Conformational motions and ligand-binding underlying gating and regulation in IP 3 R channel

Inositol-1,4,5-trisphosphate receptors (IP3Rs) are activated by IP 3 and Ca 2+ and their gating is regulated by various intracellular messengers that finely tune the channel activity. Here, using single particle cryo-EM analysis we determined 3D structures of the nanodisc-reconstituted IP 3 R1 channel in two ligand-bound states. These structures provide unprecedented details governing binding of IP 3 , Ca 2+ and ATP, revealing conformational changes that couple ligand-binding to channel opening. Using a deep-learning approach and 3D variability analysis we extracted molecular motions of the key protein domains from cryo-EM density data. We find that IP 3 binding relies upon intrinsic flexibility of the ARM2 domain in the tetrameric channel. Our results highlight a key role of dynamic side chains in regulating gating behavior of IP 3 R channels. This work represents a stepping-stone to developing mechanistic understanding of conformational pathways underlying ligand-binding, activation and regulation of the channel.

Pan-tropical daily L-band microwave land surface emissivity retrieval from GNSS-R observations

The uncertainties in microwave land surface emissivity (MLSE) measurements have long limited the use of spaceborne microwave radiometer data. As an emerging observation method, Global Navigation Satellite System Reflectometry (GNSS-R) has demonstrated great potential in several land and ocean applications. In this study, a method for obtaining daily MLSE dataset in the pan-tropical region from Cyclone GNSS (CYGNSS) observations is presented and evaluated. The CYGNSS observations are first aggregated into the Equal-Area-Scalable-Earth (EASE) 2.0 36 km grid by a combined weight function of distance, time, and signal-to-noise ratio variance. Then, the method employs a pixel-by-pixel regression algorithm to conduct the daily MLSE retrieval using reference emissivity derived from the Soil Moisture Active Passive (SMAP) brightness temperature. The CYGNSS MLSE shows good agreement with SMAP MLSE, delivering an overall root-mean-square error (RMSE) of 0.022 and 0.017 for horizontal and vertical polarization, respectively, during the training set spanning the whole year of 2018. Furthermore, on the test set from January 2019 to May 2019, the RMSE values amounted to 0.030 and 0.023 for horizontal and vertical polarization, respectively. Temperature records from the International Soil Moisture Network are employed to calculate the emissivity and for in-situ validation, which yield an RMSE of 0.034 and 0.026 for the two polarizations, respectively. The proposed algorithm provides an encouraging approach to obtain accurate daily MLSE dataset for microwave remote sensing. Compared to the SMAP MLSE, the CYGNSS MLSE has a remarkable improvement of 86% in temporal resolution, greatly complementing the existing microwave emissivity datasets.

K-Shell photoabsorption in Si 11+ : Relativistic contributions via Breit-Pauli R-matrix calculations

As a part of the investigation of the entire Si isonuclear sequence for x-ray spectral diagnostics, we focus on the Li-like Si 11+ ion, a stable component that features prominently in x-ray astrophysical spectra. In this work, we perform R-matrix calculations for the photoabsorption cross section of Si 11+ including relativistic contributions via the use of a Breit-Pauli Hamiltonian. Relativistic effects—predominantly spin-orbit splitting of resonances and mass-velocity and one-body Darwin global shifts of energies— are shown to be tractable and of importance for accuracy in x-ray spectral modeling data banks.

Elastic properties of the W 0.75 R e 0.25 alloy at high pressure up to 183 GPa

The high pressure equation of state for the W 0.75 R e 0.25 alloy is experimentally determined up to 183 GPa with synchrotron angle-dispersive powder x-ray diffraction in the diamond-anvil cell and to ∼925 GPa with density-functional theory. W-Re alloys are used in many industrial high-temperature applications and as a confining gasket material in high-pressure diamond-anvil cell research. The inclusion of 25 wt. % Re achieves the highest performance in terms of strength and ductility while also maintaining the body-centered-cubic (bcc) crystal structure, yet to date there has been no investigation into its elastic behavior at high pressure. We present the experimentally and theoretically determined volumetric and elastic pressure response and systematically compare these results to other W-Re alloys, finding that the bulk modulus of W-Re alloys varies nonlinearly with Re content and W 0.75 Re 0.25 becomes less incompressible than W at 85 GPa. Spectroscopic factors, overlaps, and isospin symmetry from an R -matrix point of view

