Towards time-resolved MicroED grid preparation using mix-and-inject gas dynamic virtual nozzles

Recent progress in gas dynamic virtual nozzle (GDVN) technologies in combination with high-brilliance synchrotron and X-ray free-electron lasers (XFELs) has allowed the visualization of protein dynamics in crystallo by mixing macromolecular protein crystals with a substrate using tunable mixing times on the order of milliseconds to seconds prior to serial X-ray diffraction data collection. This has become the method of choice for high-resolution structure determination of intermediate states. However, such experiments require large counts of crystals of proper sizes for high-resolution data collection, and premium beam times for screening efforts. Cryogenic microcrystal electron diffraction (MicroED) represents a complementary technique that may be a more accessible avenue for time-resolved nanocrystallography compared with serial X-ray diffraction experiments. MicroED can produce full diffraction datasets from just a few submicrometre-thick crystals, and the approach is more readily accessible, requiring standard cryogenic transmission electron microscopy (TEM) equipment available at many universities and institutes. Cryogenic MicroED, like other forms of cryo-EM, begins with rapidly freezing biological material on electron microscopy grids. In the case of MicroED, micro- to nano-crystals (<500 nm thick) are deposited onto electron microscopy grids and plunge-frozen for subsequent electron diffraction data collection. Here, we have incorporated GDVN technology developed originally for XFEL experiments into the freezing process as a first step towards time-resolved studies. We describe the limited deposition efficiency of the model MicroED protein proteinase K on TEM grids using GDVNs, preceding sample vitrification and successful MicroED data collection. We discuss both the initial results from such experiments and the methodological challenges in developing this approach into a reliable workflow for millisecond-to-second time-resolved structural studies of macromolecules. Our results promise a strategy to deposit crystals on grids using GDVNs and determine high-resolution structures by MicroED, constituting a first step towards development of time-resolved MicroED experiments.

Robust Model Predictive Control for Attack Mitigation of Virtual Synchronous Generators (VSGs) in an Islanded Microgrid

With the increasing application of power converters in distributed energy systems, power electronics converters face an increasing number of cyber-attacks. To mitigate the cyber-attack impact on voltage source converters (VSCs) and microgrids, a robust model predictive control (RMPC) is proposed for virtual synchronous generators (VSG) in an islanded microgrid. This RMPC is designed to compensate for the frequency deviations, thus maintaining frequency stability of the whole islanded microgrid against cyber threats. Finally, to verify the feasibility of the proposed method, comparisons between the proposed method and the conventional VSC controller are provided in different attack scenarios.

Virtual Power Plants: Pilots, Challenges, and Innovations Shaping Future Development

Virtual Power Plants (VPPs) aggregate distributed energy resources (DERs) to provide grid services traditionally delivered by centralized power plants. This article reviews the current state of VPP deployment, highlighting business models, compensation mechanisms, and global pilot projects. While VPPs offer benefits such as grid flexibility, resilience, and cost savings, challenges remain in communication infrastructure, regulatory frameworks, market access, and customer engagement. To address these, we propose a scalable, privacy-preserving hierarchical VPP architecture that coordinates with distribution utilities and preserves customer data. We also present the Integrated T&D Control Room of the Future as a key test bed for validating and accelerating VPP adoption. These innovations can help transition VPPs from pilot programs to integral components of a modern, reliable power grid.

A Virtual Space Vector Model Predictive Control for a Seven-Level Hybrid Multilevel Converter

This paper proposes a virtual space vector (VSV) model predictive control (MPC) for a three-phase seven-level (7L) hybrid multilevel converter (HMC), where each phase consists of an active-neutral-point-clamped converter with a floating H-bridge. To achieve the best current tracking, which is the primary goal of the proposed algorithm, a novel geometrical positioning approach is proposed to select the optimal voltage vector among all the realspace vectors and VSVs. Then, all the possible switching sequences that belong to the optimal voltage vector are evaluated to realize the dc capacitor voltage balancing and common-mode voltage reduction. Through an external modulator, the optimal voltage vector can be synthesized by using either one-, three-, or seven-segment switching sequence. Compared with the conventional MPC, the proposed VSV-MPC can reduce not only the computational burden but also the current THD. Both simulation and experimental results obtained on silicon carbide based 7L-HMC prototype are presented to validate the feasibility and effectiveness of the proposed VSVMPC strategy.

Enhanced Synchronization Stability of Grid-Forming Inverters With Passivity-Based Virtual Oscillator Control

In this article, a passivity-based virtual oscillator control strategy with enhanced synchronization stability for grid-forming inverters (GFMs) is proposed. By adopting the port-controlled Hamiltonian system theory for orbital stabilization problems, an energy pumping-and-damping block is proposed to render GFMs globally asymptotically stable with respect to the prespecified solutions of the power-flow equations from any initial condition. This allows for stable integrations of GFMs to any other globally asymptotically stable systems without their explicit knowledge, e.g., helping maintain synchronism with the bulk power system in a wide range of short-circuit-ratio conditions or under large disturbances and keeping synchronism among multiple GFMs in power systems. Herein, both simulations and experiments are presented to demonstrate the proposed control approach.

