Application of the Metabolic Modeling Pipeline in KBase to Categorize Reactions, Predict Essential Genes, and Predict Pathways in an Isolate Genome

The DOE Systems Biology Knowledgebase (KBase) platform offers a range of powerful tools for the reconstruction, refinement, and analysis of genome-scale metabolic models built from microbial isolate genomes. In this chapter, we describe and demonstrate these tools in action with an analysis of isoprene production in the Bacillus subtilis DSM genome. Two different methods are applied to build initial metabolic models for the DSM genome, then the models are gapfilled in three different growth conditions. Next, flux balance analysis (FBA) and flux variability analysis (FVA) techniques are applied to both study the growth of these models in minimal media and classify reactions within each model based on essentiality and functionality. The models are applied with the FBA method to predict essential genes, which are then compared to an updated list of essential genes obtained for B. subtilis 168, a very similar strain to the DSM isolate. The models are also applied to simulate Biolog growth conditions, and these results are compared with Biolog data collected for B. subtilis 168. Finally, the DSM metabolic models are applied to explore the pathways and genes responsible for producing isoprene in this strain. These studies demonstrate the accuracy and utility of models generated from the KBase pipelines, as well as exploring the tools available for analyzing these models.

Airborne infectious disease isolation units and method of making using prefabricated containers

In one embodiment, a prefabricated container is modified as a medical isolation room by replacing an original door with a clear front door. An intake louver and an adjustable damper are disposed at a lower part of the front end. An exhaust vent is disposed at an upper part of the back wall. An exhaust fan and a HEPA filter are coupled to the exhaust vent. Washable coverings cover interior sides of the container to provide washable, nonslip interior surfaces. The exhaust fan and the adjustable damper at the intake louver are controlled to produce in the medical isolation room a negative air pressure of at least about minus 0.01 inch of water gage (approximately 2.5 pascals) and a displacement ventilation exhaust flow rate through the exhaust vent of at least about 100 cubic feet per minute (cfm) greater than an intake flow rate through the intake louver.

Deep Isolation-Development of the Safety Case for Disposal of Radioactive Wastes in Horizontal Boreholes - 20028

Deep Isolation has developed a safe, secure, and permanent geological disposal method for high-level waste, including spent nuclear fuel as well as sealed sources and other highly radioactive materials. The method leverages well established directional drilling technology to create horizontal repositories deep underground. The combination of great depth (1-3 km) and the ability to precisely position repositories in a horizontal orientation provides access to geologic strata that are inaccessible for typical mined and deep vertical borehole repositories. A number of potential direct and indirect safety benefits accrue including:1) greater depth below regional freshwater aquifers; 2) increased flexibility and more geologic options for siting; 3) access to deep formations with persistent and sustained reducing conditions. In addition there are a number of safety elements related specifically to the horizontal repository geometry including: 1) passive direction of thermally driven fluid and radionuclide movement away from the vertical access hole and toward the 'dead end' portion of the repository; 2) mitigation of seismic hazards by orienting repositories parallel to local and regional fault structures. In this paper we explore and discuss some of the key technologic, geologic and hydrologic elements that support the deep horizontal borehole safety case. The stalemate seen across the globe on the disposal of spent nuclear fuel and high-level waste can be broken. Deep Isolation offers a novel option for safe, secure, and permanent deep geological disposal of nuclear waste that can be developed as centralized repositories or adapted to smaller regional or site-specific repositories located near waste sources. Isolation and spectroscopy of C 2 H + ions in helium droplets

Ethynylium (C 2 H + ) may be an important intermediate in astrochemistrybut remains poorly studied by laboratory techniques due to its high reactivity. Immersion of single acetylene molecules in helium nanodroplets followed by electron impact ionization effectively generates and isolates C 2 H + cations in an inert cryogenic environment, enabling infrared spectroscopic measurements. Here, we find that in the C-H stretching range, the C 2 H + spectrum appears as a triplet with splitting of 33.6 cm —1 and 37.5 cm —1 . The splitting is temporarily assigned to the product of interactions between the C-H stretching mode and overtones of the bending mode.

