SARS-CoV-2 propagation under nirmatrelvir and ensitrelvir pressure selects for main protease (MPro) drug-resistant mutations E166V (DRM2), L50F/E166V (DRM3), E166A/L167F (DRM4), and L50F/E166A/L167F (DRM5). DRM2-DRM5 undergoes N-terminal autoprocessing to produce mature MPro with dimer dissociation constants (K dimer ) 2–3 times larger than that of the wildtype. Co-selection of L50F restores catalytic activity of DRM2 and DRM4 from ~10 to 30%, relative to that of the wild-type enzyme, without altering K dimer . Binding affinities and thermodynamic profiles that parallel the drug selection pressure, exhibiting significant decreases in affinity through entropy/enthalpy compensation, were compared with GC373. Reorganization of the active sites due to mutations observed in the inhibitor-free DRM3 and DRM4 structures as compared to MPro WT may account for the reduced binding affinities, although DRM2 and DRM3 complexes with ensitrelvir are almost identical to MPro WT -ensitrelvir. In conclusion, chemical reactivity changes of the mutant active sites due to differences in electrostatic and protein dynamics effects likely contribute to losses in binding affinities.

Using nanoscale building blocks to construct hierarchical materials is a radical new branch point in materials discovery that promises new structures and emergent functionality. Understanding the design principles that govern nanoparticle assembly is critical to moving this field forward. By exploiting mixed ligand environments to target patchy nanoparticle surfaces, we have demonstrated a novel method of colloidal quantum dot (QD) assembly that gives rise to 2D structures. The equilibration of solutions of spherical and quasispherical QDs, including CdS, CdSe, and InP, with 2,2'-bipyridine-5,5'-diacrylic acid resulted in the preferential formation of 2D assemblies over the course of days as determined by transmission electron microscopy analysis. Small-angle X-ray scattering confirms the existence of the QD assemblies in solution. The dependence of the assembly on linker properties (length and rigidity), linker concentration, and total concentration was investigated, together with the data point to a mechanism involving ligand redistribution to create a patchy surface that maximizes the steric repulsion of neighboring QDs. By operating in an underexchanged regime, the arising patchiness results in enthalpically preferred directions of cross-linking that can be accessed by thermal equilibration.

The mechanical properties of nonequilibrium polymer hydrogels obtained from the transient cross-linking of polymer chains by a chemical fuel were investigated. Aqueous polymers featuring pendant carboxylic acids were treated with a carbodiimide to give anhydride-cross-linked gels. The anhydrides spontaneously hydrolyze back to the polymer solution, and the cycle can be repeated multiple times. Oscillatory rheology was employed to study the effects of temperature, fuel concentration, chain length, and polymer composition on the storage and loss moduli of the polymeric materials as well as the time taken for the polymers to undergo decross-linking. Regardless of the temperature used, at constant carbodiimide concentration, degelation times are more sensitive to experimental temperature than are the peak storage moduli. Decross-linking times decrease with increasing temperature. As carbodiimide concentration decreases, there is a decrease in moduli and decross-linking times. Within the scope of materials studied, the polymer structure was found to have a relatively small impact on the transient properties of gel networks compared to the fuel concentration and temperature. Furthermore, these findings facilitate the design of tunable on-demand networks and gels.
