Structural and functional ramifications of antigenic drift in recent SARS-CoV-2 variants

Defenses against SARS-CoV-2 variants Our key defense against the COVID-19 pandemic is neutralizing antibodies against the SARS-CoV-2 virus elicited by natural infection or vaccination. Recent emerging viral variants have raised concern because of their potential to escape antibody neutralization. Wang et al . identified four antibodies from early-outbreak convalescent donors that are potent against 23 variants, including variants of concern, and characterized their binding to the spike protein of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Yuan et al . examined the impact of emerging mutations in the receptor-binding domain of the spike protein on binding to the host receptor ACE2 and to a range of antibodies. These studies may be helpful for developing more broadly effective vaccines and therapeutic antibodies. —VV

Language models for the prediction of SARS-CoV-2 inhibitors

The COVID-19 pandemic highlights the need for computational tools to automate and accelerate drug design for novel protein targets. We leverage deep learning language models to generate and score drug candidates based on predicted protein binding affinity. We pre-trained a deep learning language model (BERT) on ∼9.6 billion molecules and achieved peak performance of 603 petaflops in mixed precision. Our work reduces pre-training time from days to hours, compared to previous efforts with this architecture, while also increasing the dataset size by nearly an order of magnitude. For scoring, we fine-tuned the language model using an assembled set of thousands of protein targets with binding affinity data and searched for inhibitors of specific protein targets, SARS-CoV-2 Mpro and PLpro. We utilized a genetic algorithm approach for finding optimal candidates using the generation and scoring capabilities of the language model. Our generalizable models accelerate the identification of inhibitors for emerging therapeutic targets.

An introduction to the interim digital SAR processor and the characteristics of the associated Seasat SAR imagery

Basic engineering data regarding the Interim Digital SAR Processor (IDP) and the digitally correlated Seasat synthetic aperature radar (SAR) imagery are presented. The correlation function and IDP hardware/software configuration are described, and a preliminary performance assessment presented. The geometric and radiometric characteristics, with special emphasis on those peculiar to the IDP produced imagery, are described.

Plans for the development of EOS SAR systems using the Alaska SAR facility

The Alaska SAR Facility (ASF) program for the acquisition and processing of data from the ESA ERS-1, the NASDA ERS-1, and Radarsat and to carry out a program of science investigations using the data is introduced. Agreements for data acquisition and analysis are in place except for the agreement between NASA and Radarsat which is in negotiation. The ASF baseline system, consisting of the Receiving Ground System, the SAR Processor System and the Archive and Operations System, passed critical design review and is fully in implementation phase. Augments to the baseline system for systems to perform geophysical processing and for processing of J-ERS-1 optical data are in the design and implementation phase. The ASF provides a very effective vehicle with which to prepare for the Earth Observing System (EOS) in that it will aid the development of systems and technologies for handling the data volumes produced by the systems of the next decades, and it will also supply some of the data types that will be produced by EOS.

Alaska SAR Facility (ASF5) SAR Communications (SARCOM) Data Compression System

The real-time operational requirements for SARCOM translation into a high speed image data handler and processor to achieve the desired compression ratios and the selection of a suitable image data compression technique with as low as possible fidelity (information) losses and which can be implemented in an algorithm placing a relatively low arithmetic load on the system are described.

Revision to Global Persistent SAR Sampling with the NASA-ISRO SAR (NISAR) Mission

Abstract—The National Aeronautics and Space Administration (NASA) in the United States and the Indian Space Research Organisation (ISRO) are developing an Earth-orbiting science and applications mission that will exploit synthetic aperture radar to map Earth’s surface every 12 days, persistently on ascending and descending portions of the orbit, over all land and ice-covered surfaces. The mission’s primary objectives will be to study Earth land and ice deformation, and ecosystems, in areas of common interest to the US and Indian science communities. This single spacecraft solution with an L-band (24 cm wavelength) and S-band (10 cm wavelength) radar has a swath of over 240 km at fine resolution, using full polarimetry where needed, uses a reflector-feed system whereby the feed aperture elements are individually sampled to allow a scan-on-receive (“SweepSAR”) capability at both L-band and S-band. This design is in contrast to recent concepts towards large constellations of smaller radar satellites, and is driven by the science requirements for complete coverage over the 12-day repeat cycle, using repeat pass interferometry and polarimetry to measure deformation and surface properties. A single spacecraft with enough aperture, power, duty cycle, and downlink capacity was determined to be a more practical and implementable solution that multiple smaller spacecraft. The use of a single large aperture reflector for both the L- and Sband radars enables both to have comparable performance, leading to overall development and operational efficiencies.
