Project: Corbomite - Product: ConsoleWorks REACT

TDi Technologies presents ConsoleWorks REACT, an advanced platform designed to tackle the complexities of cyber and operational risk assessment. This comprehensive solution goes beyond asset-focused approaches by considering the impact of both assets and people have on the security and operation of critical infrastructure, specifically targeting preventing gird mis-operation by evaluating real-time human interaction or commands with critical assets. The hypothesis suggests that by integrating the assessment of user commands into the overall risk assessment process, organizations can make more informed decisions, prioritize resources effectively, and respond promptly to potential threats. This hypothesis forms the basis for the development of a proactive and holistic risk management approach that is more comprehensive and context aware than traditional models which only look at assets, patch levels, configuration and threat intel by collecting that information off the network vs directly from the asset, human, and human interaction all in real-time. ConsoleWorks' unique man-in-the-middle architecture is a key feature that sets it apart in the cybersecurity landscape. This architecture enables the real-time observation, enforcement, and commands or interaction risk transparency of user interaction with critical infrastructure to be risk mitigated and audited as they are aggregated with device and human risk factors for a more comprehensive risk threat score across a device or group of devices. This architecture allows ConsoleWorks to act as a secure intermediary between users and critical assets, monitoring all interactions and ensuring that only authorized commands are sent to the asset to be executed. This not only enhances security but also provides a comprehensive audit trail of all user activities, contributing to compliance efforts and facilitating incident investigation and risk management. By integrating this unique architecture with our comprehensive risk assessment methodology, ConsoleWorks REACT provides a powerful solution for managing cyber and operational risks, enabling organizations to maintain a robust security posture and effectively mitigate potential threats. To that end, the primary objective of this project was to research and develop a robust solution that enables the energy industry to mitigate the risks associated with human actions that can compromise the security or operations of assets critical to energy delivery, generation, transmission and operation. ConsoleWorks REACT plays a pivotal role in achieving this goal by leveraging its unique capabilities to monitor and track all user activity. Through its Zero Trust approach, which emphasizes continuous verification, the platform ensures secure access to assets and serves as the centralized human response and notification platform for addressing cyber and operational issues.

Advanced Reactor Safeguards & Security Program: Cybersecurity Scenarios

The use of digital control systems and automation in advanced nuclear power systems introduces different types of vulnerabilities compared to legacy (i.e. analog) control systems that cyber adversaries can exploit. These vulnerabilities pose a challenge to reactor operators and cyber operations staff due to the dynamic nature of the event in which a human response or a lack of response can potentially evolve into a worsening plant condition. Using the Department of Homeland Security Cyber and Infrastructure Security Agency’s (CISA) critical infrastructure exercise framework, this document presents several cyber security scenarios typical of digital control systems that could be used in advanced reactor designs. These scenarios can be used in tabletop exercises to evaluate cyber security posture or conduct training on different aspects of cyber security, including detection, threat hunting using indicators of compromise, evaluating incident response, risk mitigation, incident reporting, information sharing and recovery.

Evolving theories of change: Rethinking of the effects of office lighting on occupants

This paper proposes an overall theory of change that relates lighting in offices to a holistic view of human responses that contribute to the well-being of office workers. The theoretical model considers how variations in the four fundamental lighting attributes affect visual performance, visual environment, and non-image forming responses. It moves beyond these lighting to address how external factors that are may mediate the effects on the desired lighting related outcomes, which in turn determine the final impact on occupants. The model builds on prior conceptual models of lighting effects and informs design and research planning for office lighting. Ultimately, properly supported task performance can increase occupant motivation but by itself may not achieve the holistic desired impact. Lighting attributes that support a pleasant visual environment and appropriate non-image forming responses can more fully contribute to an occupant’s mood and overall well-being.

A compendium of multi-omics data illuminating host responses to lethal human virus infections

Human infections caused by viral pathogens trigger a complex gamut of host responses that limit disease, resolve infection, generate immunity, and contribute to severe disease or death. Here, we present experimental methods and multi-omics data capture approaches representing the global host response to infection generated from 45 individual experiments involving human viruses from the Orthomyxoviridae, Filoviridae, Flaviviridae, and Coronaviridae families. Analogous experimental designs were implemented across human or mouse host model systems, longitudinal samples were collected over defined time courses, and global multi-omics data (transcriptomics, proteomics, metabolomics, and lipidomics) were acquired by microarray, RNA sequencing, or mass spectrometry analyses. For comparison, we have included transcriptomics datasets from cells treated with type I and type II human interferon. Raw multi-omics data and metadata were deposited in public repositories, and we provide a central location linking the raw data with experimental metadata and ready-to-use, quality-controlled, statistically processed multi-omics datasets not previously available in any public repository. This compendium of infection-induced host response data for reuse will be useful for those endeavouring to understand viral disease pathophysiology and network biology.

