Rational Design of Lanmodulin Variants for Size-Based Selectivity of Individual Rare Earth Elements

Rare earth elements (REEs) are essential to modern technologies, yet their high physical and chemical similarity makes separation of individual REEs difficult and environmentally taxing. Metalloproteins offer a promising alternative for selective REE binding, as they tend to have high metal ion affinity and specificity. Lanmodulin (LanM), in particular, has arisen as a potential candidate for REE separation as it exhibits picomolar affinity for elements in the REE family. Prior work has shown that the single point mutation D9N can shift LanM’s preference away from lanthanides toward actinides, motivating efforts to tune selectivity of LanM through targeted mutagenesis. Here, we tested the hypothesis that introducing selective aspartic acid to glutamic acid substitutions in the metal coordinating EF hands of LanM would impose steric constraints that would drive LanM affinity away from larger ions, such as La3+, to smaller ions, such as Y3+. To test this hypothesis, a combination of computational and experimental approaches were employed to evaluate the signal mutations LanM D5E and LanM D3E and the double mutants LanM D1ED5E and LanM D3ED9E. Surprisingly, increasing the number of mutations within the metal center did not enhance affinity for smaller REEs, or decrease affinity for larger ions. Only the single point mutation LanM D5E weakened La3+ binding by one order of magnitude relative to LanM wild type (WT), and pairing it with a second mutation to produce LanM D1ED5E drove La3+ affinity to be stronger than that seen for LanM WT. The D3E mutation alone prevented proper expression and folding, but paring it with D9E to produce LanM D3ED9E rescued expression and yielded La3+ affinities comparable to LanM WT. All variants that expressed (LanM D5E, LanM D1ED5E, LanM D3ED9E) displayed Y3+ affinities comparable to LanM WT. Overall, these results highlight the tunability of LanM’s metal-binding environment but also expose current limitations in predicting structural responses to point mutations within a protein sequence. This work establishes a foundation that can be used for refining computational and experimental strategies to engineer metalloproteins with tailored REE selectivity.

Demand-Side Grid (dsgrid) TEMPO Light-Duty Vehicle Charging Profiles v2022

Simulated hourly electric vehicle charging profiles for light-duty household passenger vehicles in the contiguous United States, 2018-2050. Profiles are differentiated by scenario, county, household and vehicle types, and charging type. Data was produced in 2022 using the Transportation Energy & Mobility Pathway Options (TEMPO) model and published in demand-side grid (dsgrid) toolkit format. Data are available for three adoption scenarios: "AEO Reference Case", which is aligned with the U.S. EIA Annual Energy Outlook 2018 (linked below), "EFS High Electrification", which is aligned with the High Electrification scenario of the Electrification Futures Study (linked below), and "All EV Sales by 2035", which assumes that average passenger light-duty EV sales reach 50% in 2030 and 100% in 2035. The charging shapes are derived from two key assumptions of which data users should be aware: "ubiquitous charger access", meaning that drivers of vehicles are assumed to have access to a charger whenever a trip is not in progress, and "immediate charging", meaning that immediately after trip completion, vehicles are plugged in and charge until they are either fully recharged or taken on another trip. These assumptions result in a bounding case in which vehicles' state of charge is maximized at all times. This bounding case would minimize range anxiety, but is unrealistic from the point of view of both electric vehicle service equipment (EVSE) (i.e., charger) access, and plug-in behavior as it can result in dozens of charging sessions per week for battery electric vehicles (BEVs) that in reality are often only plugged in a few times per week.