Structural and Catalytic Properties of Isolated Pt 2+ Sites in Platinum Phosphide (PtP 2 )

This article describes the synthesis and catalytic properties of supported, 2-3 nm platinum phosphide (PtP 2 ) nanoparticles (NPs). Depending on the P loading, two PtP 2 structures are formed, that is, a PtP 2 surface on a (metallic) Pt core (Pt@PtP 2 ) and single-phase PtP 2 NPs. The structures were determined using extended X-ray absorption fine structure , in situ synchrotron X-ray diffraction, and scanning transmission electron microscopy. In PtP 2 NPs, Pt 2+ ions are geometrically isolated by P 2 2- ions, at a Pt-Pt distance of 4.02 Å, which is much longer than 2.78 Å in (metallic) Pt NPs. The oxidation state of Pt in PtP 2 NPs was determined by in situ X-ray absorption near-edge structure and in situ X-ray photoelectron spectroscopy and was found to be consistent with Pt 2+ ions even after treatment in H 2 at 550 °C. Unlike Pt NPs, which are highly active for propylene hydrogenation at room temperature, PtP 2 NPs are not active below about 150 °C, suggesting the absence of metallic surface Pt. In contrast to metallic Pt, which is poorly selective for acetylene hydrogenation, PtP 2 NPs display high selectivity toward ethylene. PtP 2 , also has high olefin selectivity for propane dehydrogenation, although the rate per g Pt is about 7 times lower than that of metallic Pt NPs of the same size. In situ resonant inelastic X-ray scattering spectroscopy shows that the energy of the filled Pt 5d valence orbitals is 1.5 eV lower than that of metallic Pt, which leads to weaker adsorbate binding consistent with its catalytic properties. Here, a H 2 -stable Pt 2+ site suggests different catalytic applications for these catalysts as compared to Pt NPs.

Cyclically symmetric radially self-similar phononic pseudocrystal isolator for broadband, ultrasonic vibration bandstop filtering

A 2D phononic pseudocrystal isolator exhibiting cyclic symmetry and radial self-similarity is measured and demonstrated to block a wide range of ultrasonic vibration. Measurements of longitudinal and shear wave blocking effects are made and compared with computational results. The use of the bandgap edge ratio is recommended for quantifying suppression in very-wide-bandgap materials. In conclusion, the upper-to-lower suppression edge frequency ratios of 3–4 are remarkably large for shear waves and even larger for longitudinal waves upper-to-lower suppression ratio (13 at 5 dB), such that 92.5% of frequencies in that range experience ≥ 5 dB of suppression.

Isolated Single-stage Three-phase AC/DC Converter using Bidirectional Switches

The advent of the SiC Bidirectional FET (BiDFET), a monolithic 1.2 kV bidirectional switch, has rendered the single-stage three-phase AC/DC converter topology a promising approach for implementing AC/DC converters. This topology, which integrates a full-bridge converter with a single-phase to three-phase matrix converter via a high-frequency transformer, is particularly suitable for applications requiring galvanic isolation, buck-boost functionality, and bidirectional power flow. The single-stage design eliminates the need for bulky and unreliable electrolytic capacitors, and utilizes a single magnetic component for power transfer. In the matrix converter, bidirectional switches, which were traditionally implemented using combinations of multiple semiconductor devices such as MOSFETs, IGBTs, and diodes, can now be realized using the single-chip solution, BiDFET. This advancement leads to a lower switch count, compact converter implementation, with lower inductance commutation cells, thereby enhancing the overall efficiency and compactness of the system. The paper presents a unified model of the converter, considering all control parameters, including the duty cycles and phase shift of transformer voltages. Detailed expressions for power transfer, transformer currents, and currents at AC and DC ports are provided. Additionally, the paper outlines the conditions necessary for soft-switching of all switches and the commutation schemes required for the practical implementation of the matrix converter modulation scheme. A hardware prototype of a 10 kW, 480 VRMS, LL/ 800 V AC/DC system has been developed, and experimental results are presented to demonstrate its performance.

