Electrophoretic cell separation by means of immunomicrospheres

The electrophoretic mobility of fixed human red blood cells immunologically labeled with polymeric (4-vinyl)pyridine or polyglutaraldehyde microspheres was altered to a considerable extent. This observation was utilized in the preparative scale electrophoretic separation of human and turkey fixed red blood cells, whose mobilities under normal physiological conditions do not differ sufficiently to allow their separation by continuous flow electrophoresis. It is suggested that resolution in the electrophoretic separation of cell subpopulations, currently limited by finite and often overlapping mobility distributions, may be significantly enhanced by immuno-specific labeling of target populations using microspheres.

Condensation of activated diguanylates on a Poly/C/ template

The metal-ion catalysis of the oligomerization of activated diguanylate isomers on a polycytidylic acid template is studied in an investigation of possible early prebiotic polynucleotide replication mechanisms. The 5'-imidazolides of diguanylates linked 2' to 5' or 3' to 5' were reacted with polyC in a 1-methylimidazole or a 2,6-lutidine buffer in the presence of a Zn(+2) or a Pb(+2) catalyst, and reaction products were determined by paper chromatography, paper electrophoresis and liquid chromatography. In the lutidine buffer, the presence of both the Zn(+2) catalyst and the polyC template is found to result in the production of 3'-5' linked oligomers with up to 10 diguanylate units, and from diguanylates in the presence of the monomer. In the reactions conducted in the 1-methylimidazole buffer, the addition of Pb(+2) is found to lead to less marked increases in oligomerization in the presence of template, with approximately equal proportions of 2'-5' and 3'-5' oligomers formed from the 2'-5' substrate and mainly 3'-5' bonds from the 3'-5' linked dimer.

Zoological effects of variations in atmospheric oxygen levels

The role of certain gene enzymes in survival in modified atmospheres was examined. Chromosome morphology was studied. Mortality and life span were measured. Equipment to deliver various gas mixtures to the flies was designed and fabricated. To study the gene enzymes a technique called starch gel electrophoresis was needed. Equipment and supplies for this work and the study of chromosome morphology was available on the market, although some of the equipment was fabricated to save the project money.

Neurotrophic factor - Characterization and partial purification

Recent evidence suggests that neurotrophic activity is required for the normal proliferation and development of muscle cells. The present paper reports a study of the purification and characterization of a neurotrophic factor (NTF) from adult chicken ischiatic-peroneal nerves using two independent quantitative in vitro assay systems. The assays were performed by the measurement of the incorporation of tritiated thymidine or the sizes of single-cell clones by chick muscle cells grown in culture. The greatest amount of neutrotrophic activity is found to be extracted at a pH of 8; aqueous suspensions of the activity are stable to long-term storage at room temperature. The specific activity of the substance is doubled upon precipitation with ammonium sulfate or after gel filtration, and increase 4 to 5 fold after salt gradient elution from DEAE cellulose columns. The active fraction obtained after gel filtration and rechromatography on DEAE cellulose exhibits a 7 to 10-fold increase in specific activity. Electrophoresis of the most highly purified material yields a greatly concentrated band at around 80,000 daltons. Although NTF is purified almost 10-fold as indicated by the increase in specific activity, the maximum activity of the partially purified material is greatly reduced, possibly due to a requirement for a cofactor for the expression of maximum activity.

Theoretical studies in isoelectric focusing

A mathematical model has been developed which describes the steady state in an isoelectric focusing (IEF) system with ampholytes or monovalent buffers. The model is based on the fundamental equations describing the component dissociation equilibria, mass transport due to diffusion and electromigration, electroneutrality, and the conservation of charge. The validity and usefulness of the model has been confirmed by using it to formulate buffer systems in actual laboratory experiments. The model has been recently extended to include the evolution of transient states not only in IEF but also in other modes of electrophoresis.

Settling of fixed erythrocyte suspension droplets

It is pointed out that when particles behave collectively rather than individually, the fractionation of micron-size particles on the basis of size, density, and surface characteristics by centrifugation and electrophoresis is hindered. The formation and sedimentation of droplets containing particles represent an extreme example of collective behavior and pose a major problem for these separation methods when large quantities of particles need to be fractionated. Experiments are described that measure droplet sizes and settling rates for a variety of particles and droplets. Expressions relating the particle concentration in a drop to measurable quantities of the fluids and particles are developed. The number of particles in each droplet is then estimated, together with the effective droplet density. Red blood cells from different animals fixed in glutaraldehyde provide model particle groups.

Materials processing in the reduced-gravity environment of space

Studies of the effects of gravitation on the behavior of fluids and the shapes of materials for the years 1977-1982 are reviewed, with a focus on ground-based research which complements and gives direction to experimentation in the reduced-gravity environment of space. Fluid-behavior topics discussed include crystal growth from melts and from vapor, dendritic solidification and casting, multiphase alloy solidification, electrophoresis, convection behavior, and special heat-flow considerations characteristic of the space environment. With regard to solids, such subjects as containerless positioning technology and its applications (in glass preparation, liquid drop and shell studies, nucleation experiments, and measurements of fundamental properties of molten reactants), liquid floating zones, solid self-deformation, and sedimentation and buoyancy effects are considered and illustrated.

