Dissolved Inorganic Carbon-Accumulating Complexes from Autotrophic Bacteria from Extreme Environments

To facilitate the synthesis of biomass from CO 2 , autotrophic organisms use a variety of mechanisms to increase intracellular DIC concentrations. A novel type of multi-subunit complex has recently been described, which has been shown to generate measurably elevated intracellular DIC concentrations in three species of bacteria, raising the question of whether these complexes share this capability across the 17 phyla of Bacteria and Archaea where they are found.

Aerobic bacterial methane synthesis

Significance Observations summarized herein contribute to an ongoing paradigm shift in microbial ecology, documenting an emergent property of ecosystem function that further challenges the perception that biogenic methane (CH 4 ) production is strictly an anaerobic process. Relevant metabolites, a model bacterial isolate, gene, and enzyme are identified, and we show how this property can conceivably be broadly distributed in the biosphere and contribute to global CH 4 emissions. Scientifically, this study will enable lines of investigation that will expand our understanding of CH 4 synthesis and emission in nature and illustrates how CH 4 synthesis may actually serve as a nexus for the C and N cycles in nature.

An Integrated Computer-Aided Design and Manufacturing Workflow for Synthetic Biology

Biological computer-aided design and manufacturing (bioCAD/CAM) tools facilitate the design and build processes of engineering biological systems using iterative design-build-test-learn (DBTL) cycles. In this book chapter, we highlight some of the bioCAD/CAM tools developed and used at the US Department of Energy (DOE) Joint Genome Institute (JGI), Joint BioEnergy Institute (JBEI), and Agile BioFoundry (ABF). We demonstrate the use of these bioCAD/CAM tools on a common workflow for designing and building a multigene pathway in a hierarchical fashion. Additionally, each tool presented in this book chapter is specifically tailored to support one or more specific steps in a workflow, can be integrated with the others into design and build workflows, and can be deployed at academic, government, or commercial entities.