Background: Spectroscopic factors, overlaps, and isospin symmetry are often used in conjunction with single-particle wave functions for the phenomenological analysis of nuclear structure and reactions. Many differing prescriptions for connecting these quantities to physically relevant asymptotic normalization constants or widths are available in the literature, but their relationship and degree of validity are not always clear. Purpose: This paper derives relationships among the above quantities of interest using well-defined methodology and starting assumptions. Method: R -matrix theory is used as the primary tool to interoperate among the quantities of interest to this work. Particular attention is paid to effects arising from beyond the nuclear surface, where isospin symmetry is strongly violated. Results: Relationships between the quantities of interest are derived. Example applications of these methods to mirror levels in nucleon + 12 C, nucleon + 16 O, and nucleon + 26 Al are presented. A new approach to multilevel mirror symmetry is derived and applied to the first three 2 + states of 18 O and 18 Ne. Conclusions: In this work, the relationship between the quantities of interest is clarified and certain procedures are recommended. It is found that the asymptotic normalization constant of the second 2 + state in 18 Ne deduced from the mirror state in 18 O is significantly larger than found in previous work. This finding has the effect of increasing the 17 F($\textit{p,γ}$) 18 Ne reaction rate in novae.

Search for R -parity violating supersymmetric decays of the top squark to a b -jet and a lepton in s = 13 TeV p p collisions with the ATLAS detector

A search is presented for direct pair production of the stop, the supersymmetric partner of the top quark, in a decay through an R-parity violating coupling to a charged lepton and a b-quark. The dataset corresponds to an integrated luminosity of 140 fb -1 of proton-proton collisions at a center-of-mass energy of $\sqrt{s}$=13 TeV collected between 2015 and 2018 by the ATLAS detector at the LHC. The final state has two charged leptons (electrons or muons) and two b-jets. The results of the search are interpreted in the context of a Minimal Supersymmetric Standard Model with an additional B -L gauge symmetry that is spontaneously broken. No significant excess is observed over the Standard Model background, and exclusion limits on stop pair production are set at 95% confidence level. The corresponding lower limits on the stop mass for 100% branching ratios to a b-quark and an electron, muon, or tau-lepton are 1.9 TeV, 1.8 TeV and 800 GeV, respectively, extending the reach of previous LHC searches.

Realizing Kagome Band Structure in Two-Dimensional Kagome Surface States of RV 6 Sn 6 (R = Gd, Ho)

Here, we report angle resolved photoemission experiments on a newly discovered family of kagome metals RV 6 Sn 6 (R = Gd, Ho). Intrinsic bulk states and surface states of the vanadium kagome layer are differentiated from those of other atomic sublattices by the real-space resolution of the measurements with a small beam spot. Characteristic Dirac cone, saddle point, and flat bands of the kagome lattice are observed. Our results establish the two-dimensional (2D) kagome surface states as a new platform to investigate the intrinsic kagome physics.

New technology for an ancient fish: A lamprey life cycle modeling tool with an R Shiny application

Lampreys (Petromyzontiformes) are an ancient group of fishes with complex life histories. We created a life cycle model that includes an R Shiny interactive web application interface to simulate abundance by life stage. This will allow scientists and managers to connect available demographic information in a framework that can be applied to questions regarding lamprey biology and conservation. We used Pacific lamprey ( Entosphenus tridentatus ) as a case study to highlight the utility of this model. We applied a global sensitivity analysis to explore the importance of individual life stage parameters to overall population size, and to better understand the implications of existing gaps in knowledge. We also provided example analyses of selected management scenarios (dam passage, fish translocations, and hatchery additions) influencing Pacific lamprey in fresh water. These applications illustrate how the model can be applied to inform conservation efforts. This tool will provide new capabilities for users to explore their own questions about lamprey biology and conservation. Simulations can hone hypotheses and predictions, which can then be empirically tested in the real world.

Mapping Rydberg States of H 2 with the Halfium R-Matrix Method

In this article, we use the Halfium R-matrix method to investigate the Rydberg states of the H 2 molecule up to n = 20, filling the gap above the low-lying bound states already calculated with configuration interaction packages. Moreover, we show that the use of Quantum Defect Theory scaling laws, allows for a comprehensive analysis of the regular patterns resulting from the coupling between Rydberg series and doubly excited states. The results should open the door for more efficient quasi-diabatization of the potential energy curves which is required for calculating cross sections and rate coefficients of the (e + H 2 + ) collisional processes, involved in the plasma modeling for fusion devices.

