The treatment of insulin-dependent diabetes mellitus is characterized by artificial supplementation of pancreatic β-cell ability to regulate sugar levels in the blood. Even though various insulin analogs are crucial for reasonable glycemic control, understanding the dynamic mechanism of the insulin analogs may help to improve the best-protracted insulin analog to assist people with type 1 diabetes (T1D) to live comfortably while maintaining tight glycemic control. Here, we present the high-resolution crystal structure of NN304, known as insulin detemir, to 1.7 Å resolution at cryogenic temperature. We computationally further investigated our crystal structure’s monomeric-dimeric conformation and dynamic profile by comparing it with a previously available detemir structure (PDB ID: 1XDA). Our structure (PDB ID: 8HGZ), obtained at elevated pH, provides electrostatically triggered minor movements in the equilibrium between alternate conformational substates compared to the previous structure, suggesting it might induce an intermediate state in the dissociation pathway of the insulin detemir’s hexamer:dihexamer equilibrium. Supplemented with orientational cross-correlation analysis by a Gaussian network model (GNM), this alternate oligomeric conformation offers the distinct cooperative motions originated by loose coupling of distant conformational substates of a protracted insulin analog that has not been previously observed.
Switchgrass (Panicum virgatum L.) is an important cellulosic biofuel grass native to North America. Rust, caused by Puccinia spp. is the most predominant disease of switchgrass and has the potential to impact biomass conversion. In this study, virulence patterns were determined on a set of 38 switchgrass genotypes for 14 single-spore rust isolates from 14 field samples collected in seven states. Single nucleotide polymorphism (SNP) variation was also assessed in 720 sequenced cloned amplicons representing 654 base pairs of the elongation factor 1-α gene from the field samples. Five major haplotypes were identified differing by 11 out of the 39 SNP positions identified. STRUCTURE, Principal Coordinate Analysis, and phylogenetic analyses divided the rust population into two genetic clusters. Virginia and Georgia had the highest and lowest rust genetic diversity, respectively. Only nine accessions showed a differential disease response between the 14 isolates, allowing the identification of eight races, differing by 1–3 virulence factors. Overall, the results suggested clonal reproduction of the pathogen and a North–South differentiation via local adaptation. However, similar haplotypes and races were also recovered from several states, suggesting migration events, and highlighting the need to further investigate the switchgrass rust population structure and evolution in the USA.
The effective radiative forcing of anthropogenic aerosols (ERF aer ) is an important measure of the anthropogenic aerosol effects simulated by a global climate model. Here we analyze ERF aer simulated by the E3SMv1 atmosphere model using both century-long free-running atmosphere-land simulations and short nudged simulations. We relate the simulated ERF aer to characteristics of the aerosol composition and optical properties, and evaluate the relationships between key aerosol and cloud properties. In terms of historical changes from the year 1870 to 2014, our results show that the global mean anthropogenic aerosol burden and optical depth increase during the simulation period as expected, but the regional averages show large differences in the temporal evolution. The largest regional differences are found in the emission-induced evolution of the burden and optical depth of the sulfate aerosol: a strong decreasing trend is seen in the Northern Hemisphere high-latitude region after around 1970, while a continued increase is simulated in the tropics. Consequently, although the global mean anthropogenic aerosol burden and optical depth increase from 1870 to 2014, the ERF aer magnitude does not increase after around year 1970. The relationships between key aerosol and cloud properties (relative changes between preindustrial and present-day conditions) also show evident changes after 1970, diverging from the linear relationships exhibited for the period from 1870 to 2014. The ERF aer in E3SMv1 is relatively large compared to the recently published multi-model estimates; the primary reason is the large indirect aerosol effect (i.e., through aerosol-cloud interactions). Compared to other models, E3SMv1 features a stronger sensitivity of the cloud droplet effective radius to changes in the cloud droplet number concentration. Large sensitivity is also seen in the liquid cloud optical depth, which is determined by changes in both the effective radius and liquid water path. Aerosol-induced changes in liquid and ice cloud properties in E3SMv1 are found to have a strong correlation, as the evolution of anthropogenic sulfate aerosols affects both the liquid cloud formation and the homogeneous ice nucleation in cirrus clouds. The ERF aer estimates in E3SMv1 for the shortwave and longwave components are sensitive to the parameterization changes in both liquid and ice cloud processes. When the parameterization of ice cloud processes is modified, the top-of-atmosphere forcing changes in the shortwave and longwave components largely offset each other, so the net effect is negligible. This suggests that, to reduce the magnitude of the net ERF aer , it would be more effective to reduce the anthropogenic aerosol effect through liquid or mixed-phase clouds.

The ERFaer estimates in E3SMv1 for the shortwave and longwave components are sensitive to the parameterization changes in both liquid and ice cloud processes. When the parameterization of ice cloud processes is modified, the top-of-model forcing changes in the shortwave and longwave components largely offset each other, so the net effect is negligible. This suggests that, to reduce the magnitude of the net ERFaer, it would be more effective to reduce the anthropogenic aerosol effect through liquid or mixed-phase clouds.

Feedback systems stability treated by functional analysis, describing procedures for generating instability criteria
A preliminary quantitative analysis was conducted of four frames exposed by the Apollo 15 metric camera. Two of the frames show the zero-phase surge on maria and the other two show the surge on terrae. It was found that the curves resulting from brightness plotted as a function of phase angle, differ markedly according to which terrain type is involved. The magnitude of the heiligenschein was found to be significantly greater for the maria than for the terrae.
To illustrate that a relativistic field theory need not be manifestly covariant, Lorentz-invariant Lagrangian densities are constructed that yield the equation satisfied by an interacting (two-component) Sakata-Taketani spin-0 field. Six types of external field couplings are considered, two scalars, two vectors, an antisymmetric second-rank tensor, and a symmetric second-rank tensor, with the results specialized to electromagnetic interactions. For either of the two second-rank couplings, the equation is found to describe noncausal wave propagation, a property that is apparent from the dependence of the coefficients of the space derivatives on the external field; in contrast, the noncausality of the corresponding manifestly covariant Duffin-Kemmer-Petiau spin-0 equation is not so obvious. The possibilities for generalizing the results to higher spin theories involving only the essential 2(2J + 1) components for a particle with a definite spin J and mass m are discussed in considerable detail.