Countercurrent distribution of biological cells

Detailed physiochemical studies of dextran/poly(ethylene glycol) (PEG) two phase systems were carried out to characterize and provide understanding of the properties of the systems which determine cell partition and the electrophoretic behavior of phase drops responsible for electric field driven phase separation. A detailed study of the electrostatic and electrokinetic potentials developed in these systems was carried out. The salt partition was examined both in phase systems and with pure polymer solutions via equilibrium dialysis and mechanism of sulfate, chloride and phosphate partition shown to be exclusion by PEG rather than binding by dextran. Salt partition was shown to have a strong effect on the polymer compositions of the phases as well, an effect which produces large changes in the interfacial tension between them. These effects were characterized and the interfacial tension shown to obey a power law with respect to its dependence on the length of the tie line describing the system composition on a phase diagram. The electrostatic potential differences measured via salt bridges were shown to obey thermodynamic predictions. The electrophoretic mobilities measured were utilized to provide a partial test of Levine's incomplete theory of phase drop electrophoresis. The data were consistent with Levine's expression over a limited range of the variables tested.

Protein changes in leaf-sheath pulvini of barley (hordeum) induced by gravistimulation

Sodium dodecyl sulfate polyacrylamade gel electrophoresis (SDS-PAGE) pattern of salt soluble proteins elicite by gravistimulation were shown in the top and the bottom halves of the gravistimulated pulvini as follows: at least five proteins were increased in the tissues derived from the bottom halves of the pulvini in the approximate molecular weight range of 91, 57, 50, 22, 17 kilodatons. SDS densitometric scans indicated that the two of them are probably cellulase and invertase.

Pea amyloplast DNA is qualitatively similar to pea chloroplast DNA

Amyloplast DNA (apDNA), when subjected to digestion with restriction endonucleases, yields patterns nearly identical to that of DNA from mature pea chloroplasts (ctDNA). Southern transfers of apDNA and ctDNA, probed with the large subunit (LS) gene of ribulose-1,5-bisphosphate carboxylase (Rubisco), shows hybridization to the expected restriction fragments for both apDNA and ctDNA. However, Northern transfers of total RNA from chloroplasts and amyloplasts, probed again with the LS gene of Rubisco, shows that no detectable LS meggage is found in amyloplasts although LS expression in mature chloroplasts is high. Likewise, two dimensional polyacrylamide gel electrophoresis of etiolated gravisensitive pea tissue shows that both large and small subunits of Rubisco are conspicuously absent; however, in greening tissue these two constitute the major soluble proteins. These findings suggest that although the informational content of these two organelle types is equivalent, gene expression is quite different and is presumably under nuclear control.

Prochloron research

The purpose was to prepare Prochloron photosynthetic membranes for the isolation of the two major chlorophyll-proteins, the P700-chlorophyll a-protein and the light-harvesting chlorophyll a/b-protein, using SDS-polyacrylamide gel electrophoresis. The prepared proteins (purified) were examined for their cross-reactivity to polyclonal antibodies prepared from higher plant proteins. In addition, material was prepared for electron microscopy, and isolation of the DNA for determination of its general complexity (COT analysis) and similarity to barley chloroplast DNA and Anabaena DNA by using restriction-endonuclease analysis. Kleinschmidt spreads of the DNA were in the electron microscope to identify and measure the extent and size of the circlar DNA.

Ribulose 1,5-bisphosphate carboxylase and phosphoribulokinase in Prochloron

Ribulose 1,5-bisphosphate (RuBP) carboxylase and phosphoribulokinase, enzymes in the reductive pentose-phosphate cycle, were measured in cell-free extracts of Prochloran didemni. The partial purification and characterization of RuBP carboxylase were described. Prochloron RuBP carboxylase, when purified by isopycnic centrifugation in reoriented linear 0.2 to 0.8 M sucrose gradients, sedimented to a position which corresponded to that of the 520,000-dalton spinach enzyme. Sodium dodecyl sulfate polyacrylamide gel electrophoresis showed that the Prochloron enzyme was composed of large and small subunits (MW = 57,500 and 18,800). Though results established that the enzymes RuBP carboxylase and phosphoribulokinase were present in levels comparable to other CO2-fixing microorganisms, it was suggested that other enzymes in the Calvin cycle limit growth or that additional enzymic insufficiencies exist.

Purification of biomaterials by phase partitioning

A technique which is particularly suited to microgravity environments and which is potentially more powerful than electrophoresis is phase partitioning. Phase partitioning is purification by partitioning between the two immiscible aqueous layers formed by solution of the polymers poly(ethylene glycol) and dextran in water. This technique proved to be very useful for separations in one-g but is limited for cells because the cells are more dense than the phase solutions thus tend to sediment to the bottom of the container before reaching equilibrium with the preferred phase. There are three phases to work in this area: synthesis of new polymers for affinity phase partitioning; development of automated apparatus for ground-based separations; and design of apparatus for performing simple phase partitioning space experiments, including examination of mechanisms for separating phases in the absence of gravity.