Plasmonic Core–Shell–Satellites with Abundant Electromagnetic Hotspots for Highly Sensitive and Reproducible SERS Detection

In this work, we develop a Ag@Al2O3@Ag plasmonic core–shell–satellite (PCSS) to achieve highly sensitive and reproducible surface-enhanced Raman spectroscopy (SERS) detection of probe molecules. To fabricate PCSS nanostructures, we employ a simple hierarchical dewetting process of Ag films coupled with an atomic layer deposition (ALD) method for the Al2O3 shell. Compared to bare Ag nanoparticles, several advantages of fabricating PCSS nanostructures are discovered, including high surface roughness, high density of nanogaps between Ag core and Ag satellites, and nanogaps between adjacent Ag satellites. Finite-difference time-domain (FDTD) simulations of the PCSS nanostructure confirm an enhancement in the electromagnetic field intensity (hotspots) in the nanogap between the Ag core and the satellite generated by the Al2O3 shell, due to the strong core–satellite plasmonic coupling. The as-prepared PCSS-based SERS substrate demonstrates an enhancement factor (EF) of 1.7 × 107 and relative standard deviation (RSD) of ~7%, endowing our SERS platform with highly sensitive and reproducible detection of R6G molecules. We think that this method provides a simple approach for the fabrication of PCSS by a solid-state technique and a basis for developing a highly SERS-active substrate for practical applications.

Disruption of Z-Disc Function Promotes Mechanical Dysfunction in Human Myocardium: Evidence for a Dual Myofilament Modulatory Role by Alpha-Actinin 2

The ACTN2 gene encodes α-actinin 2, located in the Z-disc of the sarcomeres in striated muscle. In this study, we sought to investigate the effects of an ACTN2 missense variant of unknown significance (p.A868T) on cardiac muscle structure and function. Left ventricular free wall samples were obtained at the time of cardiac transplantation from a heart failure patient with the ACTN2 A868T heterozygous variant. This variant is in the EF 3–4 domain known to interact with titin and α-actinin. At the ultrastructural level, ACTN2 A868T cardiac samples presented small structural changes in cardiomyocytes when compared to healthy donor samples. However, contractile mechanics of permeabilized ACTN2 A868T variant cardiac tissue displayed higher myofilament Ca 2+ sensitivity of isometric force, reduced sinusoidal stiffness, and faster rates of tension redevelopment at all Ca 2+ levels. Small-angle X-ray diffraction indicated increased separation between thick and thin filaments, possibly contributing to changes in muscle kinetics. Molecular dynamics simulations indicated that while the mutation does not significantly impact the structure of α-actinin on its own, it likely alters the conformation associated with titin binding. Our results can be explained by two Z-disc mediated communication pathways: one pathway that involves α-actinin’s interaction with actin, affecting thin filament regulation, and the other pathway that involves α-actinin’s interaction with titin, affecting thick filament activation. This work establishes the role of α-actinin 2 in modulating cross-bridge kinetics and force development in the human myocardium as well as how it can be involved in the development of cardiac disease.

MoS 2 Nanoplatelets on Hybrid Core-Shell (HyCoS) AuPd NPs for Hybrid SERS Platform for Detection of R6G

In this work, a novel hybrid SERS platform incorporating hybrid core-shell (HyCoS) AuPd nanoparticles (NPs) and MoS 2 nanoplatelets has been successfully demonstrated for strong surface-enhanced Raman spectroscopy (SERS) enhancement of Rhodamine 6G (R6G). A significantly improved SERS signal of R6G is observed on the hybrid SERS platform by adapting both electromagnetic mechanism (EM) and chemical mechanism (CM) in a single platform. The EM enhancement originates from the unique plasmonic HyCoS AuPd NP template fabricated by the modified droplet epitaxy, which exhibits strong plasmon excitation of hotspots at the nanogaps of metallic NPs and abundant generation of electric fields by localized surface plasmon resonance (LSPR). Superior LSPR results from the coupling of distinctive AuPd core-shell NP and high-density background Au NPs. The CM enhancement is associated with the charge transfer from the MoS 2 nanoplatelets to the R6G. The direct contact via mixing approach with optimal mixing ratio can effectively facilitate the charges transfer to the HOMO and LUMO of R6G, leading to the orders of Raman signal amplification. The enhancement factor (EF) for the proposed hybrid platform reaches ~10 10 for R6G on the hybrid SERS platform.

