Engineering a protease-stable, oral single-domain antibody to inhibit IL-23 signaling

Interleukin (IL)-23 is a validated therapeutic target in inflammatory bowel disease. While antibodies targeting IL23 demonstrate clinical efficacy, they face challenges such as high costs, safety risks, and the necessity of parenteral administration. Here, we present a workflow to simultaneously enhance the affinity and protease stability of an inhibitory anti-IL23R VHH for oral use. Cocrystal structure analysis reveals that the anti-IL23R VHH employs both CDR and framework residues to achieve picomolar affinity for IL23R. The engineered VHH remains stable for over 8 h in intestinal fluid and 24 h in fecal samples. Oral administration of this VHH achieves deep pathway inhibition in a murine colitis model. Furthermore, a single pill provides sustained IL23R inhibition in nonhuman primate blood for over 24 h. With high potency, gut stability, high production yield, and favorable drug-like properties, oral VHHs offer a promising approach for inflammatory bowel diseases.

Discovery of GuaB inhibitors with efficacy against Acinetobacter baumannii infection

ABSTRACT Guanine nucleotides are required for growth and viability of cells due to their structural role in DNA and RNA, and their regulatory roles in translation, signal transduction, and cell division. The natural antibiotic mycophenolic acid (MPA) targets the rate-limiting step inde novoguanine nucleotide biosynthesis executed by inosine-5´-monophosphate dehydrogenase (IMPDH). MPA is used clinically as an immunosuppressant, but whetherin vivoinhibition of bacterial IMPDH (GuaB) is a valid antibacterial strategy is controversial. Here, we describe the discovery of extremely potent small molecule GuaB inhibitors (GuaBi) specific to pathogenic bacteria with a low frequency of on-target spontaneous resistance and bactericidal efficacyin vivoagainstAcinetobacter baumanniimouse models of infection. The spectrum of GuaBi activity includes multidrug-resistant pathogens that are a critical priority of new antibiotic development. Co-crystal structures ofA. baumannii, Staphylococcus aureus, andEscherichia coliGuaB proteins bound to inhibitors show comparable binding modes of GuaBi across species and identifies key binding site residues that are predictive of whole-cell activity across both Gram-positive and Gram-negative clades of Bacteria. The clearin vivoefficacy of these small molecule GuaB inhibitors in a model ofA. baumanniiinfection validates GuaB as an essential antibiotic target. IMPORTANCE The emergence of multidrug-resistant bacteria worldwide has renewed interest in discovering antibiotics with novel mechanism of action. For the first time ever, we demonstrate that pharmacological inhibition ofde novoguanine biosynthesis is bactericidal in a mouse model ofAcinetobacter baumanniiinfection. Structural analyses of novel inhibitors explain differences in biochemical and whole-cell activity across bacterial clades and underscore why this discovery may have broad translational impact on treatment of the most recalcitrant bacterial infections.

On how structures convey non-diffusive turbulence spreading

Abstract We report on comprehensive experimental studies of turbulence spreading in edge plasmas. These studies demonstrate the relation of turbulence spreading and entrainment to intermittent convective density fluctuation events or bursts (i.e. blobs and holes). The non-diffusive character of turbulence spreading is thus elucidated. The turbulence spreading velocity (or mean jet velocity) manifests a linear correlation with the skewness of density fluctuations, and increases with the auto-correlation time of density fluctuations. Turbulence spreading by positive density fluctuations is outward, while spreading by negative density fluctuations is inward. The degree of symmetry breaking between outward propagating blobs and inward propagating holes increases with the amplitude of density fluctuations. Thus, blob-hole asymmetry emerges as crucial to turbulence spreading. These results highlight the important role of intermittent convective events in conveying the spreading of turbulence, and constitute a fundamental challenge to existing diffusive models of spreading.

Transport phenomena in stratified multi-fluid flo w in the presence and absence of gravity

An experiment is being conducted to study the effects of buoyancy on planar stratified flows. A wind tunnel has been designed and constructed to generate planar flows with separate heating for the top and bottom planar air jets emerging from slot nozzles separated by an insulating splitter plate. The objective is to generate planar jet flows with well defined and well controlled velocity and temperature profiles. Magnitudes of velocity and temperature will be varied separately in each flow for both laminar and turbulent flow conditions. Both stably and unstably stratified flows will be studied by changing the temperature distributions in each air stream. This paper reports on the design of the apparatus and initial measurements of velocity and turbulence made by laser Doppler velocimetry.
