adult Bull Trout and 17 adult Westslope Cutthroat Trout from September 2008 to July 2010. Macro and micro detection zones were delineated to study fine- and large-scale movements in the tailrace. Macro zones included the spillway and powerhouse tailrace areas. Micro zones were nested within macro zones to identify regions close to the dam where fishway structures could be built. Both Bull Trout and Westslope Cutthroat Trout exhibited high mobility in the dam tailrace, transitioning between macro detection zones. Seasonal variations influenced their distribution patterns. During the spring freshet migration season, both species were primarily detected at the left powerhouse micro zone. In sedentary periods (fall/winter and summer), fish actively swam throughout the tailrace, displaying search behavior. These behavior observations suggest that potential fishway entrances located near dam concrete would be effective: the area near the left powerhouse was identified as the optimal construction location. This study highlights the feasibility of designing fish passage structures to mitigate population fragmentation and support species recovery.

Congestion Management Solutions for Enhanced Distribution System Operations with Aggregated Distribution Grid Resources Providing Grid Services and Market Participation

Microgrids and other aggregations of distribution grid resources (DGRs) are poised to actively participate in electricity markets and provide essential grid services in the coming years. In fact, DGRs already play such a role through behind-the-meter (BTM) demand response programs and small-scale BTM dispatchable generation initiatives. At the same time, the rapid growth of artificial intelligence (AI) and cryptocurrency datacenters imposes significant, often unpredictable, demands on the power distribution system. Aggregated DGRs can serve as flexible resources that help mitigate these pressures by using available transmission and distribution capacity more efficiently, supporting resource adequacy and other reliability services, and providing bridge strategies while long-term transmission infrastructure is being developed. The impacts this activity will have on distribution networks are not fully understood and could present significant challenges for distribution utilities due to capacity constraints and the need for congestion management. Technical issues include reverse power flow, variability and possible degradation of equipment integrity, voltage violations, and customer power quality concerns. These issues will likely intensify as electricity market operators across the United States implement Federal Energy Regulatory Commission Order 2222 over the next few years.
