Editorial: Multi-Omics Technologies for Optimizing Synthetic Biomanufacturing

Industrial manufacturing endures as an essential human activity yielding a variety of useful products; it plays a significant role in the global economy with huge impacts in everyday life. However, the manufacturing process requires consumption of various raw materials (especially petroleum derivatives), generates a variety of harmful waste products, causes pollution, and is energetically inefficient. Biological manufacturing from sustainable, affordable, and scalable feedstocks potentially enables the displacement of the entire portfolio of currently available products produced by industrial processes, enabling the manufacturing of renewable and eco-friendly products (Clomburg et al., 2017). Thus, successful development of a robust biomanufacturing strategy and technology platform, based on the latest advances in synthetic biology and chemical catalysis, will decrease both the cost and production time compared with previous manufacturing processes. Development of biomanufacturing processes using a synthetic biology platform requires the multidisciplinary efforts of science and engineering fields including molecular biology, microbiology, genetic engineering, informatics, metabolic modeling and chemical or process engineering (El Karoui et al., 2019).

Systematic analysis of Baobaoqu fermentation starter for Wuliangye Baijiu by the combination of metagenomics and metabolomics

Baobaoqu (BBQ) is a traditional fermenting power, which is widely applied in Nong-flavor Baijiu brewing. There are two different types of BBQ (premium BBQ and normal BBQ) used in industrial manufacture, but the reasons for the significant differences between two kinds of BBQ have not been clearly illuminated. In this study, the combination of metagenomics and metabolomics was performed to compare the differences in the composition of microbial communities and the components of flavors between premium BBQ and normal BBQ. The results showed that the glycosidase-producing microorganisms are the biomarkers of premium BBQ, contributing a better ability of carbon source utilization than normal BBQ. In addition, several important flavors (ethyl hexanoate, phenylethanol, ethyl acetate) were rich in normal BBQ, which have a significant positive correlation with the biomarkers ( Lactobacillus and Pichia kudriavzevii ) of normal BBQ. It suggests that the microbial community has an advantage in utilizing raw materials in premium BBQ, while the community was inclined to form flavors in normal BBQ. The differences between two types of BBQ at the microbial and flavor level have theoretical and practical guiding significance in the application of premium and normal BBQ and in the further improvements of taste and quality of Baijiu.

Methodology for assessing the maximum potential impact of separations opportunities in industrial processes

Separation technologies currently used in U.S. manufacturing industries are estimated to account for more than 20% of plant energy consumption. However, accurately determining the impact of new separation technology solutions can sometimes be difficult, especially when evaluating a slate of new candidate separation technologies, each of which has its own separation performance, energy demand, and capital cost. In these cases, a typical approach is to assess each new separation technology by collecting performance and cost information and then using that information to develop a techno-economic analysis to identify overall benefits. While this approach is thorough, it can be time consuming and can hinder reaching a critical understanding of the potential of a given separation challenge, especially when there is no known solution. To address these issues, we developed an assessment methodology, using industrial screening processes, that can be used to better understand the potential impacts of addressing a given separation challenge. This paper presents an overview of our separation challenge stream assessment methodology. The methodology involves defining an “ideal” separator and deriving the associated minimum separation energy. The “ideal” separator represents the most optimistic outlook of a given opportunity so the maximum impact from existing and not-yet-developed solutions can be assessed. Using established biorefinery models, we applied the methodology to 10 different separation challenge streams from two different biomass conversion platforms to identify the type of information that can be obtained. Three of the ten challenge streams assessed had maximum possible cost savings predictions >20%, and associated reductions in process energy carbon intensity ranging from 0 to 54%. Two streams had cost and energy savings potential that were < 5%. Some of the opportunity drivers from the various assessments include higher product yields, reduction or elimination of downstream equipment, new co-products, and cost savings associated with raw materials and energy consumption. The information from these assessments can help guide the selection or development of new separation technology solutions based on the various potential factors that drive the projected benefits.