Medium-Voltage Isolated Auxiliary Power Supply Design for High Insulation Capability, Ultra-Low Coupling Capacitance, and Small Size

To fully utilize the benefits of medium-voltage (MV) silicon carbide (SiC)-based converter and achieve high power density, the isolated auxiliary power supply (IAPS) needs to be as small as possible. However, the high insulation and low coupling capacitance requirements, considering the high voltage and high dv / dt , are constraints of size reduction. This article proposes an MV IAPS design for high insulation capability, ultra-low coupling capacitance, and small size. The topology selection, efficiency, load regulation, coupling capacitance, and size reduction considerations are discussed. A 2.5 W MV IAPS is designed and built for a 10 kV SiC mosfet -based 13.8 kV/100 kW three-phase converter, achieving a partial discharge (PD) inception voltage of higher than 15 kV rms, ultra-low coupling capacitance of 1.03 pF, and smaller size than that of the commercial products as well as the state-of-art research prototypes. Experimental results of PD test, 10 kV SiC mosfet -based half-bridge test, and 13.8 kV/100 kVA three-phase converter full rating test are conducted for design validation.

Utah FORGE 1-2551: Multi-Stage Fracturing System and Well Tractor to Enable Zonal Isolation During Stimulation and EGS Operations in Horizontal Wellbores - Workshop Presentation and Report

Included here are a presentation recording, slides, and report on the Multi-Stage Fracturing System and Well Tractor to Enable Zonal Isolation During Stimulation and EGS Operations in Horizontal Wellbores project by Colorado School of Mines, presented by Dr. William W. Fleckenstein. One goal of the project is to develop and demonstrate the use of low-cost, rapid multistage fracture stimulation technology with cemented casing frac sleeves that eliminates packers used in both conventional stimulation and injection flow control. The second goal is to develop and demonstrate the devices to effectively detect and control the flow of heat-carrier fluid solely from a long-reach injector through the network of induced and existing fractures and produced from an open-hole long-reach well to improve heat recovery. This presentation was featured in the Utah FORGE R&D Annual Workshop on September 7, 2023. The workshop provided a valuable opportunity to explore the progress made in each of the 17 Research and Development projects funded under Solicitation 2020-1 which aim to enhance our understanding of the crucial factors influencing the development of Enhanced Geothermal Systems (EGS) reservoirs and resources.

Lab Home Testing of Residential Isolation Space Control to Minimize Infectious Disease Transmission in Existing Single-Family Homes

Existing evidence strongly suggests that viral infectious diseases can be transmitted via an airborne route across distances in indoor environments. Accordingly, the risk of airborne transmission within homes should be managed. The public health emergency associated with SARS-CoV-2 makes controlling airborne transmission of respired viruses in indoor environments critical, especially in poorly ventilated indoor environments. The effectiveness of engineering interventions requiring minor efforts that create a negative-pressure isolation zone (IZ) for a contagious person has yet to be tested for existing residential homes. To mitigate the risk of airborne virus transmission and maximize health protection for the population in existing single-family homes, the relative effectiveness of several control strategies are investigated in this report. While very high-efficiency MERV filtration, high ventilation rates, and other controls can help be effective, most occupants are not likely to have the time or means for advanced measures found in hospitals. This project focused on testing relatively simple efforts that utilize existing or easy to acquire materials and simple processes.