Virtual Critical Coupling in High-Power Resonant Systems

Power reflections are a fundamental challenge in high-power resonant systems, causing energy loss, limiting per- formance, and potentially damaging critical components. This problem is particularly acute in applications such as lower hybrid current drive (LHCD) for tokamak-based nuclear fusion, where efficient, stable power transfer to the plasma is paramount. In this work, we introduce and experimentally demonstrate the virtual critical coupling (VCC) mechanism, a nonmechanical method to achieve reflection-free excitation in high-power S-band resonators. VCC utilizes a temporally shaped excitation signal with a precise complex frequency, tailored to the resonator’s characteristics to enable complete energy absorption. Using a custom low-level RF system and a 5-MW S-band klystron, we conducted experiments from low power (32 mW) to high power (600 kW). Furthermore, the results demonstrate a greater than ninefold reduction in the reflection coefficient compared to conventional monochromatic excitation. This signal processing-based approach offers a robust path toward enhancing the efficiency and stability of high-power resonant systems, with significant implications for particle accelerators and the advancement of nuclear fusion energy.

Nonlinear Virtual Inertia Control of WTGs for Enhancing Primary Frequency Response and Suppressing Drivetrain Torsional Oscillations

Virtual inertia controllers (VICs) for wind turbine generators (WTGs) have been recently developed to compensate the reduction of inertia in power systems. However, VICs can induce drivetrain torsional oscillations of WTGs. This paper addresses this issue and develops a novel nonlinear VIC based on objective holographic feedback theory and the definition of a completely controllable system of Brunovsky type. Simulation results under various scenarios demonstrate that the proposed technique outperforms existing VICs in terms of enhancement of system frequency nadir, suppression of WTG drivetrain torsional oscillations, fast and smooth recovery of WTG rotor speed to the original maximum power point (MPP) before the disturbance as well as preventing secondary frequency dip caused by traditional VIC. The proposed technique is also able to adaptively coordinate multiple WTGs to enhance the frequency support and the dynamic performance of each WTG.

Filling gaps in our understanding of belowground plant traits across the world: an introduction to a Virtual Issue

The belowground world is one of the final frontiers in terrestrial ecology. The tangling of plant roots with the surrounding soil below is a lifeline for the humble forbs and towering trees above, and roots play a key role in shaping ecosystem carbon, water and nutrient cycling (Bardgett et al., 2014). Ecologists have long sought to better understand the ecosystem-scale consequences of differing plant strategies, above- and belowground, by relating plant characteristics, or traits, to plant function (Grime, 1977; Pregitzer, 2002). While developing trait–function linkages is arguably more difficult for plant traits that are hidden belowground, root and rhizosphere ecologists continue to fan out across grasslands and forests with their shovels, isotopes, and specialized cameras, seeking a better understanding of the secret lives of roots. Over the years, New Phytologist has served as a virtual town square for scientists to discuss their hard-won observations on the interplay among belowground plant traits, microbial activity, and edaphic and environmental conditions from biomes around the world (Norby & Jackson, 2000; Pregitzer, 2002; Matamala & Stover, 2013; Norby & Iversen, 2017). In this Editorial we highlight the newest papers that update and add to our understanding of the role of root and rhizosphere traits in broader ecosystem processes. We focused on papers published in New Phytologist between 1 January 2019 and 31 December 2020 and extended this window to papers still in ‘early view’ up to the time of writing. Because of the overwhelming number of papers, we did not include those with a decidedly genomic focus or that served primarily as data syntheses, reviews, insights or commentaries.

High-throughput virtual laboratory for drug discovery using massive datasets

Time-to-solution for structure-based screening of massive chemical databases for COVID-19 drug discovery has been decreased by an order of magnitude, and a virtual laboratory has been deployed at scale on up to 27,612 GPUs on the Summit supercomputer, allowing an average molecular docking of 19,028 compounds per second. Over one billion compounds were docked to two SARS-CoV-2 protein structures with full optimization of ligand position and 20 poses per docking, each in under 24 hours. GPU acceleration and high-throughput optimizations of the docking program produced 350× mean speedup over the CPU version (50× speedup per node). GPU acceleration of both feature calculation for machine-learning based scoring and distributed database queries reduced processing of the 2.4 TB output by orders of magnitude. The resulting 50× speedup for the full pipeline reduces an initial 43 day runtime to 21 hours per protein for providing high-scoring compounds to experimental collaborators for validation assays.

