Super-resolution image display using diffractive decoders

High-resolution image projection over a large field of view (FOV) is hindered by the restricted space-bandwidth product (SBP) of wavefront modulators. We report a deep learning–enabled diffractive display based on a jointly trained pair of an electronic encoder and a diffractive decoder to synthesize/project super-resolved images using low-resolution wavefront modulators. The digital encoder rapidly preprocesses the high-resolution images so that their spatial information is encoded into low-resolution patterns, projected via a low SBP wavefront modulator. The diffractive decoder processes these low-resolution patterns using transmissive layers structured using deep learning to all-optically synthesize/project super-resolved images at its output FOV. This diffractive image display can achieve a super-resolution factor of ~4, increasing the SBP by ~16-fold. We experimentally validate its success using 3D-printed diffractive decoders that operate at the terahertz spectrum. This diffractive image decoder can be scaled to operate at visible wavelengths and used to design large SBP displays that are compact, low power, and computationally efficient.

Energy Efficiency and Performance Evaluation of Self-Contained, Medium Temperature, Reach-In Refrigerated Display Cases

This project was part of an effort by Commonwealth Edison Company (ComEd) to evaluate the energy and peak demand saving potential of emerging technologies in the Chicago area. This document focuses on the assessment of energy-efficient, medium-temperature, self-contained refrigerated display cases utilizing environmentally friendly refrigerants. The results of this evaluation will be considered by CLEAResult to develop a new energy efficiency rebate measure for ComEd's incentive programs. This rebate measure will become an addition to the Technical Reference Manual (TRM). In 2016, the United Nations passed the Kigali Montreal Protocol Amendment which placed restrictions on certain types of refrigerants. In compliance with this amendment, the US Environmental Protection Agency (EPA) placed a ban on the manufacture of refrigeration systems using hydrofluorocarbons including R134a starting in January 2020. Although the ban has halted manufacture, the EPA continues to allow the use of these refrigerants. Therefore, it is critical to provide incentives for replacing these refrigerants with other environmentally friendly and energy-efficient alternatives. The energy-efficient refrigerator cases evaluated here (referred to as EE Case A and B) contain environmentally-friendly refrigerants in compliance with the EPA hydrofluorocarbon ban. These consist of natural refrigerant propane (R290), and HFC drop-in hydrofluoroolefin R513a, respectively. These cases also contain other energy-efficient components including efficient lighting, more robust evaporator and condenser fans and different-sized heat exchangers. EE Case A is also built with materials that better insulate the case, which improves energy efficiency. To ascertain the energy efficiency contribution of these design components, the consumption of the evaporator and condenser fan motors, compressor, and lighting/controller were evaluated individually. The medium-temperature, self-contained reach-in refrigerated display case was selected due to its widespread use in convenience stores and small supermarkets. Self-contained refrigeration has also seen increased use in restaurants due to curbside pickup during the COVID-19 pandemic. For this study, the refrigerated display cases' performance was evaluated in a controlled environmental chamber at representative indoor dry-bulb and humidity conditions found in supermarkets within ComEd's service territory climate zone. The test method used in this project was foundationally inspired by the ANSI/ASHRAE 72-2018 method of testing. However, modifications to the ANSI/ASHRAE test methodology were implemented to better represent customer operation of the units. In addition to the indoor supermarket conditions, the cases were also evaluated at the "upper target," or environmental conditions used in ANSI/ASHRAE 72-2018. The case total power and case components were metered to obtain their daily power (kW) and energy consumption(kWh). The cases were filled with thermal filler mass to replicate thermal mass of product loading. Additionally, product simulators were used to provide product temperature information. Door actuators were mounted to each of the cases' three doors to replicate regular door openings and effects of shopper traffic.

Development of a cell surface display system in Chlamydomonas reinhardtii

Cell-surface display systems are biotechnological techniques used to express heterologous proteins on the cell surface. Their application depends directly on the cell system used, as well as on the anchoring point for the surface displayed protein. To meet most application demands an inexpensive, safe, and scalable production platform, that reduces the economic barriers for large scale use is needed. Toward this goal, we screened three possible cell surface anchoring points in the green algae Chlamydomonas by fusing mVenus to prospective anchors moieties. The vectors harboring mVenus:anchor were screened for mVenus fluorescence and tested for cellular localization by confocal laser scanning microscopy. This strategy allowed the identification of two functional anchors, one for the cytoplasmic membrane using the MAW8 GPI-anchor signal, and one for the cell wall using the GP1 protein. We also exploited GP1 chemical and biological traits to release the fused proteins efficiently during cell wall shedding. Our work provides a foundation for surface engineering of C reinhardtii supporting both cell biology studies and biotechnology applications.