Equation-Free Coarse Control of Distributed Parameter Systems via Local Neural Operators

The control of high-dimensional distributed parameter systems (DPS) remains a challenge when explicit coarse-grained equations are unavailable. Classical equation-free (EF) approaches rely on fine-scale simulators treated as black-box timesteppers. However, repeated simulations for steady-state computation, linearization, and control design are often computationally prohibitive, or the microscopic timestepper may not even be available, leaving us with data as the only resource. We propose a data-driven alternative that uses local neural operators, trained on spatiotemporal microscopic/mesoscopic data, to obtain efficient short-time solution operators. These surrogates are employed within Krylov subspace methods to compute coarse steady and unsteady-states, while also providing Jacobian information in a matrix-free manner. Krylov-Arnoldi iterations then approximate the dominant eigenspectrum, yielding reduced models that capture the open-loop slow dynamics without explicit Jacobian assembly. Both discrete-time Linear Quadratic Regulator (dLQR) and pole-placement (PP) controllers are based on this reduced system and lifted back to the full nonlinear dynamics, thereby closing the feedback loop.

Structural insights into mRNA reading frame regulation by tRNA modification and slippery codon–anticodon pairing

Modifications in the tRNA anticodon loop, adjacent to the three-nucleotide anticodon, influence translation fidelity by stabilizing the tRNA to allow for accurate reading of the mRNA genetic code. One example is the N1-methylguanosine modification at guanine nucleotide 37 (m 1 G37) located in the anticodon loop andimmediately adjacent to the anticodon nucleotides 34, 35, 36. The absence of m 1 G37 in tRNA Pro causes +1 frameshifting on polynucleotide, slippery codons. Here, we report structures of the bacterial ribosome containing tRNA Pro bound to either cognate or slippery codons to determine how the m 1 G37 modification prevents mRNA frameshifting. The structures reveal that certain codon–anticodon contexts and the lack of m 1 G37 destabilize interactions of tRNA Pro with the P site of the ribosome, causing large conformational changes typically only seen during EF-G-mediated translocation of the mRNA-tRNA pairs. These studies provide molecular insights into how m 1 G37 stabilizes the interactions of tRNA Pro with the ribosome in the context of a slippery mRNA codon.

Thermal performance evaluation of Solar Energy Products Company (SEPCO) 'Soloron' collector tested outdoors

The test article, Model EF-212, Serial Nr. 002, is a single glazed collector with a nonselective absorber plate, using flowing air as the heat transfer medium. The absorber plate and box frame are aluminum and the insulation is one inch isocyanurate foam board with thermal conductivity of 0.11 (BTU/sq ft Hr0/ft.) The tests included the following. (1) time constant test, (2) collector efficiency test, (3) collector stagnation test, (4) incident angle modifier test, (5) load test, (6) weathering test, and (7) absorber plate optical properties test. The results of these tests are tabulated, graphed, or otherwise recorded.

Indoor thermal performance evaluation of the SEPCO air collector

The procedures used and the results obtained during the evaluation test program on the Solaron solar air collector, model EF-212, under simulated conditions for comparison with data collected in outdoor tests on the same collector are given. The test article was a single glazed collector with a nonsensitive absorber plate, aluminum box frame, and one inch isocyanurate foam insulation.

Radio emission from AM Herculis - The quiescent component and an outburst

The VLA has been used to search for radio emission from the AM Her-type binaries VV Pup, EF Eri, PG 1550 + 191, CW 1103 + 354, and AN UMa, at 4.9 GHz. A remarkable 10-min outburst was detected from AM Her at 4.9 GHz, which was about 20 times more intense than the quiescent emission and was essentially 100 percent circularly polarized. It is suggested that the quiescent emission of AM Her can be accounted for by 500-keV electrons trapped in the magnetosphere of the white dwarf, provided that the electron energy spectrum is quite hard and that the spectral hardness or number density of energetic electrons increases with radius, while the outburst is probably due to an electron-cyclotron maser operating near the surface of the red dwarf companion. The implied existence of a 1000-gauss localized magnetic field and a corona on the red dwarf has consequences for mass transfer, field line interactions, and variable activity.