Microcystin-LR aerosol induces inflammatory responses in healthy human primary airway epithelium

Harmful algal blooms plague bodies of freshwater globally. These blooms are often composed of outgrowths of cyanobacteria capable of producing the heptapeptide Microcystin-LR (MC-LR) which is a well-known hepatotoxin. Recently, MC-LR has been detected in aerosols generated from lake water. However, the risk for human health effects due to MC-LR inhalation exposure have not been extensively investigated. In this study, we exposed a fully differentiated 3D human airway epithelium derived from 14 healthy donors to MC-LR-containing aerosol for 3 minutes per day for 3 days. Concentrations of MC-LR ranged from 100 pM to 1 µM. Although there were little to no detrimental alterations in measures of the airway epithelial function (i.e. cell survival, tissue integrity, mucociliary clearance, or cilia beating frequency), a distinct shift in the transcriptional activity was found. Genes related to inflammation were found to be upregulated such as C-C motif chemokine 5 (CCL5; log2FC = 0.56 , p = 0.02) and C-C chemokine receptor type 7 (CCR7; log2FC = 0.83, p = 0.03). Functionally, conditioned media from MC-LR exposed airway epithelium was also found to have significant chemo-attractive properties for primary human neutrophils. Additionally, increases were found in the concentration of secreted chemokine proteins in the conditioned media such as CCL1 (log2FC = 5.07 , p = 0.0001) and CCL5 (log2FC = 1.02, p = 0.046). These results suggest that MC-LR exposure to the human airway epithelium is capable of inducing an inflammatory response that may potentiate acute or chronic disease.

Capsular polysaccharide correlates with immune response to the human gut microbe Ruminococcus gnavus

Active inflammatory bowel disease (IBD) often coincides with increases of Ruminococcus gnavus, a gut microbe found in nearly everyone. It was not known how, or if, this correlation contributed to disease. We investigated clinical isolates of R. gnavus to identify molecular mechanisms that would link R. gnavus to inflammation. Here, we show that only some isolates of R. gnavus produce a capsular polysaccharide that promotes a tolerogenic immune response, whereas isolates lacking functional capsule biosynthetic genes elicit robust proinflammatory responses in vitro. Germ-free mice colonized with an isolate of R. gnavus lacking a capsule show increased measures of gut inflammation compared to those colonized with an encapsulated isolate in vivo. These observations in the context of our earlier identification of an inflammatory cell-wall polysaccharide reveal how some strains of R. gnavus could drive the inflammatory responses that characterize IBD.

The human plasma lipidome response to exertional heat tolerance testing

The year of 2023 displayed the highest average global temperatures recorded in history— the duration and severity of extreme heat are projected to increase. Rising global temperatures represent a major public health threat, especially to occupations exposed to hot environments, such as construction and agricultural workers, and first responders. Despite efforts of the scientific community, there is still a need to characterize the pathophysiological processes leading to heat related illness and develop biomarkers that can predict its onset. Here, we performed a plasma lipidomic analysis on male and female subjects who underwent heat tolerance testing (HTT), consisting of a 2-h treadmill walk at 5 km/h with 2% inclination at a controlled temperature of 40oC. We identified 995 lipids from 27 classes, with nearly half of all detected lipids being responsive to HTT. Lipid classes related to substrate utilization were predominantly affected by HTT, with a downregulation of triacylglycerols and upregulation of free fatty acids and acyl-carnitines. We additionally examined correlations between changes in plasma lipids by using the even chain acyl-carnitines is physiological strain index (PSI). Here, even chain acyl-carnitines, bi-products of incomplete beta oxidation, and diacylglycerols displayed the highest correlation to PSI. PSI did not correlate with plasma lactate levels, which suggests that correlations with related to metabolic efficiency versus physical exertion. Overall, our results show that HTT has a strong impact on the plasma lipidome and that incomplete beta oxidation may underlie heat intolerance.

A Mathematical Model of the Dynamics of Cytokine Expression and Human Immune Cell Activation in Response to the Pathogen Staphylococcus aureus

Cell-based mathematical models have previously been developed to simulate the immune system in response to pathogens. Mathematical modeling papers which study the human immune response to pathogens have predicted concentrations of a variety of cells, including activated and resting macrophages, plasma cells, and antibodies. This study aims to create a comprehensive mathematical model that can predict cytokine levels in response to a gram-positive bacterium, S. aureus by coupling previous models. To accomplish this, the cytokines Tumor Necrosis Factor Alpha (TNF-α), Interleukin 6 (IL-6), Interleukin 8 (IL-8), and Interleukin 10 (IL-10) are included to quantify the relationship between cytokine release from macrophages and the concentration of the pathogen, S. aureus, ex vivo. Partial differential equations (PDEs) are used to model cellular response and ordinary differential equations (ODEs) are used to model cytokine response, and interactions between both components produce a more robust and more complete systems-level understanding of immune activation. In the coupled cellular and cytokine model outlined in this paper, a low concentration of S. aureus is used to stimulate the measured cellular response and cytokine expression. Results show that our cellular activation and cytokine expression model characterizing septic conditions can predict ex vivo mechanisms in response to gram-negative and gram-positive bacteria. Our simulations provide new insights into how the human immune system responds to infections from different pathogens. Novel applications of these insights help in the development of more powerful tools and protocols in infection biology.

Broad and potent neutralizing human antibodies to tick-borne flaviviruses protect mice from disease

Tick-borne encephalitis virus (TBEV) is an emerging human pathogen that causes potentially fatal disease with no specific treatment. Mouse monoclonal antibodies are protective against TBEV, but little is known about the human antibody response to infection. Here, we report on the human neutralizing antibody response to TBEV in a cohort of infected and vaccinated individuals. Expanded clones of memory B cells expressed closely related anti-envelope domain III (EDIII) antibodies in both groups of volunteers. However, the most potent neutralizing antibodies, with IC50s below 1 ng/ml, were found only in individuals who recovered from natural infection. These antibodies also neutralized other tick-borne flaviviruses, including Langat, louping ill, Omsk hemorrhagic fever, Kyasanur forest disease, and Powassan viruses. Structural analysis revealed a conserved epitope near the lateral ridge of EDIII adjoining the EDI–EDIII hinge region. Prophylactic or early therapeutic antibody administration was effective at low doses in mice that were lethally infected with TBEV.