Survey of Thirteen Novel Pseudomonas putida Bacteriophages

Bacteriophages have been widely investigated as a promising treatment of food, medical equipment, and humans colonized by antibiotic-resistant bacteria. Phages pose particular interest in combating those bacteria which form biofilms, such as the medically important human pathogen Pseudomonas aeruginosa and several plant pathogens, including P. syringae . In an undergraduate lab course, P. putida was used as the host to isolate novel anti-pseudomonal bacteriophages. Environmental samples of soil and water were collected, and purified phage isolates were obtained. After Illumina sequencing, genomes of these phages were assembled de novo and annotated. Assembled genomes were compared with known genomes in the literature and GenBank to identify taxonomic relations and to refine their functional annotations. The thirteen phages described are sipho-, myo-, and podoviruses in several families of Caudoviricetes , spanning several novel genera, with genomes ranging from 40,000 to 96,000 bp. One phage (DDSR119) is unique and is the first reported P. putida siphovirus. The remaining 12 can be clustered into four distinct groups. Six are highly related to each other and to previously described Autotranscriptaviridae phages: Waldo5, PlaquesPlease, and Laces98 all belong to the Waldovirus genus, whereas Stalingrad, Bosely, and Stamos belong to the Troedvirus genus. Zuri was previously classified as the founding member of a new genus Zurivirus within the family Schitoviridae . Ebordelon and Holyagarpour each represent different species within Zurivirus , whereas Meara is a more distantly related member of the Schitoviridae . Dolphis and Jeremy are similar enough to form a genus but have only a few distant relatives among sequenced phages and are notable for being temperate. We identified the lysis cassettes in all 13 phages, compared tail spike structures, and found auxiliary metabolic genes in several. Studies like these, which isolate and characterize infectious virions, enable the identification of novel proteins and molecular systems and also provide the raw materials for further study, evaluation, and manipulation of phage proteins and their hosts.

Calcium Carbonate Cement: A Carbon Capture, Utilization, and Storage (CCUS) Technique

A novel calcium carbonate cement system that mimics the naturally occurring mineralization process of carbon dioxide to biogenic or geologic calcium carbonate deposits was developed utilizing carbon dioxide-containing flue gas and high-calcium industrial solid waste as raw materials. The calcium carbonate cement reaction is based on the polymorphic transformation from metastable vaterite to aragonite and can achieve >40 MPa compressive strength. Due to its unique properties, the calcium carbonate cement is well suited for building materials applications with controlled factory manufacturing processes that can take advantage of its rapid curing at elevated temperatures and lower density for competitive advantages. Examples of suitable applications are lightweight fiber cement board and aerated concrete. The new cement system described is an environmentally sustainable alternative cement that can be carbon negative, meaning more carbon dioxide is captured during its manufacture than is emitted.

TiNi-Based Bi-Metallic Shape-Memory Alloy by Laser-Directed Energy Deposition

In this study, laser-directed energy deposition was applied to build a Ti-rich ternary Ti–Ni–Cu shape-memory alloy onto a TiNi shape-memory alloy substrate to realize the joining of the multifunctional bi-metallic shape-memory alloy structure. The cost-effective Ti, Ni, and Cu elemental powder blend was used for raw materials. Various material characterization approaches were applied to reveal different material properties in two sections. The as-fabricated Ti–Ni–Cu alloy microstructure has the TiNi phase as the matrix with Ti2Ni secondary precipitates. The hardness shows no high values indicating that the major phase is not hard intermetallics. A bonding strength of 569.1 MPa was obtained by tensile testing, and digital image correlation reveals the different tensile responses of the two sections. Differential scanning calorimetry was used to measure the phase-transformation temperatures. The austenite finishing temperature of higher than 80 °C was measured for the Ti–Ni–Cu alloy section. For the TiNi substrate, the austenite finishing temperature was tested to be near 47 °C at the bottom and around 22 °C at the upper substrate region, which is due to the repeated laser scanning that acts as annealing on the substrate. Finally, the multiple shape-memory effect of two shape-memory alloy sides was tested and identified.

Prediction of Silicon Content in a Blast Furnace via Machine Learning: A Comprehensive Processing and Modeling Pipeline

Silicon content plays an important role in determining the operational efficiency of blast furnaces (BFs) and their downstream processes in integrated steelmaking; however, existing sampling methods and first-principles models are somewhat limited in their capability and flexibility. Current data-based prediction models primarily rely on a limited set of manually selected furnace parameters. Additionally, different BFs present a diverse set of operating parameters and state variables that are known to directly influence the hot metal’s silicon content, such as fuel injection, blast temperature, and raw material charge composition, among other process variables that have their own impacts. The expansiveness of the parameter set adds complexity to parameter selection and processing. This highlights the need for a comprehensive methodology to integrate and select from all relevant parameters for accurate silicon content prediction. Providing accurate silicon content predictions would enable operators to adjust furnace conditions dynamically, improving safety and reducing economic risk. To address these issues, a two-stage approach is proposed. First, a generalized data processing scheme is proposed to accommodate diverse furnace parameters. Second, a robust modeling pipeline is used to establish a machine learning (ML) model capable of predicting hot metal silicon content with reasonable accuracy. The method employed herein predicted the average Si content of the upcoming furnace cast with an accuracy of 91% among 200 target predictions for a specific furnace provisioned by the XGBoost model. This prediction is achieved using only the past shift’s operating conditions, which should be available in real time. This performance provides a strong baseline for the modeling approach with potential for further improvement through provision of real-time features.

