Liquid Chromatography Mass Spectrometry study of eutectic bis(2,2-dinitropropyl) acetal/formal

This study is the first attempt to document methodology development undergone using liquid chromatography tandem quadrupole time of flight mass spectrometry (LC-QTOF) to investigate degradation products of eutectic bis(2,2-dinitropropyl) acetal/formal nitroplasticizer (called NP here). Method properties investigated are: desolvation temperature (°C) and spray voltage (V) of the electrospray ionization source, and the development of an acetone system rinse to prevent any residual contamination between sample injections. Details are given on why it is essential to investigate method optimization with changes shown in MS/MS analysis in addition to MS results. Trends in MS/MS analytic results reveal important relationships between baseline and aged materials. In addition to verification of previously proposed fragments, insights offered by this newly developed methodology will also identify new degradation products and shed light on the complexity of NP degradation chemistry.

Identification of 2,2-dinitropropanol, a Hydrolyzed Product of Aged Eutectic Bis(2,2-dinitropropyl) Acetal – Bis(2,2-dinitropropyl) Formal Mixture

Eutectic bis(2,2-dinitropropyl) acetal - bis(2,2-dinitropropyl) formal mixture, nitroplasticizer (herein called NP) has historically been produced by either the ter Meer or oxidative nitration synthesis process, wherein 2,2-dinitropropanol (DNPOH) is produced as an intermediate step in both processes. Therefore that DNPOH, could be present in NP either as a production or hydrolysis degradation product is worth investigation. Here, we synthesized DNPOH, validated the synthesis using NMR, and identified DNPOH in aged NP using liquid chromatography tandem time of flight – quadrupole mass spectrometry (LC-QTOF). Using these results, for the first time we positively identify that DNPOH is absent from NP after its production, but is present as a degradation product through hydrolysis from a thermo-chemical aging profile of NP. To hydrolyze NP, prerequisite is the presence of both water and acid. Despite the presence of water in NP, DNPOH is only generated in the late stage of the aging process, when acid concentration is sufficiently high. It has been previously shown both theoretically and experimentally that a primary step of NP degradation is HONO elimination followed by decomposition, wherein nitric acid, nitrous acid, and water are produced (NP→NP’+2HONO and 2HONO→NO+NO 2 +H 2 O). It is shown from this reaction series that water slows HONO decomposition and therefore in small quantities, 100 s of ppm, water actually stabilizes NP against hydrolysis by equilibrium and reducing acidity.

Combining cross-pivot flexures to generate improved kinematically equivalent flexure systems

In this work, we show that new kinematic equivalents with improved performance of standard flexure elements can be systematically synthesized by combination of cross-pivot flexures. Cross-pivot flexures provide a unique feature of kinematic stability under both high loading and large displacement conditions which can be exploited to synthesize a range of kinematic equivalents to standard flexure elements which retain much greater stiffness, load capacity and range capacity than the traditional elements. Cross-pivot synthetic elements provide a means to expand the performance of a large range of flexure-based structures including motion stages, manufacturing equipment and optical systems. This could result in better data collection, smaller systems, and less distortion in operation.

Vapor kinetic energy for the detection and understanding of atmospheric rivers

Poleward water vapor transport in the midlatitudes mainly occurs in meandering filaments of intense water vapor transport, spanning thousands of kilometers long and hundreds of kilometers wide and moving eastward. The water vapor filaments are known as atmospheric rivers (ARs). They can cause extreme wind gusts, intense precipitation, and flooding along densely populated coastal regions. Many recent studies about ARs focused on the statistical analyses of ARs, but a process-level understanding of ARs remains elusive. Here we show that ARs are streams of air with enhanced vapor kinetic energy (VKE) and derive a governing equation for Integrated VKE to understand what contributes to the evolution of ARs. We find that ARs grow mainly because of potential energy conversion to kinetic energy, decay largely owing to condensation and turbulence, and the eastward movement is primarily due to horizontal advection of VKE. Our VKE framework complements the integrated vapor transport framework, which is popular for identifying ARs but lacks a prognostic equation for understanding the physical processes.

A hybrid topological quantum state in an elemental solid

Topology and interactions are foundational concepts in the modern understanding of quantum matter. Their nexus yields three important research directions: (1) the competition between distinct interactions, as in several intertwined phases, (2) the interplay between interactions and topology that drives the phenomena in twisted layered materials and topological magnets, and (3) the coalescence of several topological orders to generate distinct novel phases. The first two examples have grown into major areas of research, although the last example remains mostly unexplored, mainly because of the lack of a material platform for experimental studies. Here, using tunnelling microscopy, photoemission spectroscopy and a theoretical analysis, we unveil a ‘hybrid’ topological phase of matter in the simple elemental-solid arsenic. Through a unique bulk-surface-edge correspondence, we uncover that arsenic features a conjoined strong and higher-order topology that stabilizes a hybrid topological phase. Although momentum-space spectroscopy measurements show signs of topological surface states, real-space microscopy measurements unravel a unique geometry of topologically induced step-edge conduction channels revealed on various natural nanostructures on the surface. Using theoretical models, we show that the existence of gapless step-edge states in arsenic relies on the simultaneous presence of both a non-trivial strong Z 2 invariant and a non-trivial higher-order topological invariant, which provide experimental evidence for hybrid topology. Finally, our study highlights pathways for exploring the interplay of different band topologies and harnessing the associated topological conduction channels in engineered quantum or nano-devices.

