Time-series RNA metabarcoding of the active Populus tremuloides root microbiome reveals hidden temporal dynamics and dormant core members

The rhizosphere is a critical interface between plant roots and soil, harboring diverse microbial communities that are essential to plant and ecosystem health. Although these communities exhibit stark temporal dynamics, their dormancy/activity transitions remain poorly understood. Such transitions may enable microbes to rapidly adjust functional contributions faster than community turnover alone would allow. Here, we used RNA metabarcoding to characterize the active fraction of microbial communities on the roots of quaking aspen (Populus tremuloides) in a time-series study across a natural environmental gradient. We explore cryptic temporal microbial community dynamics of rhizosphere communities at the ecosystem scale. The active rhizosphere bacterial and fungal communities were more temporally dynamic than total communities, while total communities exhibited a stronger response to site-specific conditions. Notably, some core microbiome members were often inactive, yielding a smaller “active core” subset. The fungal endophyte Hyaloscypha finlandica was the only microbe that was both present and active in all plots across all timepoints. Soil temperature strongly influenced both total and active community composition, with the fungal class Eurotiomycetes showing a temperature-dependent seasonal decline in abundance. Together, these results reveal that modulation of microbial activity levels is a key mechanism by which the plant root holobiont responds to environmental variation, and that even dominant symbionts may frequently persist in dormancy within the rhizosphere.

Visualizing the Hidden Half: Plant-Microbe Interactions in the Rhizosphere

Plant roots and the associated rhizosphere constitute a dynamic environment that fosters numerous intra- and interkingdom interactions, including metabolite exchange between plants and soil mediated by root exudates and the rhizosphere microbiome. These interactions affect plant fitness and performance, soil health, and the belowground carbon budget.

Unraveling Hidden Order and Dynamics in a Heterogeneous Ferroelectric System Using Machine Learning

We uploaded two batches of datasets: Batch-a) Equilibrium dynamics at a constant temperature at zero electric-field, with one trajectory each for 4-different defect structures (i.e. SETs 1 to 4). Each trajectory has a time-step of 0.25 fs, and is run for 7775000 time-steps, with snapshots written out every 4 time-steps (i.e. every 1fs); and Batch-b) Non-Equilibrium dynamics at a constant temperature with the same 0.25 fs time-step, but data dumped every 500 times-steps (i.e. every 125fs) for each of the SETs. The total trajectory of each defect structure (i.e. each SETs 1 to 4) is 2800000 time-steps (5600 snapshots), with stepping of electric-field by 0.01 V/Ã…, after every 100,000 time-steps (i.e. every 200 snapshots), from E=0 to E=0.05 V/Ã… to E = -0.05 V/Ã… to E=0.05 V/Ã….

Reply to “Comment on ‘The Reduced Detection Rate of Signals That Are Hidden by Earthquakes: Case Studies with Spotlight Detectors That Operate at Seismic Arrays,’ by Joshua D. Carmichael, Brent G. Delbridge, and Richard Alfaro-Diaz” by Paul G. Richards

We reply to a comment that Paul G. Richards (Richards, 2026) directed to an article by Carmichael et al. (2025). Richards expressed concern over two issues. The first issue relates to the size of the explosions discussed in the article. The second relates to the probability that earthquakes and explosions sourced near the same location coincide in time, by chance. We do not address the first issue because the original article does not assign significance to the size of the explosions from the point of view of an experimental party. We do respond to Richards’ comment on the second issue and claim that it can be explained through a clarification about conditional versus joint probabilities. Our present discussion, therefore, describes the difference between the conditional probability that a correlator fails, given that an explosion and earthquake coincidently occur in the same place, and the joint probability that a correlator fails while an earthquake and explosion also coincidentally occur in the same place. This latter, joint probability appears to be rare. We clarify that our original article did not treat the joint probability, only the conditional probability.