Global Regulations for Sustainable Battery Recycling: Challenges and Opportunities

With the rapid expansion of transportation electrification worldwide, the demand for electric vehicles (EVs) has increased dramatically, creating new and sustainable growth opportunities for the global economy. However, as the most expensive component of EVs, lithium-ion batteries pose significant sustainability challenges due to raw material consumption and supply chain constrains, as well as the complexities of end-of-life battery disposal and recycling. To address these concerns, many countries are actively establishing regulations to promote sustainable pathways for battery reuse and recycling. Despite these efforts, existing battery recycling regulations remain often inefficient and vary significantly across different countries in legal enforcement, producer responsibility, waste classification, recycling targets, design standards, public engagement, and financial incentives, particularly given the complexities of the global supply chain and resource distribution within the battery industry. Understanding these regulatory differences and establishing a unified framework are therefore crucial to ensuring sustainable and efficient battery recycling. This review provides a comprehensive analysis of the necessity of establishing robust regulations for sustainable development of battery recycling industry. The evolution and refinement of battery recycling regulations are deeply reviewed to identifying persistent gaps and challenges in key countries. Furthermore, we discuss the challenges associated with regulatory enforcement and propose strategies for developing a more cohesive legislative framework to ensure the effective utilization of retired batteries.

Optimization of a Lightweight Floating Offshore Wind Turbine with Water Ballast Motion Mitigation Technology

Floating offshore wind turbines are a promising technology for addressing energy needs by utilizing wind resources offshore. The current state of the art is based on heavy, expensive platforms to survive the ocean environment. Typical design techniques do not involve optimization because of the computationally expensive time domain solvers used to model motions and loads in the ocean environment. However, this design uses an efficient frequency domain solver with a genetic algorithm to rapidly optimize the design of a novel floating wind turbine concept. The concept utilizes a liquid ballast mass to mitigate motions on a lightweight post-tensioned concrete platform. The simple cruciform-shaped design of the platform made of post-tensioned concrete is less expensive than steel, reducing the raw material and manufacturing cost. The use of ballast water to behave as a tuned mass damper allows a smaller platform to achieve the same motions as a much larger platform, thus reducing the mass and cost. The optimization techniques applied with these design innovations resulted in a design with a levelized cost of energy of USD 0.0753/kWh, roughly half the cost of the current state of the art.

America's Strategy to Secure the Supply Chain for a Robust Clean Energy Transition Factsheet

Demand for clean energy technologies such as wind turbines and batteries for electric vehicles has increased significantly as technology costs have plummeted over the last decade and countries seek to diversify their energy systems with more reliable clean energy sources to lower costs for businesses and consumers. The global clean energy market is expected to grow exponentially — reaching $23 trillion at a minimum by 2030. Without new domestic raw materials production and manufacturing capacity, the U.S. will continue to rely on clean energy imports, exposing the nation to supply chain vulnerabilities while simultaneously losing out on the enormous job opportunities associated with the energy transition. Yet, in many cases, the United States has untapped potential to support greater domestic production.

Achieving American Leadership in the Rare Earth Magnets Supply Chain Factsheet

Rare earth permanent magnets are a critical technology for the clean energy transition and electrified transport revolution. The global supply chain for rare earth metals and magnets is almost completely dominated by China, which controlled 58% of rare earth mining and 92% of magnet manufacturing in 2020. The United States produces only 15% of the global supply of raw materials for these magnets, and while the nation has several nascent downstream processing and manufacturing efforts, current domestic production is limited. Investments and incentives to boost the domestic supply chain, especially in the separation and refining phases, point the way to securing stable supplies of these critical clean energy components.

Sustainable Manufacturing and the Circular Economy

The manufacturing industry predominantly follows a linear model of production in which raw materials are mined or grown, transformed into manufactured goods, used for a period, and then discarded. However, this once-through flow of materials to manufactured products is not sustainable. Many high-grade ore bodies have been consumed, increasing the energy and material impacts per ton of mined material. At the other end of the cycle, there are challenges with the efficiency and effectiveness of our waste disposal system.
