Creation of a High-Yield AAV Vector Production Platform in Suspension Cells Using a Design-of-Experiment Approach

Recombinant adeno-associated virus (rAAV) vectors are a leading gene delivery platform, but vector manufacturing remains a challenge. New methods are needed to increase rAAV yields and reduce costs. Past efforts to improve rAAV production have focused on optimizing a single variable at a time, but this approach does not account for the interactions of multiple factors that contribute to vector generation. Here, we utilized a design-of-experiment (DOE) methodology to optimize rAAV production in a HEK293T suspension cell system. We simultaneously varied the transgene, packaging, and helper plasmid ratios, the total DNA concentration, and the cell density to systematically evaluate the impact of each variable across 52 conditions. The results revealed a unique set of parameters with a lower concentration of transgene plasmid, a higher concentration of packaging plasmid, and a higher cell density than previously described protocols. Using this DOE-optimized protocol, we achieved unpurified yields approaching 3 × 10 14 viral genomes (VGs)/L of cell culture. Additionally, we incorporated polyethylene glycol (PEG)-based virus precipitation, pH-mediated protein removal, and affinity chromatography to our downstream processing, enabling average purified yields of >1 × 10 14 VGs/L for rAAV-EGFPs across 13 serotypes and capsid variants.

Porous architecture and thermal properties of thermal barrier coatings deposited by suspension plasma spray

Besides the intrinsic low heat transfer capability of material, the thermal insulation property of thermal barrier coatings (TBCs) also relies on their microstructures. For better understanding the relationship between process parameters, porous architecture, and thermal properties of coatings, YSZ coatings were firstly manufactured by suspension plasma spray (SPS). Afterwards their total porosities were characterized by using the technique of X-ray transmission, the nano/submicro pores in those coatings were detected with Ultra-Small Angle X-ray Scattering (USAXS), and the thermal properties of coatings were measured using the laser flash method. The results indicated that: i) the porous architecture of SPS coatings can be tailored by adapting process parameters. ii) increasing total porosity is an effective means for reducing the heat transfer capability of SPS coatings. iii) nano/submicro pores have higher influence sensitivity on thermal properties than that of larger pores. Increasing the content of nano-submicro pores is therefore more effective for improving the thermal insulation property of SPS coatings. The potential of SPS technique for fabricating TBCs was discussed as well.

Heating of aqueous suspensions of silicon nanoparticles by a 808-nm diode laser for application in local photohyperthermia

3D simulation of the distribution of thermal fields in aqueous suspensions of silicon nanoparticles upon irradiation by a high-power 808-nm laser diode operating in cw and quasi-cw repetitively pulsed regimes is performed. It is shown that irradiation by a repetitively pulsed laser with a peak power of 10 W and a pulse duration of 300 ms can form a region with a maximum temperature exceeding 42 °C, which is promising for application in cancer photohyperthermia for local heating of biotissues at a given depth. Onset of interfacial waves in the terahertz spectrum of a nanoparticle suspension

We used inelastic x-ray scattering to gain insight into the complex terahertz dynamics of a diluted Au-nanoparticle suspension in glycerol. Here, we observe that, albeit sparse, Au nanoparticles leave clear signatures on the dynamic response of the system, the main one being an additional mode propagating at the nanoparticle-glycerol interface. A Bayesian inferential analysis of the line shape reveals that such a mode, at variance with conventional acoustic modes, keeps a hydrodynamiclike behavior well beyond the continuous limit and down to subnanometer distances.

JET EROSION OF PARTICLE BEDS: PROJECTING CRITICAL SUSPENSION VELOCITIES FROM EFFECTIVE CLEARING / CLEANING RADII

Here we explore the relationship between effective cleaning/clearing radii, ECR, and critical suspension velocities, Ucs. Although one set of physics controls both the radial extent of erosion (i.e., the ECR) and the velocity needed to erode from the center of jet impingement to any given location (i.e., Ucs, typically defined from nozzle center to the vessel center), the relationship between these two remains unexplored quantitatively. Here we advance the model of Kuhn, et al., (PNNL-22816, 2013) as described by Pease, et al. (FEDSM2017-69444, 2017) to evaluate the relationship between the effective clearing/cleaning radius and the nozzle velocity on flat surfaces including flat bottomed vessels. Two governing dimensionless groups are identified. We present both a closed form analytical but transcendental solution and a non-iterative approximation modeled after the Serghides approximation of Colebrook’s nonlinear equation for both ECR versus nozzle velocity and Ucs. Comparison of the model to data from flume testing on a flat surface finds reasonable agreement.