The Hidden Flexibility of the Natural Gas Network for Electric Power Operations: A Case Study of a Near-Miss Winter Event

The U.S. power sector has become increasingly reliant on gas pipeline networks to deliver fuel to natural gas power plants. In addition to supplying relatively low-cost fuel, gas networks offer generators flexibility in their operations through the ability to deliver fuel when needed by using gas storage facilities or linepack if the gas network is at an operating point below its design capacity. However, disruptions or stress events on the gas network - like those occurring in the Northeast and Texas in recent years - can result in limitations on gas availability to generators at times when generation is in short supply. Here we examine a period of stress that occurred in the winter of 2022 in the Western United States. Using data on the region's natural gas pipeline network and electric generators, we build an integrated gas and electric model that closely replicates the actual dispatch of the period. We then evaluate the implications of removing flexibility employed by the gas network operator, which during that period curtailed scheduled gas deliveries to other parties to increase deliveries to natural gas power plants, which requested more gas than initially forecasted. We find that without the flexibility supplied by the gas network operator, there would have been curtailment of gas generation due to gas offtake constraints, requiring the power system operator to redispatch relying on more expensive generation or to potentially shed load. A sensitivity exploring a wind drought further exacerbates the strain, illustrating the potential challenge of managing gas and grid interactions as systems move to higher shares of variable renewable electricity. Based on this example, we discuss potential coordination strategies between the two system operators to ensure that the power system can successfully utilize and rely on the flexibility offered by natural gas networks.

Hidden Sectors from Cosmos to Colliders

The major aims of this project are to (i) to systematically explore the cosmological history of dark matter born out of a larger dark sector and to establish new discovery opportunities for dark matter in both astrophysical and terrestrial experiments, and (ii) to develop new techniques to discover very weakly coupled new physics at colliders, especially particles produced in exotic decays of the Higgs boson and/or with a macroscopic proper lifetime. This report summarizes how the research carried out in this project addresses these goals. Selected highlights from the final year of the project include (i) establishing new probes of dark sectors in the early universe through visible footprints in cosmic microwave background, big bang nucleosynthesis, and gravitational wave observables; (ii) developing new links between self-interacting dark sectors and dark matter microstructure; (iii) surveying progress in the search for exotic decays of the Higgs and discussing the resulting consequences for models of nature.

Uncovering hidden market opportunities for advanced nuclear reactors

Decarbonizing to meet aggressive climate change mitigation targets requires energy transition within all sectors. In the industrial sector, global emissions will need to decrease by 65–90% by 2050 to avert warming greater than 1.5°C (Pörtner et al., 2022). Recent U.S. laws, including the Inflation Reduction Act (IRA), Bipartisan Infrastructure Law (BIL), Defense Production Act, Creating Helpful Incentives to Produce Semiconductors (or CHIPS), state programs, and other recent laws, have clean energy requirements and provide financial incentives to accelerate the use of clean energy technologies in the industrial sector. These new laws include supporting mechanisms with direct financial support for nuclear power (e.g., advanced reactor development and hydrogen production). However, advanced nuclear could also gain these financial benefits by coupling with low-carbon industrial projects. For example, microreactors could supply low-carbon energy to producers of critical metals that (1) are eligible to receive investment and production tax credits and favorable loans, and (2) the low-carbon product could gain preference in emerging markets for green products.

Hidden in Plain Sight How Interior Furnishings Management Can Help Los Alamos National Laboratory Meet the United States Department of Energy's Sustainability Goals [Slides]

Annually close to 50,000 lbs. of furniture avoids landfills through this program. Over a ten year (2020-2030) over 224 tons of furniture will avoid landfills through the program, that is the weight of the Statue of Liberty. The ReUse program saves LANL $\$172,875$ annually based on the salvage value of the furniture. If the product reused were to be purchased new, LANL would spen close to $1,200,000.

Hidden in Plain Sight: How Interior Furnishings Management Can Help Los Alamos National Laboratory Meet the United States Department of Energy's Sustainability Goals [Capstone Project]

In the last three years, LANL has purchased over $50 million in new interior products with a focus on sustainable building and high-quality interior furnishings, carpet, and paint. Purchasing interior products that focus on sustainable manufacturing, however, is only the beginning of properly operating and maintaining an interior space over its useable life. Sustainable commercial furniture, if well maintained and warrantied, can last well beyond the accounting depreciation of the asset, ultimately saving taxpayer dollars. Given DOE’s recently announced built environment sustainability goals, now is the time to look at interior products as an important part of the overall sustainability plan for LANL’s green building initiatives. The scope of this thesis provides an environmentally sustainable O&M process for the procurement, use, and proper decommissioning of interior products. The thesis also provides environmental and financial data to assist with LANL’s overall DOE Matrix milestones, currently due in 2030.