Rapid identification of neutralizing antibodies against SARS-CoV-2 variants by mRNA display

The increasing prevalence of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) variants with the ability to escape existing humoral protection conferred by previous infection and/or immunization necessitates the discovery of broadly reactive neutralizing antibodies (nAbs). Utilizing mRNA display, we identify a set of antibodies against SARS-CoV-2 spike (S) proteins and characterize the structures of nAbs that recognize epitopes in the S1 subunit of the S glycoprotein. These structural studies reveal distinct binding modes for several antibodies, including the targeting of rare cryptic epitopes in the receptor-binding domain (RBD) of S that interact with angiotensin-converting enzyme 2 (ACE2) to initiate infection, as well as the S1 subdomain 1. Further, we engineer a potent ACE2-blocking nAb to sustain binding to S RBD with the E484K and L452R substitutions found in multiple SARS-CoV-2 variants. We demonstrate that mRNA display is an approach for the rapid identification of nAbs that can be used in combination to combat emerging SARS-CoV-2 variants.

Experimental characterization and analysis of phase change material-based thermal energy storage system for refrigerated display case

Refrigerated display cases that are used to store and exhibit food products in supermarkets and retail spaces consume a significant portion of the buildings’ total electricity. Importantly, the refrigeration-related energy cost and demand charges are greatly affected by the time-of-use electricity pricing and demand rates, which are at their maximum during peak hours, typically when refrigeration energy consumption is also high. Using energy storage to shift the refrigeration load from peak to off-peak hours can greatly reduce the operational costs in the supermarket. This study demonstrates a phase change material-based thermal energy storage (TES) system, specifically designed in stackable units, that can be integrated with an open vertical refrigerated display case. We perform numerical and experimental characterization that includes finite-difference modeling for the TES, prototype fabrication, and laboratory evaluation, followed by a preliminary system-level analysis to predict the impact of TES on the refrigerated case performance, energy use, and energy cost. The results show that the dedicated latent TES for refrigerated cases can provide a specific energy of 50.4 Wh/kg and a specific power of 15.5 W/kg. The TES can be charged during 12 h of the off-peak period and discharged at various rates during 4 to 6 h of the peak period, thereby shifting the refrigeration load from the peak to the off-peak period. Consequently, annual cost savings up to 19% can be achieved, depending on the thermal load, the summer/winter peak electricity pricing, and the transition temperature of the phase change material used.

Multiplex Evolution of Antibody Fragments Utilizing a Yeast Surface Display Platform

Advances in high-throughput synthetic biology technologies based on the CRISPR/Cas9 system have enabled a comprehensive assessment of mutations conferring desired phenotypes, as well as a better understanding of genotype-phenotype correlations in protein engineering. Engineering antibodies to enhance properties such as binding affinity and stability plays an essential role in therapeutic applications. Here we report a method, multiplex navigation of antibody structure (MINAS), that combines a CRISPR/Cas9-based trackable editing method and fluorescent-activated cell sorting (FACS) of yeast-displayed libraries. We designed mutations in all of the complementarity-determining and framework regions of a well-characterized scFv antibody and mapped the contribution of these regions to enhanced properties. We identified specific mutants that showed higher binding affinities up to 100-fold compared to the wild-type. This study expands the applicability of CRISPR/Cas9-based trackable protein engineering by combining it with a surface display platform.

Realizing Wide-Gamut Human-Centric Display Lighting with K 3 AlP 3 O 9 N:Eu 2+

Abstract Computers, televisions, and smartphones are revolutionized by the invention of InGaN blue light-emitting diode (LED) backlighting. Yet, continual exposure to the intense blue LED emission from these modern displays can cause insomnia and mood disorders. Developing “human-centric” backlighting that uses a violet-emitting LED chip and a trichromatic phosphor mixture to generate color images is one approach that addresses this problem. The challenge is finding a blue-emitting phosphor that possesses a sufficiently small Stokes’ shift to efficiently down-convert violet LED light and produce a narrow blue emission. This work reports a new oxynitride phosphor that meets this demand. K 3 AlP 3 O 9 N:Eu 2+ exhibits an unexpectedly narrow (45 nm, 2206 cm -1 ), thermally robust, and efficient blue photoluminescence upon violet excitation. Computational modeling and temperature-dependent optical property measurements reveal that the narrow emission arises from a rare combination of preferential excitation and site-selective quenching. The resulting chromaticity coordinates of K 3 AlP 3 O 9 N:Eu 2+ lie closer to the vertex of the Rec. 2020 than a blue LED chip and provides access to ≈10% more colors than a commercial tablet when combined with commercial red- and green-emitting phosphors. Alongside the wide gamut, tuning the emission from the violet LED and phosphor blend can reduce blue light emissions to produce next-generation, human-centric displays.

