Uncovering the basis of protein-protein interaction specificity with a combinatorially complete library

Protein-protein interaction specificity is often encoded at the primary sequence level. However, the contributions of individual residues to specificity are usually poorly understood and often obscured by mutational robustness, sequence degeneracy, and epistasis. Using bacterial toxin-antitoxin systems as a model, we screened a combinatorially complete library of antitoxin variants at three key positions against two toxins. This library enabled us to measure the effect of individual substitutions on specificity in hundreds of genetic backgrounds. These distributions allow inferences about the general nature of interface residues in promoting specificity. We find that positive and negative contributions to specificity are neither inherently coupled nor mutually exclusive. Further, a wild-type antitoxin appears optimized for specificity as no substitutions improve discrimination between cognate and non-cognate partners. By comparing crystal structures of paralogous complexes, we provide a rationale for our observations. Collectively, this work provides a generalizable approach to understanding the logic of molecular recognition.

An evaluation of the Goddard Space Flight Center Library

The character and degree of coincidence between the current and future missions, programs, and projects of the Goddard Space Flight Center and the current and future collection, services, and facilities of its library were determined from structured interviews and discussions with various classes of facility personnel. In addition to the tabulation and interpretation of the data from the structured interview survey, five types of statistical analyses were performed to corroborate (or contradict) the survey results and to produce useful information not readily attainable through survey material. Conclusions reached regarding compatability between needs and holdings, services and buildings, library hours of operation, methods of early detection and anticipation of changing holdings requirements, and the impact of near future programs are presented along with a list of statistics needing collection, organization, and interpretation on a continuing or longitudinal basis.

The Control Point Library Building System

The Earth Resources Observation System (EROS) Data Center in Sioux Falls, South Dakota distributes precision corrected Landsat MSS and RBV data. These data are derived from master data tapes produced by the Master Data Processor (MDP), NASA's system for computing and applying corrections to the data. Included in the MDP is the Control Point Library Building System (CPLBS), an interactive, menu-driven system which permits a user to build and maintain libraries of control points. The control points are required to achieve the high geometric accuracy desired in the output MSS and RBV data. This paper describes the processing performed by CPLBS, the accuracy of the system, and the host computer and special image viewing equipment employed.

Documentation for the machine-readable version of A Library of Stellar Spectra (Jacoby, Hunter and Christian 1984)

The machine readable library as it is currently being distributed from the Astronomical Data Center is described. The library contains digital spectral for 161 stars of spectral classes O through M and luminosity classes 1, 3 and 5 in the wavelength range 3510 A to 7427 A. The resolution is approximately 4.5 A, while the typical photometric uncertainty of each resolution element is approximately 1 percent and broadband variations are 3 percent. The documentation includes a format description, a table of the indigenous characteristics of the magnetic tape file, and a sample listing of logical records exactly as they are recorded on the tape.

Spectral synthesis in the ultraviolet. I - Far-ultraviolet stellar library

A library of mean stellar energy distributions for use in population synthesis of the ultraviolet spectra of active star-forming galaxies is derived from the ultraviolet spectrophotometry in the IUE Spectral Atlas. The spectra extend from 1230 to 1930 A with a resolution of 6 A. The library contains eight main-sequence groups from O3 to A7 V, four giant groups from O5 to B9 III, and three supergiant groups from O9 to A0 I. Several continuum and spectral line indices are computed, and their usefulness as temperature and luminosity discriminants is discussed. The advantages of analyzing composite stellar populations in the far-ultraviolet, where cool starlight is strongly suppressed, are examined.

Common Graphics Library (CGL). Volume 1: LEZ user's guide

Users are introduced to and instructed in the use of the Langley Easy (LEZ) routines of the Common Graphics Library (CGL). The LEZ routines form an application independent graphics package which enables the user community to view data quickly and easily, while providing a means of generating scientific charts conforming to the publication and/or viewgraph process. A distinct advantage for using the LEZ routines is that the underlying graphics package may be replaced or modified without requiring the users to change their application programs. The library is written in ANSI FORTRAN 77, and currently uses a CORE-based underlying graphics package, and is therefore machine independent, providing support for centralized and/or distributed computer systems.

Development of an Ada package library

A usable prototype Ada package library was developed and is currently being evaluated for use in large software development efforts. The library system is comprised of an Ada-oriented design language used to facilitate the collection of reuse information, a relational data base to store reuse information, a set of reusable Ada components and tools, and a set of guidelines governing the system's use. The prototyping exercise is discussed and the lessons learned from it have led to the definition of a comprehensive tool set to facilitate software reuse.

A design for a reusable Ada library

A goal of the Ada language standardization effort is to promote reuse of software, implying the existence of substantial software libraries and the storage/retrieval mechanisms to support them. A searching/cataloging mechanism is proposed that permits full or partial distribution of the database, adapts to a variety of searching mechanisms, permits a changine taxonomy with minimal disruption, and minimizes the requirement of specialized cataloger/indexer skills. The important observation is that key words serve not only as indexing mechanism, but also as an identification mechanism, especially via concatenation and as support for a searching mechanism. By deliberately separating these multiple uses, the modifiability and ease of growth that current libraries require, is achieved.

Common Graphics Library (CGL). Volume 2: Low-level user's guide

The intent is to instruct the users of the Low-Level routines of the Common Graphics Library (CGL). The Low-Level routines form an application-independent graphics package enabling the user community to construct and design scientific charts conforming to the publication and/or viewgraph process. The Low-Level routines allow the user to design unique or unusual report-quality charts from a set of graphics utilities. The features of these routines can be used stand-alone or in conjunction with other packages to enhance or augment their capabilities. This library is written in ANSI FORTRAN 77, and currently uses a CORE-based underlying graphics package, and is therefore machine-independent, providing support for centralized and/or distributed computer systems.

Availability of a library of infrared (2.1-25.0 microns) mineral spectra

All previously published libraries of infrared mineral spectra are in the form of transmitance. Reflectance spectra are, however, more useful for remote sensing and some potential laboratory applications, such as the use of an infrared microscope for mineral identification on polished sections. This note points out that construction of a new library of infrared (2.1-25.0 microns) mineral spectra is in progress. Both transmittance and reflectance measurements of a selection of 63 different, well-characterized minerals have been published to date. These data are available in both hard copy and digital form.