Synchronization of magnetic stars in binary systems

Asynchronous rotation of magnetic stars in close binary systems drives substantial field-aligned electrical currents between the magnetic star and its companion. The resulting magnetohydrodynamic torque is able to account for the heretofore unexplained synchronous rotation of the strongly magnetic degenerate dwarf component in systems like AM Her, VV Pup, AN UMa, and EF Eri as well as the magnetic A type component in systems like HD 98088 and 41 Tauri. The electric fields produced by even a small asynchronism are large and may accelerate some electrons to high energies, producing radio emission. The total energy dissipation rate in systems with degenerate dwarf spin periods as short as 1 minute may reach 10 to the 33rd ergs/s. Total luminosities of this order may be a characteristic feature of such systems.

Comparison of the X-ray observations of the AM Herculis objects to theory

The X-ray observations of the original AM Her objects (AM Her, VV Pup, EF Eri, and AN UMa are reexamined. The work differs from that carried out previously in that the observed X-ray spectral properties are compared to theoretical models calculated using a dipolar accretion geometry rather than a spherically symmetric one, and the observed X-ray light curves are compared to theoretical light curves calculated assuming that the X-ray sources are cataclysmic variable-like binaries which contain a strongly magnetic white dwarf. The added constraints offered by the light curve properties restrict the possible kinds of X-ray emission scenarios in some cases. For instance, it is found that the X-ray observations of AM Her can be self-consistently explained by accretion powered radiative shock models. Further, the observations cannot be self-consistently explained by conduction dominated models as has been suggested by previous workers.

Orbital transfer vehicle 3000 LBF thrust chamber assembly hot fire test program

The Aerojet Orbital Transfer Vehicle (OTV) Thrust Chamber Assembly (TCA) concept consists of a hydrogen cooled chamber, and annular injector, and an oxygen cooled centerbody. The hot fire testing of a heat sink version of the chamber with only the throat section using hydrogen cooling is documented. Hydraulic performance of the injector and cooled throat were verified by water flow testing prior to TCA assembly. The cooled throat was proof tested to 3000 psia to verify the integrity of the codeposited EF nickel-cobalt closeout. The first set of hot fire tests were conducted with a heat sink throat to obtain heat flux information. After demonstration of acceptable heat fluxes, the heat sink throat was replaced with the LH2 cooled throat section. Fourteen tests were conducted with a heat sink chamber and throat at chamber pressures of 85 to 359 psia. The injector face was modified at this time to add more face coolant flow. Ten tests were then conducted at chamber pressures of 197 to 620 psia. Actual heat fluxes at the higher chamber pressure range were 23 percent higher than the average of 10 Btu/in 2 predicted.

Automation - Changes in cognitive demands and mental workload

The effect of partial automation on mental workloads in man/machine tasks is investigated experimentally. Subjective workload measures are obtained from six subjects after performance of a task battery comprising two manual (flight-path control, FC, and target acquisition, TA) tasks and one decisionmaking (engine failure, EF) task; the FC task was performed in both a fully manual (altitude and lateral control) mode and in a semiautomated mode (autmatic latitude control). The performance results and subjective evaluations are presented in graphs and characterized in detail. The automation is shown to improve objective performance and lower subjective workload significantly in the combined FC/TA task, but not in the FC task alone or in the FC/EF task.