RF Harmonic Kicker R&D Demonstration and Its Application to the RCS Injection of the EIC

The Rapid Cycling Synchrotron (RCS) of the Electron-Ion Collider (EIC) at Brookhaven National Laboratory (BNL) is an accelerating component of the electron injection complex, which provides polarized electrons in electron-ion collisions in the main Electron Storage Ring (ESR). We present the injection scheme into the RCS based on an ultra-fast harmonic kicker, whose "five odd-harmonic modes" prototype was developed in the context of the Jefferson Lab EIC (JLEIC) conceptual design. In its early stage of R&D, the sharp (~3 ns width) waveform construction, beam dynamics, and pulsed power operation with short ramping time (~10 us) will be discussed together with the fabrication work of the JLEIC prototype.

ambrosia: An R package for calculating and analyzing food demand that is responsive to changing incomes and prices

The ambrosia R package was developed to calculate food demand for staples and non-staple commodities that is responsive to changing levels of incomes and prices. ambrosia implements the framework to quantify food demand as established by Edmonds et al. (Edmonds et al. 2017) and allows the user to explore and estimate different variables related to the food demand system. Currently ambrosia provides 3 main functions: (1) calculation of food demand for any given set of parameters including income levels and prices (2) estimation of parameters within a given a dataset. Note: ambrosia is used to calculate parameters for the food demand model implemented in the Global Change Analysis Model(GCAM) (3) exploration and preparation of raw data before starting a parameter estimation

Determination of Solar System R-Process Abundances using ENDF/B-VIII.0 and TENDL-2015 libraries

Recent multi-messenger detection of the binary neutron star merger (GW170817) energized the astrophysical community and encouraged further research for determination of nuclear physics observables. Comprehensive studies of atomic nuclei in the cosmos provide an opportunity for investigating these astrophysical phenomena and acquiring complementary information on stellar nucleosynthesis processes that can be verified using the latest nuclear data. Evaluated Nuclear Data File (ENDF) libraries contain complete collections of reaction cross sections over the energy range relevant to astrophysics, fission yields and decay data. These data collections have been used worldwide in nuclear science, industry and national security applications. There is great interest in exploring the ENDF/B-VIII.0 and TALYS Evaluated Nuclear Data Library (TENDL-2015) for nuclear astrophysics purposes and comparing findings with the Karlsruhe Astrophysical Database of Nucleosynthesis in Stars (KADoNiS). The Maxwellian-averaged cross sections (MACS) and astrophysical reaction rates have been calculated using the ENDF/B-VIII.0 and TENDL-2015 evaluated data sets. The calculated cross sections were combined with the solar system abundances and fitted using the classical model of stellar nucleosynthesis. Astrophysical rapid- and slow-neutron capture, r- and s-process, respectively, abundances were obtained from present data and compared with available values. Further analysis of MACS reveals potential evaluated libraries data deficiencies and a strong need for new measurements. The current results demonstrate a large nuclear astrophysics potential of evaluated libraries and mutually beneficial relations between nuclear industry and research e orts.

How to Create, and Sustain, R&D Leadership A blueprint for international collaboration

How the United States can compete in the twenty-first century is one of the most vexing challenges for policy makers and academics today. Many studies have shown that most impactful science and technology advances happen in open and diverse environments, under excellent research conditions, with sufficient funding. Since the end of World War II, the United States has been the most attractive country for open research and commercialization of new discoveries. This strategy—partly articulated in science administrator Vannevar Bush’s Endless Frontier report of 1945—leverages federal government investments in research and development (R&D) to enhance economic competitiveness and national security. The combination of a dynamic, open research ecosystem and world-leading science infrastructure provided an exciting work environment leading to groundbreaking discoveries, and propelled economic growth through the second half of the twentieth century.

Extended Plate Wall System Expands Adoption of High-R Envelope Solutions

For decades, the residential building industry has worked to expand opportunities to increase the thermal resistance of walls. Multiple high-R wall construction methods have been developed, yet market adoption remains low because of perceived complexity and cost. One appealing solution is an extended plate wall system (EPW). Framing for top and bottom plates is one nominal dimension larger than for studs, which provides space for a 2-inch layer of foam plastic insulative sheathing (FPIS). Wood structural sheathing (OSB or plywood) is then installed on the exterior, fastened directly to the plates and nailed through the foam to the studs. In contrast to the conventional method of installing FPIS as a fully exterior layer, the EPW greatly simplifies air sealing, installation and detailing of windows and doors, an application of a weather resistive barrier, and attachment of cladding.