Hot electron preheat effects in two-photon polymerization plastic lattice laser-driven shock-tube experiments on the OMEGA laser

We present simulations of Omega shock tube experiments designed to investigate hot electron preheat effects in 3D-printed, two-photon polymerization (2PP) plastic lattices. Preheat is inferred in the experiments from the expansion of a plastic witness disk embedded in the lattice. Using the Eulerian radiation-hydrodynamics code xRAGE, we model shock propagation and preheat from both radiative and hot electron energy sources to evaluate their relative impact. To simulate the transport of laser-generated hot electrons, the nonlocal electron heat transport model proposed by Schurtz, Nicolaï, and Busquet (SNB) is extended with a hot electron source term and an energy cascade algorithm. We explore how variations in ablator, lattice geometry, and laser drive affect the shock velocity and witness disk expansion. Simulations show that the inclusion of a 5 μm gold layer reduces shock pressure by 60% and shock speeds by 30%–40% but does not significantly reduce the hot electron preheat, and that different lattice geometries lead to enhanced shock velocities—up to 40% faster than in homogeneous foams. However, radiative and conductive preheat from classical mechanisms alone fail to match experiment. By including a hot electron source term, we reproduce experimental observables such as disk expansion rates and spatial radiographic features. We find that a hot electron population corresponding to 4%–8% of the incident laser energy with T hot = 50 keV produces expansion which agrees with the experimental data, suggesting hot electron preheat is the most plausible explanation.

Metabolic flux, metabolite, and transcript analysis uncover reprogramming of metabolism toward higher seed oil

Overexpression of WRINKLED1 (WRI1), a master regulator of glycolysis and fatty acid biosynthesis, together with DIACYLGLYCEROL ACYLTRANSFERASE1 (DGAT1), which catalyzes the final step of triacylglycerol assembly, is a promising strategy for enhancing seed oil content. However, how these regulators coordinate system-wide metabolic reprogramming at the levels of gene expression, metabolite pools, and fluxes remains poorly understood. To address this, we performed 13 C-metabolic flux analysis, metabolomics, and transcriptomics on in vitro cultured pennycress (Thlaspi arvense L.) embryos overexpressing the native WRI1 and DGAT1 homologs. Here, in cultured embryos, WRI1/DGAT1 overexpression increased triacylglycerol accumulation by 28% while reducing protein content by 34%, relative to the wild type. Embryos showed ∼20-fold and 50-fold upregulation of WRI1 and DGAT1 along with induction of WRI1 target genes in glycolysis and fatty acid biosynthesis. Genes associated with photosynthesis and Calvin cycle functions were also upregulated, whereas genes encoding ribosomal proteins and seed storage proteins were strongly repressed, consistent with the observed lipid–protein tradeoff. Flux analysis revealed that enhanced triacylglycerol biosynthesis is supported by increased flux through the Rubisco shunt and cytosolic pyruvate kinase, while the oxidative pentose phosphate pathway and malic enzyme contributed little to NADPH or pyruvate supply. Metabolomic profiling revealed extensive perturbations in glycolytic intermediates, tricarboxylic acid cycle metabolites, and amino acids. In plant grown seeds, WRI1/DGAT1 lines also showed a modest but significant increase in total lipid content. Collectively, these findings reveal how WRI1 and DGAT1 reprogram central metabolism to enhance oil accumulation, with relevance to mature seeds.

Induction of durable remission by dual immunotherapy in SHIV-infected ART-suppressed macaques

The eradication of the viral reservoir represents the major obstacle to the development of a clinical cure for established HIV-1 infection. Here, we demonstrate that the administration of N-803 (brand name Anktiva) and broadly neutralizing antibodies (bNAbs) results in sustained viral control after discontinuation of antiretroviral therapy (ART) in simian-human AD8 (SHIV-AD8)–infected, ART-suppressed rhesus macaques. N-803+bNAbs treatment induced immune activation and transient viremia but only limited reductions in the SHIV reservoir. Furthermore, upon ART discontinuation, viral rebound occurred in all animals, which was followed by durable control in approximately 70% of all N-803+bNAb–treated macaques. Viral control was correlated with the reprogramming of CD8 + T cells by N-803+bNAb synergy. Thus, complete eradication of the replication-competent viral reservoir is likely not a prerequisite for the induction of sustained remission after discontinuation of ART.