FY 2021 Isolated Grids and Grid-Connected Turbine Reference Systems; Microgrids, Infrastructure Resilience, and Advanced Controls Launchpad (MIRACL)

For individuals, businesses, and communities focused on building resilient electrical grid infrastructure, wind energy can provide an affordable, accessible, and compatible distributed energy resource option that also enhances the capabilities of local grid operations. The Microgrids, Infrastructure Resilience, and Advanced Controls Launchpad (MIRACL) project is a multi-year distributed wind research effort, driven through a partnership between four Department of Energy National Laboratories and industry to develop and improve the planning, design, and operation of wind-centered microgrids to complement solar, energy storage, and other distributed energy resources for grid-tied and isolated operation (U.S. Department of Energy, 2021). This report documents the application of methods developed through the initial three years of the MIRACL project to two real-world distributed wind reference systems. Specifically, the methods demonstrated in this report include 1) a market valuation framework to comprehensively value the services distributed wind can provide and 2) a resilience framework that enables stakeholders to characterize distribution system resilience and compare grid investment decisions from a resilience perspective. Additional methods mentioned in this report include distributed hybrid system design methods for grid resilience, advanced control co-simulation platforms, and power hardware-in-the-loop (PHIL) models. Preliminary results from these additional methods are presented in this report and will be demonstrated and/or applied to the reference systems in the coming year. The purpose of applying these methods to reference systems is to drive technology transfer of the theories, methodologies, and technologies developed under the MIRACL project and increase the number of referenceable case studies available to stakeholders interested in additional value-added capabilities of wind systems beyond bulk energy supply (i.e. kilowatt-hours).

System Study: Isolation Condenser 1998-2020

This report presents an unreliability evaluation of the isolation condenser (ISO) system at four U.S. boiling water reactors. Demand, run hours, and failure data from calendar year 1998 through 2020 for selected components were obtained from the Institute of Nuclear Power Operations (INPO) Industry Reporting and Information System (IRIS), formerly the INPO Consolidated Events Database (ICES). The unreliability results are trended for the most recent 10 year period while yearly estimates for system unreliability are provided for the entire active period. No statistically significant increasing or decreasing trends were identified in the ISO results.

The unreliability results are trended for the most recent 10-year period while yearly estimates for system unreliability are provided for the entire active period. No statistically significant increasing or decreasing trends were identified in the RCIC results.

Modular HF Isolated MV String Inverters Enable a New Paradigm for Large PV Farms

The “Modular HF Isolated MV String Inverters Enable a New Paradigm for Large PV Farms” project focuses on exploring alternative power converter and system-level plant configurations to achieve the lowest cost and highest energy output for a given solar plus storage (e.g., PV + battery) plant, including the use of medium voltage (MV) collection while taking into account detailed models of all elements. To realize this objective, four approaches were utilized (i) employ a novel Medium Voltage String Inverter (MVSI) topology (soft switching solid state transformer – S4T) to convert 1000 Vdc to 4.16 kVac; (ii) plant collection using standard, low-cost overhead MV distribution network; (iii) enable energy storage integration without additional converter cost to achieve dispatchability of the PV resource; and (iv) provide advanced functionality (autonomous operation, track ISO signals for dynamic balancing and ancillary services, and PV farm operation as a virtual grid resource). Subsequently and in alignment with the previously mentioned approaches, the project was structured in five efforts (i) S4T MVSI simulation and design; (ii) system analysis and storage optimization; (iii) financial analysis; (iv) power converter prototype build and test; and (v) regulatory and commercial impact study. The outcomes provided by each effort can be summarized as follows (i) Project explored the use of MV AC distribution architecture for hybrid PV+storage utility-scale PV farms; (ii) Detailed loss and LCOE analysis for AC and DC side BESS architecture, including multiple converter topologies, as well as for proposed MVSI/MDCT systems; (iii) MVSI was built and holds promise but needs lower-cost high-voltage Si-C devices, which does not seem possible in the near term; (iv) MDCT provides a simpler modular building block – validated through HIL and farm level modeling, simulation and experimental validation; (v) 300 kVA MDCT prototype built and tested, technology is being commercialized; and (vi) Regulatory model of utility building PV plants, where PV panels are treated as DC generation (IPP), seems viable and can allow improved grid integration.

Statistically significant decreasing trends were identified in both the RCIC system start-only unreliability and 8-hour mission unreliability.
