Measurement report: Vertically resolved atmospheric properties observed over the Southern Great Plains with the ArcticShark uncrewed aerial system

Abstract. This study presents the unique capability of the Department of Energy (DOE) ArcticShark – a mid-size fixed-wing uncrewed aerial system (UAS) – for measuring vertically resolved atmospheric properties over the Southern Great Plains (SGP) of the United States. Focusing on atmospheric states, such as ambient temperature, wind, and aerosol properties, we overview measurements from 32 research flights (∼ 97 flight hours) in 2023. The August operations, aided by a visual observer on a chase plane, allowed for extensive UAS coverage, surpassing typical UAS operation envelopes. Our data from March, June, and August 2023 reveal distinctive seasonal patterns within the atmospheric column through unique chemical composition measurements. In situ measurements combined with remote sensing retrievals and radiosonde measurements provided valuable insights into their consistency and complementarity. Furthermore, we demonstrate the capabilities of the ArcticShark through several case studies, including the analyses of correlations between UAS-derived atmospheric profiles and conventional radiosonde measurements, as well as the derivation of vertically resolved profiles of aerosol chemical, optical, and microphysical properties. These case studies highlight the versatility of the ArcticShark UAS as a powerful tool for comprehensive atmospheric research, effectively bridging data gaps and enhancing our understanding of vertical atmospheric structures in the region.

High-Resolution Image Products Acquired from Mid-Sized Uncrewed Aerial Systems for Land–Atmosphere Studies

We assess the viability of deploying commercially available multispectral and thermal imagers designed for integration on small uncrewed aerial systems (sUASs, <25 kg) on a mid-size Group-3-classification UAS (weight: 25–600 kg, maximum altitude: 5486 m MSL, maximum speed: 128 m/s) for the purpose of collecting a higher spatial resolution dataset that can be used for evaluating the surface energy budget and effects of surface heterogeneity on atmospheric processes than those datasets traditionally collected by instrumentation deployed on satellites and eddy covariance towers. A MicaSense Altum multispectral imager was deployed on two very similar mid-sized UASs operated by the Atmospheric Radiation Measurement (ARM) Aviation Facility. This paper evaluates the effects of flight on imaging systems mounted on UASs flying at higher altitudes and faster speeds for extended durations. We assess optimal calibration methods, acquisition rates, and flight plans for maximizing land surface area measurements. We developed, in-house, an automated workflow to correct the raw image frames and produce final data products, which we assess against known spectral ground targets and independent sources. We intend this manuscript to be used as a reference for collecting similar datasets in the future and for the datasets described within this manuscript to be used as launching points for future research.

Modes of strain accommodation in Cu-Nb multilayered thin film on indentation and cyclic shear

Two-phase layered thin films with a high density of semi-coherent interfaces exhibit excellent mechanical properties and thermal stability. Here, in this study, a magnetron-sputtered Cu-Nb dual-layered thin film (~500 nm for Cu and ~150 nm for Nb) having an amorphous interface between Cu and Nb with a high density of aligned growth twins in Cu is subjected to severe surface deformation. The material is loaded using indentation and cyclic shear under tribological testing. The strain accommodation in the subsurface microstructure after deformation varies based on the local structure and deformation mode. Grain refinement and crack formations in the stressed region of the Nb layer and localized crystallization of the amorphous interface are observed after indentation and scratch testing. Pronounced detwinning of growth twins in the Cu layer under the cyclic shear strain leaves large dislocations sites and loops which are observed both by high-resolution transmission electron microscopy and experiment-guided molecular dynamic (MD) simulations. Our simulations provided insights into understanding the pathway for the detwinning process under cyclic shear loading.

High-throughput and high-efficiency sample preparation for single-cell proteomics using a nested nanowell chip

Abstract Global quantification of protein abundances in single cells could provide direct information on cellular phenotypes and complement transcriptomics measurements. However, single-cell proteomics is still immature and confronts many technical challenges. Herein we describe a nested nanoPOTS (N2) chip to improve protein recovery, operation robustness, and processing throughput for isobaric-labeling-based scProteomics workflow. The N2 chip reduces reaction volume to <30 nL and increases capacity to >240 single cells on a single microchip. The tandem mass tag (TMT) pooling step is simplified by adding a microliter droplet on the nested nanowells to combine labeled single-cell samples. In the analysis of ~100 individual cells from three different cell lines, we demonstrate that the N2 chip-based scProteomics platform can robustly quantify ~1500 proteins and reveal membrane protein markers. Our analyses also reveal low protein abundance variations, suggesting the single-cell proteome profiles are highly stable for the cells cultured under identical conditions.

High-temperature NMR MAS probe with optimized temperature gradient across sample rotor

A rotor housing assembly for NMR spectroscopy. An elongate rotor has a distal drive end, a proximal end and an internal sample space positioned along its length between the drive and proximal ends. The rotor is driveable about a rotation axis by a drive gas flow. A rotor housing has an interior space in which the rotor is at least partially received. At least one first heated gas flow inlet is positioned opposite the internal sample space, through which a first heated gas flow is controllably flowable into the interior space to heat it and the rotor. At least a pair of spaced apart second heated gas flow outlets are axially spaced from the first heated gas flow inlet to controllably convey a second heated gas flow to heat distal and proximal areas of the sample space to minimize a temperature gradient extending axially within the sample space.