Using phage display for rational engineering of a higher-affinity humanized 3’ phosphohistidine-specific antibody

Abstract Histidine phosphorylation is a non-canonical post-translational modification (PTM), with 1-phosphohistidine (1-pHis) and 3-phosphohistidine (3-pHis) isoforms, that is understudied due to a lack of robust reagents, including high-affinity pHis-specific antibodies. Engineering pHis antibodies is challenging due to the labile nature of its phosphoramidate (P-N) bond. We developed a strategy for in vitro engineering of antibodies for the detection of native 3-pHis targets, in which the rabbit SC44-8 anti-3-pTza mAb is humanized into a scaffold (hSC44) that is suitable for phage display. Six unique Fab phage-displayed hSC44 scaffold libraries were screened for antibodies that bound 3-pHis with higher affinity and had specificity for 3-pHis versus 3-pTza. hSC44.20N32F L , the best engineered antibody, has ~10-fold higher affinity for 3-pHis than parental hSC44. Eleven new Fab structures, including the first antibody-pHis peptide structures, together with structural and quantum mechanical calculations, provided molecular insights into 3-pHis and 3-pTza discrimination by hSC44.20N32F L and the increased affinity obtained through engineering. We demonstrated the utility of these high-affinity 3-pHis-specific antibodies for the recognition of pHis proteins in mammalian cells by immunoblotting and immunofluorescence staining. Our work describes a general method for engineering labile PTM-specific antibodies and provides novel antibodies for investigating the role of 3-pHis in cell biology.

Markers of mitochondrial function and oxidative metabolism in skeletal muscle do not display intrinsic circadian regulation in female mice

Mitochondria are key regulators of metabolism and ATP supply in skeletal muscle, while circadian rhythms influence many physiological processes. However, whether mitochondrial function is intrinsically regulated in a circadian manner in mouse skeletal muscle is inadequately understood. Accordingly, we measured postabsorptive transcript abundance of markers of mitochondrial autophagy, dynamics, and metabolism [extensor digitorum longus (EDL), soleus, gastrocnemius], protein abundance of electron transport chain complexes (EDL and soleus), enzymatic activity of succinate dehydrogenase (tibialis anterior and plantaris), and maximal mitochondrial respiration (tibialis anterior) in different skeletal muscles from female C57BL/6NJ mice at four zeitgeber times: 1, 7, 13, and 19. Our findings demonstrate that markers of mitochondrial function and oxidative metabolism do not display intrinsic time-of-day regulation at the gene, protein, enzymatic, or functional level. The core-clock genes Bmal1 and Dbp exhibited intrinsic circadian rhythmicity in skeletal muscle (i.e., EDL, soleus, gastrocnemius) and circadian amplitude varied by muscle type. These findings demonstrate that female mouse skeletal muscle does not display circadian regulation of markers of mitochondrial function or oxidative metabolism over 24 h.

Towards a Public Event Display for DUNE

The Deep Underground Neutrino Experiment (DUNE) is a next generation long baseline neutrino experiment based at Fermilab, with a near detector near the beam target and a Far Detector (FD) in South Dakota. As the experiment prepares for its first data runs, creating pathways for public engagement and data transparency is essential. We present the first-ever DUNE event display designed for public outreach and education. Developed using data from the ProtoDUNE detectors at the CERN Neutrino Platform, this tool provides an intuitive and interactive interface that allows non-experts to visualise and explore particle interactions in a Liquid Argon Time Projection Chamber (LArTPC). By translating raw experimental data into a browser-accessible format, we establish the essential infrastructure for DUNE’s pathway to open data. This talk will detail the technical development of the display, its current implementation with ProtoDUNE data, and the strategic roadmap for integrating it into DUNE’s long-term open-access framework.

A genomic analysis reveals the diversity of cellulosome displaying bacteria

Introduction Several species of cellulolytic bacteria display cellulosomes, massive multi-cellulase containing complexes that degrade lignocellulosic plant biomass (LCB). A greater understanding of cellulosome structure and enzyme content could facilitate the development of new microbial-based methods to produce renewable chemicals and materials. Methods To identify novel cellulosome-displaying microbes we searched 305,693 sequenced bacterial genomes for genes encoding cellulosome proteins; dockerin-fused glycohydrolases (DocGHs) and cohesin domain containing scaffoldins. Results and discussion This analysis identified 33 bacterial species with the genomic capacity to produce cellulosomes, including 10 species not previously reported to produce these complexes, such asAcetivibrio mesophilus. Cellulosome-producing bacteria primarily originate from theAcetivibrio, Ruminococcus, Ruminiclostridium, andClostridiumgenera. A rigorous analysis of their enzyme, scaffoldin, dockerin, and cohesin content reveals phylogenetically conserved features. Based on the presence of a high number of genes encoding both scaffoldins and dockerin-fused GHs, the cellulosomes inAcetivibrioandRuminococcusbacteria possess complex architectures that are populated with a large number of distinct LCB degrading GH enzymes. Their complex cellulosomes are distinguishable by their mechanism of attachment to the cell wall, the structures of their primary scaffoldins, and by how they are transcriptionally regulated. In contrast, bacteria in theRuminiclostridiumandClostridiumgenera produce ‘simple’ cellulosomes that are constructed from only a few types of scaffoldins that based on their distinct complement of GH enzymes are predicted to exhibit high and low cellulolytic activity, respectively. Collectively, the results of this study reveal conserved and divergent architectural features in bacterial cellulosomes that could be useful in guiding ongoing efforts to harness their cellulolytic activities for bio-based chemical and materials production.
