Data for "Anti-Pdc1p Nanobody as a Genetically Encoded Inhibitor of Ethanol Production Enables Dual Transcriptional and Post-translational Controls of Yeast Fermentations"

Microbial fermentation provides a sustainable method of producing valuable chemicals. Adding dynamic control to fermentations can significantly improve titers, but most systems rely on transcriptional controls of metabolic enzymes, leaving existing intracellular enzymes unregulated. This limits the ability of transcriptional controls to switch off metabolic pathways, especially when metabolic enzymes have long half-lives. We developed a two-layer transcriptional/post-translational control system for yeast fermentations. Specifically, the system uses blue light to transcriptionally activate the major pyruvate decarboxylase PDC1 , required for cell growth and concomitant ethanol production. Switching to darkness transcriptionally inactivates PDC1 and instead activates the anti-Pdc1p nanobody, NbJRI, to act as a genetically encoded inhibitor of Pdc1p accumulated during the growth phase. This dual transcriptional/post-translational control improves the production of 2,3-BDO and citramalate by up to 100 and 92% compared to using transcriptional controls alone in dynamic two-phase fermentations. This study establishes the NbJRI nanobody as an effective genetically encoded inhibitor of Pdc1p that can enhance the production of pyruvate-derived chemicals.

Project Liberty: Launch of an Integrated Bio-Refinery with Eco-Sustainable and Renewable Technologies. Conversion of Corn Stover Biomass to Bio-Ethanol, Final Report

POET-DSM Advanced Biofuels, in collaboration with parent companies, POET and DSM, has successfully designed, constructed, and continuously operated Project Liberty, a 770 bone-dry ton per day (tpd) nameplate capacity, 20 million gallon (cellulosic bioethanol) per year demonstration scale integrated biorefinery (IBR). The IBR produced cellulosic bio-ethanol from agriculture waste (corn stover) feedstock using next-generation pretreatment and enzymatic conversion technologies. The IBR plant was built and demonstrated operations in Emmetsburg, Iowa. The schedule for this IBR project is illustrated in Figure E1. Due to uncontrollable market factors and COVID-19 pandemic, cellulosic bioethanol production ceased near the end of 2019 and remaining operations were idled June of 2020.

Ethanol to Para-xylene via Methyl Benzaldehyde

Para-xylene is an intermediate compound to several valuable commodity chemicals including polyethylene terephthalate (PET) and currently produced from fossil feedstock through a complex sequence of steps utilizing energy intensive process. Recent work from PNNL has demonstrated the conversion of ethanol produced from waste feedstocks such as municipal solid waste to para-xylene via simple three step process.

Integrated Renewable Combined Heat and Power for Ethanol

Conference presentation at 48th International Technical Conference on Clean Energy (Clearwater Clean Energy Conference), Clearwater, Florida, June 16–19, 2024. This project evaluates the technical and economic feasibility of small‑scale combined heat and power (CHP) systems using corn stover to support bioenergy with carbon capture and storage (BECCS) at ethanol facilities. The work includes resource characterization, pilot‑scale gasification and CO 2 capture testing to support techno‑economic analysis, economic evaluation of CHP deployment and potential carbon revenue, and life‑cycle assessment of gasification‑based CHP systems.

Method of converting ethanol to higher alcohols

A method and catalyst for forming higher alcohols from lower alcohol feedstocks. In one application a highly selective and stable copper pseudo-single-atom supported on MgO—Al2O3 catalyst is provided which provides ethanol condensation to higher alcohols at ˜50% yields and ˜85% selectivity is demonstrated with stable catalyst lifetime over 500 hours in a continuous flow system. In some applications a Guerbet condensation process is further utilized to yield a higher alcohol at a selectivity of near ˜90%.

Low-Temperature Plasma Assisted Kinetics Study of Ethanol

Abstract: Non- equilibrium plasma assisted kinetics of ethanol at 0.5 atm over a temperature range of 523-1203 K were studied at stoichiometric conditions by using a plasma flow reactor (PFR). The effects of plasma chemistry on fuel and its influence on neutral chemistry is decoupled with both experimental and modelling efforts. Enhanced reactivity was observed experimentally in both plasma-assisted oxidation and pyrolysis cases with fuel consumption beginning at temperatures as low as 523 K. Absolute fuel consumption was observed at temperatures nearly 200 K earlier than pure thermal reactions highlighting the effectiveness of non-equilibrium plasma to promote reactivity. Modelling results illustrated the effect of electron impact reactions towards accelerating fuel consumption and promoting low temperature chemistry by generating excited state N2 and reactive O and OH radicals.