Liquid Chromatography Mass Spectrometry Instrumentation and Methodology

This report reviews instrumentation and techniques utilized in liquid chromatography mass spectrometry (LC-MS). The analytical advantages and disadvantages of each major component of different LC-MS systems are discussed, along with a comparison between LC-MS, liquid chromatography tandem mass spectrometry (LC-MS/MS), and gas chromatography mass spectrometry (GC-MS). This report also details future work regarding the use of LC-MS/MS to analyze and identify degradation products from an extensive accelerated aging process of nitroplasticizer (NP). This report is the first of a series on the use of LC-MS to understand the degradation mechanisms of NP.

CERES: CRISPR Engineering for the Rapid Enhancement of Strains

Previous strain development efforts for cyanobacteria have failed to achieve the necessary productivities needed to support economic biofuel production. We proposed to develop CRISPR Engineering for Rapid Enhancement of Strains (CERES). We developed genetic and computational tools to enable future high-throughput screening of CRISPR interference (CRISPRi) libraries in the cyanobacterium Synechococcus sp. PCC 7002, including: (1) Operon- SEQer: an ensemble of algorithms for predicting operon pairs using RNA-seq data, (2) experimental characterization and machine learning prediction of gRNA design rules for CRISPRi, and (3) a shuttle vector for gene expression. These tools lay the foundation for CRISPR library screening to develop cyanobacterial strains that are optimized for growth or metabolite production under a wide range of environmental conditions. The optimization of cyanobacterial strains will directly advance U.S. energy and climate security by enabling domestic biofuel production while simultaneously mitigating atmospheric greenhouse gases through photoautotrophic fixation of carbon dioxide.

Aging and Lifetimes Program Annual Report - Part I: Update to BDNPA-BDNPF phase diagram (FY2022)

The eutectic mixture nitroplasticizer (NP) contains water, without exception, on the order of hundreds to thousands of parts per million depending on the exposure conditions. Therefore, to evaluate the properties of NP, one must consider that NP is not simply comprised of two constituents. Here we supplement previous work on the physical properties of NP (Edgar et al., 2020) by expanding on the experimental work therein and addressing inaccuracies detailed in a subsequent comment article (Brown, 2021). Specifically, the purpose of this work is to clarify and modify the phase diagram of the bis(2,2-dinitropropyl) acetal with bis(2,2-dinitropropyl) formal eutectic mixture by considering the effect of water concentrations on the nucleation process.

Cleaner Cooking: Exploring Tools to Measure and Understand the Long-term Adoption and Environmental Significance of Cookstoves in India

About 40% of the world’s population, or roughly 3 billion people, rely on solid biomass fuels like coal, wood, dung, and crop residues to cook and meet their household energy needs. This outdated energy system has severe social, health, and environmental implications. Women are disproportionately affected as they predominantly bear the burden of cooking and collecting fuelwood, which exacerbates the “time poverty” trap that restricts them from participating in economic and educational activities. Exposure to indoor solid fuel combustion, also known as household air pollution, is responsible for 3-4 million premature deaths per year and is a leading risk factor for chronic obstructive pulmonary disease, childhood pneumonia, stroke, ischemic heart disease, and lung cancer. Solid-fuel cooking contributes to 16% of global ambient air pollution, emitting CO 2 and other climate-forcing pollutants like carbon monoxide, black carbon, and methane.

A mechanistic model for creep and thermal aging in Alloy 709

This report describes a physics-based model for creep and thermal aging in Alloy 709. Alloy 709 is an advanced austenitic alloy, targeted for use in future Sodium Fast Reactors (SFRs) and other advanced reactors. The material has superior high temperature properties compared to currently qualified 316 and 304 stainless steels. However, the available creep and thermal aging test database for Alloy 709 is significantly more limited compared to the historical materials. The physics-based model developed here is one way to accelerate the qualification of the material by providing more accurate long-term predictions for creep properties and thermal aging, compared to current empirical time-extrapolate techniques. The crystal plasticity finite element model is used to predict the deformation and failure of alloy 709. The same setup for the CPFE model is used in both the baseline model calibration process and the simulation campaigns for parameter inference. Specific constitutive choices are made for Alloy 709 to capture the primary deformation mechanisms. The dislocation creep formulation developed by Hu and Cocks is extended to account for coupled precipitation formation and the grain boundary cavitation model developed by Sham, Needleman, et al. is used to model grain boundary cavitation-induced failure. A novel update algorithm is proposed to render the semi-discrete constitutive update for the Sham-Needleman model unconditionally stable. A progressive calibration approach is adopted based on the observations that several types of material responses can be effectively decoupled. A surrogate model is trained based on full-fledged CPFE simulations to accelerate the forward model evaluations, and stochastic variational inference (SVI) is used to calibrate the unknown microstructural model parameters. The calibrated mechanistic model is used to predict the long-term creep life of Alloy 709, and the predictions are compared against classical empirical approaches.