Research and development of HgZnTe as an infrared material

Interfacial morphology and Fermi level pinning behavior at the interfaces of Al, Ag, and Pt with UHV-cleaved CdTe and ZnTe have been studied using X-ray and ultraviolet photoemission spectroscopies. Results are compared to metal/HgCdTe interface formation, where the weak HgTe bond and consequent ease of Hg loss strongly influence semiconductor disruption and metal-semiconductor intermising. For Al/CdTe, the strong Al-Te reaction yields a significantly more extensive Al-Te reacted region than has been observed for HgCdTe. The Al/ZnTe interface is observed to be more abrupt than Al/CdTe. The final Fermi level pinning positions, Ef-Evbm for Al, Ag, and Pt on p-type CdTe and p-ZnTe have been determined. Efi is found to be roughly the same for both CdTe and ZnTe, with the value for ZnTe lying approximately 0.2 eV closer to the VBM for all three metals. From these results, one would expect Schottky barriers of about the same height for these metals on p-CdTe and p-ZnTe; and also that, in principle, metal interfaces with the two alloys HgCdTe and HgZnTe would have the same properties. Comparisons and implications for electrical behavior of metal contacts to the alloys are discussed.

Millisecond optical photometry of the DQ Herculis objects

The results of millisecond optical photometry of the DQ Herculis stars EX Hya, H2252-035, V1223 Sgr, and AE Aqr obtained at the Las Campanas Observatory and Cerro Tololo Inter-American Observatory during the years 1985, 1986, and 1987 are reported. The data for coherent and incoherent features are searched for. Coherent features (other than those previously known) were not detected for frequencies between 0.1 and 250 Hz at 4-sigma upper limits of 0.05-0.4 percent. Evidence is not found for 1-3 s, quasi-coherent features. Such features have been detected in the optical emission of the AM Her objects AN UMa, EF Eri, and E1405-451. Because of the similarities between AM Her and DQ Her objects, it was suggested that such quasi-coherent features might also be found in the DQ Her objects.

Magnetism and superconductivity of some Tl-Cu oxides

Many copper oxide based Thallium compounds have now been discovered. In comparison to the Bi-compounds, the Tl-system shows a richer diversity; viz., High Temperature Superconductors (HTSC) can be obtained with either one or two Tl-0 layers (m = 1,2); also, the triple-digit phases are easier to synthesize. The value of d, oxygen stoichiometry, is critical to achieving superconductivity. The Tl system is robust to oxygen loss; Tl may be lost or incorporated by diffusion. A diffusion coefficient equal to 10 ms at 900 C was determined. Both ortho-rhombic and tetragonal structures are evidenced, but HTSC behavior is indifferent to the crystal symmetry. This system has the highest T(sub c) confirmed. T(sub c) generally increases with p, the number of CuO layers, but tends to saturate at p = 3. Zero resistance as high as 125K has been observed. Most of these HTSC's are hole type, but the Ce-doped specimens may be electronic. The magnetic aspects were studied; because in addition to defining the perfectly diamagnetic ground state as in the conventional superconductors, magnetism of the copper oxides show a surprising variety. This is true of both the normal and the superconducting states. Also, due to the large phonon contribution to the specific heat at the high T(sub c) accurate thermal measurement of important parameters such as the sp. heat jump, electronic density of states, D(Ef) and coherence length are uncertain, and thus, are estimated from the magnetic results. Results from the Tl-system CuO, LaBaCuO, 120 and the Bi-CuO compounds are discussed. The emphasis is on the role of magnetism in the TlCuO HTSC, but technological aspects are also pointed out.

Bowen fluorescence in AM Herculis stars

The study relates the observations of the Bowen fluorescence lines in AM Her and EF Eri, two AM Her systems. The implications of source variability and the checks performed in order to verify the accuracy of the data are reviewed. The line ratios for H I and He II are shown to deviate considerably from those expected; in this light, the H I and He II emission in AM Her is analyzed in detail. This approach makes it possible to put limits on line and continuum optical depths as well as temperatures. These limits along with the observed Bowen emission are used to construct a picture of the emission-line regions in the two sources, producing the values for the size, density, and temperature of a region. The derived value of the size is found to be significantly less than the size of the Roche lobe of the white dwarf, while the large range of densities is attributed to the exponential temperature dependence in the expression used for density.
