PASS: A computer program for Preliminary Aircraft Structural Synthesis

A computer code for Preliminary Aircraft Structural Synthesis provides rapid and accurate analysis for aircraft structures that can be adequately modeled by beam finite elements. The philosophy used in developing the program was to provide a basic framework that can be used for structural synthesis. It is anticipated that a user will need to add detail to this framework in order to perform his specific task. With this philosophy in mind, the program was written so that it is easily divided into segments, thereby making it readily adaptable. The theoretical portion of this manual describes the basic structure of the program and details the development of the unique beam element that is used. The present capability of the algorithm is stated and suggestions are made regarding enhancements to this capability. User information is also given that provides an overview of the program's construction, identifies the required inputs, describes the program output, provides some comments on the program use, and exhibits results for a simple example.

Synthesis of stiffened shells of revolution

Computer programs for the synthesis of shells of various configurations were developed. The conditions considered are: (1) uniform shells (mainly cones) using a membrane buckling analysis, (2) completely uniform shells (cones, spheres, toroidal segments) using linear bending prebuckling analysis, and (3) revision of second design process to reduce the number of design variables to about 30 by considering piecewise uniform designs. A perturbation formula was derived and this allows exact derivatives of the general buckling load to be computed with little additional computer time.

Reuse: A knowledge-based approach

This paper describes our research in automating the reuse process through the use of application domain models. Application domain models are explicit formal representations of the application knowledge necessary to understand, specify, and generate application programs. Furthermore, they provide a unified repository for the operational structure, rules, policies, and constraints of a specific application area. In our approach, domain models are expressed in terms of a transaction-based meta-modeling language. This paper has described in detail the creation and maintenance of hierarchical structures. These structures are created through a process that includes reverse engineering of data models with supplementary enhancement from application experts. Source code is also reverse engineered but is not a major source of domain model instantiation at this time. In the second phase of the software synthesis process, program specifications are interactively synthesized from an instantiated domain model. These specifications are currently integrated into a manual programming process but will eventually be used to derive executable code with mechanically assisted transformations. This research is performed within the context of programming-in-the-large types of systems. Although our goals are ambitious, we are implementing the synthesis system in an incremental manner through which we can realize tangible results. The client/server architecture is capable of supporting 16 simultaneous X/Motif users and tens of thousands of attributes and classes. Domain models have been partially synthesized from five different application areas. As additional domain models are synthesized and additional knowledge is gathered, we will inevitably add to and modify our representation. However, our current experience indicates that it will scale and expand to meet our modeling needs.

Strategies for Constructing and Operating DNA Origami Linear Actuators

Abstract Linear actuators are ubiquitous components at all scales of engineering. DNA nanotechnology offers a unique opportunity for bottom‐up assembly at the molecular scale, providing nanoscale precision with multiple methods for constructing and operating devices. In this paper, DNA origami linear actuators with up to 200 nm travel, based on a rail threading a topologically locked slider, are demonstrated. Two strategies, one‐ and two‐pot assembly, are demonstrated whereby the two components are folded from one or two DNA scaffold strands, respectively. In order to control the position of the slider on the rail, the rail and the inside of the slider are decorated with single‐stranded oligonucleotides with distinct sequences. Two positioning strategies, based on diffusion and capture of signaling strands, are used to link the slider reversibly to determined positions on the rail with high yield and precision. These machine components provide a basis for applications in molecular machinery and nanoscale manufacture including programmed chemical synthesis.

Development of a clostridia-based cell-free system for prototyping genetic parts and metabolic pathways

