SARS-CoV-2 raw wastewater surveillance from student residences on an urban university campus

The COVID-19 pandemic brought about an urgent need to monitor the community prevalence of infection and detect the presence of SARS-CoV-2. Testing individual people is the most reliable method to measure the spread of the virus in any given community, but it is also the most expensive and time-consuming. Wastewater-based epidemiology (WBE) has been used since the 1960s when scientists implemented monitoring to measure the effectiveness of the Polio vaccine. Since then, WBE has been used to monitor populations for various pathogens, drugs, and pollutants. In August 2020, the University of Tennessee-Knoxville implemented a SARS-CoV-2 surveillance program that began with raw wastewater surveillance of the student residence buildings on campus, the results of which were shared with another lab group on campus that oversaw the pooled saliva testing of students. Sample collection began at 8 am, and the final RT-qPCR results were obtained by midnight. The previous day’s results were presented to the campus administrators and the Student Health Center at 8 am the following morning. The buildings surveyed included all campus dormitories, fraternities, and sororities, 46 buildings in all representing an on-campus community of over 8,000 students. The WBE surveillance relied upon early morning “grab” samples and 24-h composite sampling. Because we only had three Hach AS950 Portable Peristaltic Sampler units, we reserved 24-h composite sampling for the dormitories with the highest population of students. Samples were pasteurized, and heavy sediment was centrifuged and filtered out, followed by a virus concentration step before RNA extraction. Each sample was tested by RT-qPCR for the presence of SARS-CoV-2, using the CDC primers for N Capsid targets N1 and N3. The subsequent pooled saliva tests from sections of each building allowed lower costs and minimized the total number of individual verification tests that needed to be analyzed by the Student Health Center. Our WBE results matched the trend of the on-campus cases reported by the student health center. The highest concentration of genomic copies detected in one sample was 5.06 × 10 7 copies/L. Raw wastewater-based epidemiology is an efficient, economical, fast, and non-invasive method to monitor a large community for a single pathogen or multiple pathogen targets.

The Essential Role of Integrated Nuclear-Renewable Energy Systems in Achieving Economy-wide Net Zero Solutions

Background/Objectives. The Biden administration has committed to full decarbonization of the U.S. electricity grid by 2035 and economy-wide net zero emissions by 2050. These aggressive goals demand immediate action if we are to be successful, and they require us to think more holistically about our clean energy options. Focused laboratory initiatives, such as the INL Integrated Energy Systems (IES) Initiative, and multiple programs within the Department of Energy are working together to address these holistic solutions. Approach/Activities. Traditionally, electricity generation and management and meeting energy demands for industry and transportation are considered independently. As we seek to achieve net zero, we need to reassess our energy demands. When we consider overall energy use, only one-third is in the form of electricity. Additional energy demands are in the form of heat or steam for industrial processes, as well as transportation. Emissions across these sectors are much harder to abate, and electrification may not be the best option. Reducing environmental emissions at an affordable cost, while maintaining grid reliability and resilience, will require us to use all of the clean energy resources that we have available. That means coordinating the use of nuclear, renewables, and fossil fuels with carbon capture to meet growing demands for electricity, industrial applications, and mobility. The primary focus of IES research is to assess the technical and economic potential of various IES solutions to enhance the flexibility and utilization of nuclear energy generators working alongside renewable generators to meet an array of energy demands—thereby maximizing the utilization of clean energy resources across all energy sectors. Various energy applications and product streams beyond electricity are being evaluated, ranging from generation of potable water to production of hydrogen, fertilizers, synthetic fuels, and various chemicals. Results/Lessons Learned. This presentation will highlight the wide array of RD&D being conducted to develop and deploy nuclear-based IES that will be key to achieving our net zero goals. By working with key collaborators in the nuclear industry, analytical studies are now becoming a reality in multiple demonstration projects.

Eligibility Assessment of TA-52-1 and TA-52-11: The Ultra-High Temperature Reactor Experiment (UHTREX) Complex

The U.S. Department of Energy (DOE), National Nuclear Security Administration, Los Alamos Field Office (NA-LA) requests the State Historic Preservation Officer (SHPO) to concur with the eligibility determinations contained in this report for Buildings 1 and 11 in Technical Area 52 (TA-52) at Los Alamos National Laboratory (LANL or the Laboratory). Triad National Security LLC cultural resources staff have completed the evaluation of two buildings, called the UHTREX Complex, for inclusion in the National Register of Historic Places (Register). This complex includes the UHTREX Reactor Building (TA-52-1), and an associated Mechanical Assembly Building (TA-52-11). As part of LANL’s Footprint Reduction Program, both facilities of the UHTREX Complex are scheduled for characterization and demolition. In addition to evaluating their eligibility in the Register, the properties at TA-52 were assessed for potential adaptive reuse, long-term preservation, and public interpretation. Based on the findings in this assessment report, both TA-52-1 and TA-52-11 have been determined to not be eligible for inclusion in the Register. The history of the UHTREX Complex lacks association with exceptionally significant Cold War events of scientific developments. Both TA-52-1 and TA-52-11 lack the necessary internal historic integrity suitable for long-term preservation or public interpretation. And both facilities contain legacy radioactive contamination, which prohibits their reuse. In addition to its loss of integrity and context, TA-52- 11 has been determined ineligible due to its status as a support building of secondary or minor importance. In compliance with Section 106 and Section 110 of the National Historic Preservation Act of 1966, as amended, and with the Programmatic Agreement among the U.S. Department of Energy, National Nuclear Security Administration, Los Alamos Field Office, the New Mexico State Historic Preservation Office, and the Advisory Council on Historic Preservation Concerning Management of the Historic Properties at Los Alamos National Laboratory, Los Alamos, New Mexico, the SHPO is requested to concur with the eligibility determinations contained in this report for the UHTREX Complex in TA-52.

Georectified polygon database of ground-mounted large-scale solar photovoltaic sites in the United States.

Over 4,400 large-scale solar photovoltaic (LSPV) facilities operate in the United States as of December 2021, representing more than 60 gigawatts of electric energy capacity. Of these, over 3,900 are ground-mounted LSPV facilities with capacities of 1 megawatt direct current (MW dc ) or more. Ground-mounted LSPV installations continue increasing, with more than 400 projects appearing online in 2021 alone; however, a comprehensive, publicly available georectified dataset including spatial footprints of these facilities is lacking. The United States Large-Scale Solar Photovoltaic Database (USPVDB) was developed to fill this gap. Using US Energy Information Administration (EIA) data, locations of 3,699 LSPV facilities were verified using high-resolution aerial imagery, polygons were digitized around panel arrays, and attributes were appended. Quality assurance and control were achieved via team peer review and comparison to other US PV datasets. Data are publicly available via an interactive web application and multiple downloadable formats, including: comma-separated value (CSV), application programming interface (API), and GIS shapefile and GeoJSON.
