Hierarchically Informed Engineering Models for Predictive Modeling of Turbulent Premixed Flame Propagation in Pre- chamber Turbulent Jet Ignition

The goal of the project is to improve the predictive accuracy and efficiency of turbulent combustion sub-models for pre-chamber turbulent jet ignition (TJI). This goal is achieved through the development of a hierarchically informed engineering model for turbulent combustion in TJI. The model development starts with the highest level of model description of turbulent combustion with direct numerical simulation (DNS) from which fundamental characteristics and scaling properties of turbulent premixed flame propagation under TJI relevant conditions are obtained.

Multi-regime mixing modeling for local extinction and re-ignition in turbulent non-premixed flame by using LES/FDF method

Local extinction and re-ignition occur in turbulent non-premixed combustion when the Damköhler number is not large enough and the combustion is not fully mixing controlled. The occurrence of local extinction introduces locally extinguished flame holes followed by a premixed flame propagation toward the hole center to potentially reignite the flame. The co-existence of the non-premixed and premixed combustion regimes complicates the modeling since traditional combustion models are mostly for a single regime. In this work, we examine the effect of multi-regime mixing modeling in the transported filtered density function (FDF) method on the predictions of local extinction and reignition. Predictions of local extinction and reignition remain a challenge for the FDF method despite the progress made in the past. To account for the multi-regime combustion, two different mixing timescale models for non-premixed and premixed combustion are combined. A flame index based on the gradients of fuel and oxidizer is used to define a weighting factor to blend the two mixing timescale models. A turbulent jet non-premixed flame with substantial local extinction, the Sydney piloted jet flame L, is adopted as a test case to examine the performance of the multi-regime model in large-eddy simulation/FDF modeling. It is found that the traditional non-premixed mixing timescale model when combined with the modified Curl mixing leads to global extinction for the Sydney flame L without the presence of the premixed combustion regime. After accounting for the multi-regime combustion with proper detection of the different combustion regimes, the predictions for the flame statistics and the amount of local extinction are significantly improved. Furthermore, it suggests the need of including multi-regime combustion for the predictions of local extinction and re-ignition in turbulent non-premixed combustion configurations.

Broad-spectrum CRISPR-mediated inhibition of SARS-CoV-2 variants and endemic coronaviruses in vitro

A major challenge in coronavirus vaccination and treatment is to counteract rapid viral evolution and mutations. Here we demonstrate that CRISPR-Cas13d offers a broad-spectrum antiviral (BSA) to inhibit many SARS-CoV-2 variants and diverse human coronavirus strains with >99% reduction of the viral titer. We show that Cas13d-mediated coronavirus inhibition is dependent on the crRNA cellular spatial colocalization with Cas13d and target viral RNA. Cas13d can significantly enhance the therapeutic effects of diverse small molecule drugs against coronaviruses for prophylaxis or treatment purposes, and the best combination reduced viral titer by over four orders of magnitude. Using lipid nanoparticle-mediated RNA delivery, we demonstrate that the Cas13d system can effectively treat infection from multiple variants of coronavirus, including Omicron SARS-CoV-2, in human primary airway epithelium air-liquid interface (ALI) cultures. Our study establishes CRISPR-Cas13 as a BSA which is highly complementary to existing vaccination and antiviral treatment strategies.

A Right Transfer Access Control Model of Internet of Things Based on Smart Contract

Sensor nodes play a crucial role in the promotion of development of Internet of Things (IoT). Through this transaction, RO defines access control policies in script form based on ABAC's access control model to grant access right. The identity of all users in the model is identified by address. This paper builds a more flexible right transfer access control model by means of combining the Attribute-Based Access Control model (ABAC) and blockchain technology. Owing to the characteristics of ABAC’s attributes and right association, the massive problems of some sensor nodes can be solved. At the same time, for the sake of addressing the dynamic problems such as node access and right transfer, right transfer contract (TS) and access control contract (CS) are employed on the chain to ensure efficient and safe transmission of rights. To solve on-chain storage problems and ensure transparency of the operation, the idea of Rollup in Ethereum expansion is used to upload the final state of protocol policy and right exchange to the chain. Any user can know the policy and current right transfer status at any time. Finally, comparative and security analysis show that the model presented here can solve IoT devices’ massive and dynamic problems more effectively and it is more secure than the traditional models.

Examination of probability distribution of mixture fraction in LES/FDF modelling of a turbulent partially premixed jet flame

