Rapid T cell engineering to counter emerging threats (Full Technical Report)

Instead of genetic engineering, we sought to determine if a rapid method of membrane protein delivery could generate functional CAR-T cells in a rapid, safer, cost-effective manner. Using cell free protein synthesis, we generated CAR proteins embedded in a nanodisc, and thus effectively solubilized the protein. We demonstrated the first functional CAR proteins outside of a cellular context. We next tested uptake into immune cells and found extremely high uptake into T cells and multiple other populations of immune cells. We found some evidence of cytotoxicity in vitro conferred by the nanodisc delivered protein.

Review of Design Elements within Power Infrastructure Cyber–Physical Test Beds as Threat Analysis Environments

Cyber–physical systems (CPSs) are an integral part of modern society; thus, enhancing these systems’ reliability and resilience is paramount. Cyber–physical testbeds (CPTs) are a safe way to test and explore the interplay between the cyber and physical domains and to cost-effectively enhance the reliability and resilience of CPSs. Here a review of CPT elements, broken down into physical components (simulators, emulators, and physical hardware), soft components (communication protocols, network timing protocols), and user interfaces (visualization-dashboard design considerations) is presented. Various methods used to validate CPS performance are reviewed and evaluated for potential applications in CPT performance validation. Last, initial simulated results for a CPT design, based on the IEEE 33 bus system, are presented, along with a brief discussion on how model-based testing and fault–injection-based testing (using scaling and ramp-type attacks) may be used to help validate CPT performance.

Reliable cyber-threat detection in rapidly changing environments

In some embodiments, a plurality of monitoring nodes each generate a series of current monitoring node values over time that represent a current operation of the industrial asset. An attack detection computer platform may receive the series of current monitoring node values and generate a set of current feature vectors including a current feature for capturing transients (e.g., local transients and/or global transients). The attack detection computer platform may also access an attack detection model having at least one decision boundary that was created using at least one of a set of normal feature vectors and/or a set of attacked feature vectors. The attack detection model may then be executed such that an attack alert signal is transmitted by the attack detection computer platform, when appropriate, based on the set of current feature vectors (including the current feature to capture transients) and the at least one decision boundary.
