ASTROCULTURE (TM) root metabolism and cytochemical analysis

Physiology of the root system is dependent upon oxygen availability and tissue respiration. During hypoxia nutrient and water acquisition may be inhibited, thus affecting the overall biochemical and physiological status of the plant. For the Astroculture (TM) plant growth hardware, the availability of oxygen in the root zone was measured by examining the changes in alcohol dehydrogenase (ADH) activity within the root tissue. ADH activity is a sensitive biochemical indicator of hypoxic conditions in plants and was measured in both spaceflight and control roots. In addition to the biochemical enzyme assays, localization of ADH in the root tissue was examined cytochemically. The results of these analyses showed that ADH activity increased significantly as a result of spaceflight exposure. Enzyme activity increased 248% to 304% in dwarf wheat when compared with the ground controls and Brassica showed increases between 334% and 579% when compared with day zero controls. Cytochemical staining revealed no differences in ADH tissue localization in any of the dwarf wheat treatments. These results show the importance of considering root system oxygenation in designing and building nutrient delivery hardware for spaceflight plant cultivation and confirm previous reports of an ADH response associated with spaceflight exposure.

Separation of mouse testis cells on a Celsep (TM) apparatus and their usefulness as a source of high molecular weight DNA or RNA

The use of a self-contained unit-gravity cell separation apparatus for separation of populations of mouse testicular cells is described. The apparatus, a Celsep (TM), maximizes the unit area over which sedimentation occurs, reduces the amount of separation medium employed, and is quite reproducible. Cells thus isolated have been good sources for isolation of DNA, and notably, high molecular weight RNA.

Ho:Tm:Er:LuAG and Two Wavelength Oscillation

Ho:Tm:Er:LuAG demonstrated simultaneous laser oscillation associated with two different transitions and different atoms. Two wavelength oscillation, or TWO, can be achieved in a single laser and single resonator. Moreover, the wavelength can be electronically controlled by varying the pump pulse length, Either the short wavelength, the Ho (5)1(sub 7) to (5)1(sub 8) transition at 2.1 microns, or the long wavelength, the Er (4)1(sub 11/2) to (4)1(sub 13/2) transition at 2.7 microns, or both wavelengths simultaneously can be produced without resorting to mechanical intervention. Consequently, a single laser system can easily switch between producing either of these useful wavelengths. Laser operation of both transitions has been characterized and is presented.

Simultaneous measurement of multiple radiation-induced protein expression profiles using the Luminex(TM) system

Space flight results in the exposure of astronauts to a mixed field of radiation composed of energetic particles of varying energies, and biological indicators of space radiation exposure provides a better understanding of the associated long-term health risks. Current methods of biodosimetry have employed the use of cytogenetic analysis for biodosimetry, and more recently the advent of technological progression has led to advanced research in the use of genomic and proteomic expression profiling to simultaneously assess biomarkers of radiation exposure. We describe here the technical advantages of the Luminex(TM) 100 system relative to traditional methods and its potential as a tool to simultaneously profile multiple proteins induced by ionizing radiation. The development of such a bioassay would provide more relevant post-translational dynamics of stress response and will impart important implications in the advancement of space and other radiation contact monitoring. c2004 COSPAR. Conductively Cooled Ho:Tm:LuLiF Laser Amplifier

A conductively-cooled Ho:Tm:LuLiF laser head can amplify 80mJ/340ns probe pulses into 400mJ when the pump pulse energy is close to amplified spontaneous emission (ASE) threshold, 5.6J. For a small signal, the double-pass amplification exceeds 25.

Summary of Current Radiometric Calibration Coefficients for Landsat MSS, TM, ETM+, and EO-1 ALI Sensors

This paper provides a summary of the current equations and rescaling factors for converting calibrated Digital Numbers (DNs) to absolute units of at-sensor spectral radiance, Top-Of- Atmosphere (TOA) reflectance, and at-sensor brightness temperature. It tabulates the necessary constants for the Multispectral Scanner (MSS), Thematic Mapper (TM), Enhanced Thematic Mapper Plus (ETM+), and Advanced Land Imager (ALI) sensors. These conversions provide a basis for standardized comparison of data in a single scene or between images acquired on different dates or by different sensors. This paper forms a needed guide for Landsat data users who now have access to the entire Landsat archive at no cost.

20 years of Tm:Ho:YLF and LuLF Laser Development for Global Winds Measurements

NASA Langley Research Center has a long history of developing 2 micron lasers. From fundamental spectroscopy research, theoretical prediction of new materials, laser demonstration and engineering of lidar systems, it has been a very successful program spanning around two decades. This article covers the program development from the early research to the present instrumentation. A brief historical perspective of Tm:Ho work by early researchers is also given.