An accurate prediction of the probability density function (PDF) of the mixture fraction is crucial to the prediction of combustion since mixing plays an important role in turbulent non-premixed and partially premixed flames. This work provides an assessment of the large-eddy simulation (LES)/filtered density function (FDF) method for the prediction of the PDF of the mixture fraction. The advantage of the LES/FDF method is that it provides the full predictions of the statistical distribution of scalars including the mixture fraction. The predictive accuracy of the method for the PDF is yet to be fully validated. Assessing the prediction of the PDF of the mixture fraction, a conserved scalar, is an important starting point. The Sydney/Sandia inhomogeneous inlet jet flame is used as a test case. A quick comparison shows that the LES/FDF predicted PDF shapes of the mixture fraction deviate significantly from the commonly presumed Beta-PDF as well as from the experimental data in the flame. Here, to examine the source of the discrepancy, we clarify the different PDF definitions used in the comparison among the predictions, measurements, and the presumed shape PDFs. The discrepancy observed from the comparison is largely reconciled by clarifying the difference between the PDFs that are examined. The PDF of the resolved mixture fraction is shown to be close to the Beta-PDF in both the measurements and predictions, while the PDF directly deduced from the LES/FDF particles deviates significantly from the Beta-PDF. A multimodal PDF analysis and a pseudo convergence analysis are conducted to provide plausible evidence to support the predicted multimodal PDF shapes. The sub-filter scale FDF is shown to be close to the Beta-PDF too through the construction of a synthesized PDF, which supports the common presumed Beta-PDF assumption used in the presumed PDF methods when combined with LES.

Machine learning assisted modeling of mixing timescale for LES/PDF of high-Karlovitz turbulent premixed combustion

Accurate modeling of mixing in the transported probability density function (PDF) method remains a great challenge, especially for turbulent premixed combustion under extreme conditions such as high Karlovitz number Ka. Recently, a power-law based mixing timescale model was developed for the large-eddy simulations (LES)/PDF modeling of high-Ka number turbulent premixed flames. It is found in this work that the power-law mixing timescale model is highly sensitive to the model parameters. It is thus critically needed to develop accurate calibration of these model parameters. The empirical specification of the model parameters developed in Zhang et. al. is found to be inadequate for accurate modeling of the mixing timescale. Here, machine learning is introduced as an attractive alternative in this work for the specification of the model parameters. A high-Ka number DNS jet flame is used as the training and validation of the machine learning models. The choices of the input parameters are discussed and compared for the machine learning models. The effect of differential molecular diffusion on mixing is examined by including the effect of the Lewis number in the training of the machine learning models. The performance of different machine learning algorithms is compared for the specification of the mixing model parameters. Overall, excellent performance of the machine learning models is observed for assisting the mixing modeling. The feasibility, interpretability, applicability, generality, and portability of using machine learning are discussed in general to provide a perspective on applying data-driven machine learning for turbulent combustion modeling studies.

Fully consistent Eulerian Monte Carlo fields method for solving probability density function transport equations in turbulence modeling

The probability density function (PDF) transport equation method is a sophisticated model for the closure of turbulent mixing and turbulent reactive flows. An efficient solution approach for solving the PDF transport equation has been vital for the method to be widely used in applications. The Eulerian Monte Carlo fields (EMCF) method has been developed to solve the PDF transport equation efficiently for decades. A recent work by Wang et al. revealed a serious issue of the EMCF method for not being fully consistent with the PDF transport equation for which the method is designed to solve. This work advances the state of the art by introducing fully consistent EMCF methods for solving the PDF transport equation. The fully consistent EMCF formulations are derived for two different PDF equation forms. The consistency of the EMCF formulations is mathematically confirmed by examining the derived moment transport equations from the EMCF formulations and from the PDF transport equation. The method of manufactured solutions is employed to further verify the consistency and convergence of the different EMCF formulations numerically. The newly introduced EMCF formulations bring the EMCF method to full consistency with the PDF transport equations for the first time.

Clinically applicable 53-Gene prognostic assay predicts chemotherapy benefit in gastric cancer: A multicenter study

BACKGROUND:We previously established a 53-gene prognostic signature for overall survival (OS) of gastric cancer patients. This retrospective multi-center study aimed to develop a clinically applicable gene expression detection assay and to investigate the prognostic value of this signature. METHODS:A TCGA gastric adenocarcinoma cohort (TCGA-STAD) was used for comparing 53-gene signature with other gene signatures. A high-throughput mRNA hybridization gene expression assay was developed to quantify the expression of 53-genes in formalin-fixed paraffin-embedded tissues of 540 patients enrolled from three hospitals. 180 patents were randomly selected from two hospitals to build a prognostic prediction model based on the 53-gene signature using leave-p-out (one-third out) cross-validation method together with Cox regression and Kaplan-Meier analysis, and the model was assessed on three validation cohorts. FINDINGS:In the evaluation phase, studies based on TCGA-STAD showed that the 53-gene signature was significantly superior to other three prognostic signatures and was independent of TCGA molecular subtypes and clinical factors. For clinical validation and utility, the prognostic scores were generated using the newly developed assay, which was reliable and sensitive, in 100 sampling training sets and were significantly associated with OS in 100 sampling validation sets. The scores were significantly associated with OS in three independent and combined validation cohorts, and in patients with stages II and III/IV. The multivariate Cox regression demonstrated that the prognostic power of the score was independent of clinical factors, consistent with those findings in the TCGA dataset. Finally, patients with good prognostic scores exhibited significantly a better 5-year OS rate from adjuvant FOLFOX chemotherapy after surgery than from other chemotherapies. INTERPRETATION:The 53-gene prognostic score system is clinically applicable for predicting the OS of patients independent of clinical factors in gastric cancers, which could also be a promising predictive biomarker for FOLFOX regimen.