Gas fermentation by autotrophic bacteria, such as clostridia, offers a sustainable path to numerous bioproducts from a range of local, highly abundant, waste and low-cost feedstocks, such as industrial flue gases or syngas generated from biomass or municipal waste. Unfortunately, designing and engineering clostridia remains laborious and slow. The ability to prototype individual genetic part function, gene expression patterns, and biosynthetic pathway performance in vitro before implementing designs in cells could help address these bottlenecks by speeding up design. Unfortunately, a high-yielding cell-free gene expression (CFE) system from clostridia has yet to be developed. Here in this paper, we report the development and optimization of a high-yielding (236 ± 24 μg/mL) batch CFE platform from the industrially relevant anaerobe, Clostridium autoethanogenum. A key feature of the platform is that both circular and linear DNA templates can be applied directly to the CFE reaction to program protein synthesis. We demonstrate the ability to prototype gene expression, and quantitatively map aerobic cell-free metabolism in lysates from this system. We anticipate that the C. autoethanogenum CFE platform will not only expand the protein synthesis toolkit for synthetic biology, but also serve as a platform in expediting the screening and prototyping of gene regulatory elements in non-model, industrially relevant microbes.

VISION: a modular AI assistant for natural human-instrument interaction at scientific user facilities

Scientific user facilities, such as synchrotron beamlines, are equipped with a wide array of hardware and software tools that require a codebase for human-computer-interaction. This often necessitates developers to be involved to establish connection between users/researchers and the complex instrumentation. The advent of generative AI presents an opportunity to bridge this knowledge gap, enabling seamless communication and efficient experimental workflows. Here we present a modular architecture for the Virtual Scientific Companion by assembling multiple AI-enabled cognitive blocks that each scaffolds large language models (LLMs) for a specialized task. With VISION, we performed LLM-based operation on the beamline workstation with low latency and demonstrated the first voice-controlled experiment at an x-ray scattering beamline. The modular and scalable architecture allows for easy adaptation to new instruments and capabilities. Development on natural language-based scientific experimentation is a building block for an impending future where a science exocortex—a synthetic extension to the cognition of scientists—may radically transform scientific practice and discovery.

Development of a scalable, robust electrocatalytic technology for conversion of CO 2 to formic acid via microstructured materials

This project was funded by the Bioenergy Engineering for Product Synthesis (BEEPS) program under the Funding Opportunity Announcement (FOA) DE-FOA-0001916 Topic Area 5 “Rewiring Carbon Utilization”. This FOA sought projects that would electrocatalytically reduce CO 2 to a carbon intermediate and then upconvert to a multi-carbon product or fuel via non-photosynthetic biological system engineering. The project sought to combine the expertise of OCO Chem, whose chief scientist had previously developed an efficient electrocatalytic reactor for conversion of CO 2 to formate with potential to scale, Montana State University investigators who had recently patented a method for laterally grading membranes with the potential of improving reactant distribution and more uniform efficiency across membrane-based reactors, and University of South Florida which had recently discovered a novel C-C bond forming reaction not known to naturally occur in biological systems and which uses C1 substrates and circumvents the central microbial metabolism.

Transonic transport study: Economics

An economic analysis was performed to evaluate the impact of advanced materials, increased aerodynamic and structural efficiencies, and cruise speed on advanced transport aircraft designed for cruise Mach numbers of .90, .98, and 1.15. A detailed weight statement was generated by an aircraft synthesis computer program called TRANSYN-TST; these weights were used to estimate the cost to develop and manufacture a fleet of aircraft of each configuration. The direct and indirect operating costs were estimated for each aircraft, and an average return on investment was calculated for various operating conditions. There was very little difference between the operating economics of the aircraft designed for Mach numbers .90 and .98. The Mach number 1.15 aircraft was economically marginal in comparison but showed significant improvements with the application of carbon/epoxy structural material. However, the Mach .90 and Mach .98 aircraft are the most economically attractive vehicles in the study.

Design of turbofan engine controls using output feedback regulator theory

A multivariable control design procedure based on output feedback regulator (OFR) theory is applied to the F100 turbofan engine. Results for the OFR design are compared to a design based on linear quadratic regulator (LQR) theory. This LQR design was obtained as part of the F100 Multivariable Control Synthesis (MVCS) program. In the MVCS program the LQR feedback control was designed in a reduced dimension state space and then applied to the original system. However, the OFR feedback control is designed in the full order state space and thus eliminates any need for model reduction techniques. Using the performance measure and control structure of the MVCS program LQR design, an equivalent OFR feedback control is obtained. The flexibility of the OFR as a control design procedure is demonstrated and differing feedback control structures are evaluated.
