Nutrient requirements and other factors involved in the culture of human kidney cells on microcarrier beads

The culture of human kidney cells on microcarrier beads in the Bioprocessing Laboratory at the Johnson Space Center is described. These were the first series of studies performed before and during 1983 to determine optimum conditions, including medium type, bead type and density. The composition of several medium types and the molecular weights of some common culture medium supplements and cellular proteins are included. The microgravity cell-to-bead attachment experiment performed on Space Transportation System Flight 8 is described.

Cell culture experiments planned for the space bioreactor

Culturing of cells in a pilot-scale bioreactor remains to be done in microgravity. An approach is presented based on several studies of cell culture systems. Previous and current cell culture research in microgravity which is specifically directed towards development of a space bioprocess is described. Cell culture experiments planned for a microgravity sciences mission are described in abstract form.

Bioseparations - Flight experiment support research

While important principles of electrokinetic technology, such as electrophoresis, have been demonstrated for microgravity environments and promise to be able to alleviate many of the practical limitations imposed by gravity, the minimal crew time and sample-handling facilities available in orbit call for almost total reliance on postflight analysis of separated fractions. Since cell separations are the most difficult to accomplish in 1-g, two model cell types have been studied intensively to determine the basic relationships between cell surface charge and secretory functions. A rationale has been developed for selecting and qualifying new cell types and proteins that warrant processing in microgravity.

Electrophoretic separation and analysis of living cells from solid tissues by several methods - Human embryonic kidney cell cultures as a model

The use of free-fluid electrophoresis methods to separate tissue cells having a specific function is discussed. It is shown that cells suspended by trypsinization from cultures of human embryonic kidney are electrophoretically heterogeneous and tolerate a wide range of electrophoresis buffers and conditions without significant attenuation of function. Moreover, these cells do not separate electrophoretically on the basis of size or cell position alone and can be separated according to their ability to give rise to progeny that produce specific plasminogen activators.