Virtual Engineering: Python framework for engineering process design

Virtual Engineering (VE) is a Python software framework designed to accelerate the research and development of engineering processes that are fundamentally defined by multiple unit operations executed in series. VE supports a wide variety of different multi-physics models and integrates them to simulate a complete end-to-end process. To automate the execution of this model sequence, VE provides (i) a robust method to communicate between models, (ii) a high-level, user-friendly interface to set model parameters and enable optimization, and (iii) an overall model-agnostic approach that allows new computational units to be swapped in and out of workflows. Although the VE framework was developed to support the biochemical conversion of biomass to fuel, we have designed each component to easily accommodate new domains and unit models.

A Virtual Element Method for the Full MHD system

In this manuscript we present a novel discretization for the incompressible MHD system. Our approach follows the framework of the Virtual Element Method and offers two main advantages. The method can be implemented in unstructured meshes making it highly versatile and capable of handling a wide array of problems involving interfaces, free-boundaries or adaptive refinements on the mesh. The second advantage involves the divergence of the magnetic field, our approach guarantees that it remains solenoidal. We include a theoretical proof of the condition on the magnetic field as well as energy estimates and a well-posedness study. The latter sheds light as to the stability properties of the method.

Virtual Power Plants and Energy Justice

The Federal Energy Regulatory Commission's (FERC) Order 2222, issued in September 2020, removes barriers for distributed energy resources (DERs) to participate in wholesale energy markets by allowing DERs to aggregate and participate in wholesale markets as a single entity, known as a virtual power plant (VPP). VPPs can provide balancing, reliability, and resiliency grid services and can participate in capacity, energy and ancillary services markets. They can also increase customer energy access and lower electricity bills. This report focuses on VPP business models, including considerations of energy justice (EJ). Through an analysis of the VPP value chain, business models, programs, and pilots, several VPP applications are identified and grouped by their ability to have quantitatively measurable or monetized benefits. Benefits and barriers specific to underserved communities are outlined, and VPP programs with an intentional focus on underserved communities are compared to those without such a focus.

Virtual Resource Management Framework (CRADA Final Report)

There is a need for advanced and widespread business automation within the nuclear industry to drive down operational costs while sustaining or improving safe operations. While there are many business process automation platforms commercially available, the difficulty is that business processes typically rely on a mixture of resource types to accomplish the desired activities and there is no universal software framework virtualizing diverse resource types for the purpose of automation. In this context, we are referring to any capability, physical or intangible, that can be used by an organization to achieve its objectives as a resource. To deploy business automation broadly and enable integrated operations for nuclear (ION), a framework is needed to represent all resource types and their associated disparate data within a plant and to enable seamless flow of resource information to the technologies used for process automation and resource optimization.

Virtual Power Plant Architecture and Resilient Design

Virtual Power Plants (VPPs) represent a fundamental shift in electric grid operations, aggregating distributed energy resources (DERs) such as solar panels and battery storage to deliver utility-scale grid services traditionally provided by centralized power plants. This report examines the unique architectural, operational, and digital assurance considerations that distinguish VPPs from conventional utility infrastructure as they scale from pilot projects to mainstream deployment across the United States. While VPPs offer significant opportunities for grid modernization and enhanced flexibility, their distributed, multi-stakeholder architecture introduces distinct security challenges that differ fundamentally from traditional generation facilities. The analysis identifies risks in VPP operations, including device-level security gaps, platform vulnerabilities, and communication protocol weaknesses that create expanded attack surfaces compared to centralized power plants. Through examination of real-world incidents and emerging threat patterns, the report demonstrates how some VPPs' reliance on consumer-owned devices, public internet infrastructure, and complex vendor ecosystems require new approaches to digital assurance and operational security. The findings provide practical guidance for utilities, regulators, and aggregators to implement robust security frameworks and operational best practices essential for maintaining grid reliability as VPP deployment accelerates under the Federal Energy Regulatory Commission (FERC) Order 2222 and related regulatory initiatives.

Drugsniffer: An Open Source Workflow for Virtually Screening Billions of Molecules for Binding Affinity to Protein Targets

The SARS-CoV2 pandemic has highlighted the importance of efficient and effective methods for identification of therapeutic drugs, and in particular has laid bare the need for methods that allow exploration of the full diversity of synthesizable small molecules. While classical high-throughput screening methods may consider up to millions of molecules, virtual screening methods hold the promise of enabling appraisal of billions of candidate molecules, thus expanding the search space while concurrently reducing costs and speeding discovery. Here, we describe a new screening pipeline, called drugsniffer, that is capable of rapidly exploring drug candidates from a library of billions of molecules, and is designed to support distributed computation on cluster and cloud resources. As an example of performance, our pipeline required ~40,000 total compute hours to screen for potential drugs targeting three SARS-CoV2 proteins among a library of ~3.7 billion candidate molecules.
