Integration of proteomics and metabolomics into the Design, Build, Test, Learn cycle to improve 3-hydroxypropionic acid production in Aspergillus pseudoterreus

Biological engineering of microorganisms to produce value-added chemicals is a promising route to sustainable manufacturing. However, overproduction of metabolic intermediates at high titer, rate, and yield from inexpensive substrates is challenging in non-model systems where limited information is available regarding metabolic flux and its control in production conditions. Integrated multi-omic analyses of engineered strains offers an in-depth look at metabolites and proteins directly involved in growth and production of target and non-target bioproducts. Here we applied multi-omic analyses to overproduction of the polymer precursor 3-hydroxypropionic acid (3HP) in the filamentous fungus Aspergillus pseudoterreus. A synthetic pathway consisting of aspartate decarboxylase, beta-alanine pyruvate transaminase, and 3HP dehydrogenase was designed and built for A. pseudoterreus. Strains with single- and multi-copy integration events were isolated and multi-omics analysis consisting of intracellular and extracellular metabolomics and targeted and global proteomics was used to interrogate the strains in shake-flask and bioreactor conditions. Production of a variety of co-products (organic acids and glycerol) and oxidative degradation of 3HP were identified as metabolic pathways competing with 3HP production. Intracellular accumulation of nitrogen as 2,4-diaminobutanoate was identified as an off-target nitrogen sink that may also limit flux through the engineered 3HP pathway. Elimination of the high-expression oxidative 3HP degradation pathway by deletion of a putative malonate semialdehyde dehydrogenase improved the yield of 3HP by 3.4× after 10 days in shake-flask culture. This is the first report of 3HP production in a filamentous fungus amenable to industrial scale biomanufacturing of organic acids at high titer and low pH.

Design Basis Document / Owner’s Technical Specification for Nitrate Salt Systems in CSP Projects: Hypothesis of Hot Tank Failure

Work has begun on a Design Basis Document / Owner’s Technical Specification for parabolic trough and central receiver power plants using nitrate salt as the heat transport fluid and the thermal storage medium. A principal topic of recent interest is the failures of the hot salt tanks in central receiver projects. The paper outlines a hypothesis for the source of the failures, and discusses a range of possible solutions.

Spaceborne computer executive routine functional design specification. Volume 1: Functional design of a flight computer executive program for the reusable shuttle

A flight computer functional executive design for the reusable shuttle is presented. The design is given in the form of functional flowcharts and prose description. Techniques utilized in the regulation of process flow to accomplish activation, resource allocation, suspension, termination, and error masking based on process primitives are considered. Preliminary estimates of main storage utilization by the Executive are furnished. Conclusions and recommendations for timely, effective software-hardware integration in the reusable shuttle avionics system are proposed.

GENENG 2: A program for calculating design and off-design performance of two- and three-spool turbofans with as many as three nozzles

A computer program which calculates steady-state design and off-design jet engine performance for two- or three-spool turbofans with one, two, or three nozzles is described. Included in the report are complete FORTRAN 4 listings of the program with sample results for nine basic turbofan engines that can be calculated: (1) three-spool, three-stream engine; (2) two-spool, three-stream, boosted-fan engine; (3) two-spool, three-stream, supercharged-compressor engine; (4) three-spool, two-stream engine; (5) two-spool, two-stream engine; (6) three-spool, three-stream, aft-fan engine; (7) two-spool, three-stream, aft-fan engine; (8) two-spool, two-stream, aft-engine; and (9) three-spool, two-stream, aft-fan engine. The simulation of other engines by using logical variables built into the program is also described.

Space shuttle low pressure auxiliary propulsion subsystem design definition: Design handbook

A detailed description of recommended low pressure auxiliary propulsion subsystems (APS) for a space shuttle orbiter and booster is presented. The APS designs are the product of a study to identify and evaluate APS concepts, and to perform in-depth design and performance analyses for the most attractive of these. Selected APS baselines use the main engine propellant tanks as low pressure gas accumulators. For the orbiter, propellants from separate liquid tanks are used for main engine tank resupply. Resupply propellants are first circulated through tubular, passive heat exchangers, where they are vaporized and superheated prior to injection into the main engine tanks. Warm propellant vapors from the main engine tanks are mixed with additional liquid propellants in a downstream liquid/vapor mixer and supplied to the engines at constant temperature and pressure (constant density). The booster APS requires no separate propellant storage, since propellant residuals, trapped in the main engine tanks following boost, are sufficient to meet APS propellant demands.

Application of EVA guidelines and design criteria. Volume 2: EVA workstation conceptual designs

Several EV workstation concepts were developed and are documented. The workstation concepts were developed following a comprehensive analysis of potential EV missions, functions, and tasks as interpreted from NASA and contractor space shuttle and space station studies, mission models, and related reports. The design of a versatile, portable EVA workstation is aimed at reducing the design and development costs for each mission and aiding in the development of on-orbit serviceable payloads.

Compilation, design tests: Energetic particles Satellite S-3 including design tests for S-3A, S-3B and S-3C

A compilation of engineering design tests which were conducted in support of the Energetic Particle Satellite S-3, S-3A, and S-3b programs. The purpose for conducting the tests was to determine the adequacy and reliability of the Energetic Particles Series of satellites designs. The various tests consisted of: (1) moments of inertia, (2) functional reliability, (3) component and structural integrity, (4) initiators and explosives tests, and (5) acceptance tests.

Analysis and design of three dimensional supersonic nozzles. Volume 3: A design technique for multiple nozzle configurations

An iteration procedure for optimum nozzle design with application to hypersonic aircraft is discussed. In the procedure, intial values of several of the design parameters are assumed from simplified approximate considerations and are then varied systematically in order to determine the value of each parameter which optimizes nozzle performance. Improvements in nozzle performance are obtained by perturbing the initially assumed values of the geometric parameters on the basis of a computer analysis.
