The Efficacy and Safety of an Intra-articular Dual-Acting Antibacterial Agent (TNP-2092) for Implant Infection–Associated Methicillin-Resistant Staphylococcus aureus

Abstract Owing to the presence of microbial biofilm on the implant, the eradication of biofilm-associated infections poses a challenge for antibiotic therapies. The study aimed to investigate the efficacy and safety of the novel antibiotic agent TNP-2092 in the context of implant infections. In vivo, rats with periprosthetic joint infection (PJI) treated with antibiotics showed an increase in body weight and decrease in swelling, temperature, and width of knee, compared with the control group. Meanwhile, inflammatory markers in synovium and serum were decreased in the TNP-2092 group, consistent with the pathological results. Moreover, TNP-2092 was effective in eliminating bacteria and disruption biofilm formation, and further alleviated the abnormal bone absorption and reactive bone changes around the prosthesis. In conclusion, intra-articular injection of TNP-2092 is safe and effective in treating knee PJI in a rat model. The study provides a foundation for the future utilization of TNP-2092 in the management of implant-related infections.

Unnamed Pt(Cu 0.67 Sn 0.33 ) from the Bolshoy Khailyk River, Western Sayans, Russia, and a Review of Related Compounds and Solid Solutions

We describe a potentially new species of a platinum cupride–stannide mineral (PCSM) of composition Pt(Cu 0.67 Sn 0.33 ). It occurs in a placer deposit in the River Bolshoy Khailyk, southern Krasnoyarskiy kray, Russia. A synthetic equivalent of PCSM was obtained and characterized. The PCSM occurs as anhedral or subhedral grains up to 15 μm × 30 μm in association with various platinum-group minerals, Rh–Co-rich pentlandite and magnetite, all hosted by a placer grain of Cu–Au–Pt alloy. Synchrotron micro-Laue diffraction studies indicate that the PCSM mineral is tetragonal and belongs to the inferred space-group P4/mmm (#123). Its unit-cell parameters are a = 2.838 (3) Å, c = 3.650 (4) Å, and V = 29.40 (10) Å 3 , and Z = 1. The c:a ratio calculated from the unit-cell parameters is 1.286. These characteristics are in good agreement with those obtained for specimens of synthetic Pt(Cu 0.67 Sn 0.33 ). A review on related minerals and unnamed phases is provided to outline compositional variations and extents of solid solutions in the relevant systems PtNi–PtFe–PtCu, PdCu–PdHg–PdAu, PdHg–PtHg, and AuCu–PtCu. The PCSM-bearing mineralization appears to be related genetically with an ophiolitic source-rock of the Aktovrakskiy complex of the western Sayans. The unnamed phase likely crystallized from microvolumes of a highly fractionated melt rich in Cu and Sn.

Fleetite, Cu 2 RhIrSb 2 , a New Species of Platinum-Group Mineral from the Miass Placer Zone, Southern Urals, Russia

Fleetite, Cu 2 RhIrSb 2 , a new species of platinum-group mineral (PGM), was discovered intergrown with an Os–Ir–Ru alloy in the Miass Placer Zone (Au–PGE), southern Urals, Russia. A single grain 50 μm across was found. Osmium, ruthenium, and iridium are the main associated minerals; also present are Pt–Fe alloys, laurite, Sb-rich irarsite, Rh-rich tolovkite, kashinite, anduoite, ferronickelplatinum, heazlewoodite, PGE-bearing pentlandite and digenite, as well as micrometric inclusions of forsterite (Fo 93.7 ), chromite–magnesiochromite, and Mg-rich edenite. In reflected light, fleetite is light gray; it is opaque, isotropic, non-pleochroic, and non-bireflectant. We report reflectance values measured in air. A mean of seven point-analyses (wavelength-dispersive spectrometry) gave Cu 13.93, Ni 8.60, Fe 0.10, Ir 28.07, Rh 7.91, Ru 1.96, Sb 39.28, total 99.85 wt.%, corresponding to (Cu 1.41 Ni 0.58 Fe 0.01 ) Σ2.00 (Rh 0.49 Ni 0.36 Ru 0.12 ) Σ0.97 Ir 0.95 Sb 2.08 on the basis of six atoms per formula unit, taking into account the structural results. The calculated density is 10.83 g/cm 3 . Single-crystal X-ray studies show that fleetite is cubic, space group Fdm (#227), a = 11.6682(8) Å, V = 1588.59(19) Å 3 , and Z = 16. A least-squares refinement of X-ray powder-diffraction data gave a = 11.6575(5) Å and V = 1584.22(19) Å 3 . The strongest five reflections in the powder pattern [d in Å(I)(hkl)] are: 6.70(75)(111), 4.13(100)(220), 3.52(30)(311), 2.380(50)(422), 2.064(40)(440). Results of synchrotron micro-Laue diffraction experiments are consistent [a = 11.66(2) Å]. The crystal structure of fleetite was solved and refined to R = 0.0340 based upon 153 reflections with F o > 4σ(F o ). It is isotypic with Pd 11 Bi 2 Se 2 and best described as intermetallic, with all metal atoms in 12-fold coordination. Fleetite and other late exotic phases were formed by reaction of the associated alloy phases with a fluid phase enriched in Sb, As, and S in circulation in the cooling ophiolite source-rock. The mineral is named after Michael E. Fleet (1938–2017) in recognition of his significant contributions to the Earth Sciences.

Oxidative stress induced apoptosis mediated anticancer activity of Rhus typhina fruits extract in human colon cancer

The present work was tested the anti-proliferation activity of Rhus typhina fruits extract (RTFE) in HEK293 and HT-29 cells, and studied their underlying mechanisms through the analysis of oxidative stress and apoptosis-related genes expressions by flow cytometer and RT-qPCR. In addition, the bioactive constituents were identified from RTFE by chromatography and NMR analysis. Among the various type of the fractions from the RTFE, the ethyl acetate fraction (EAF) was showed the potent anti proliferation effect in HT-29 cells. Thus EAF was selected for the further extensive analysis. Cytotoxcity and RT-qPCR results revealed that the treatment of EAF significantly inhibit the growth of the HT-29 cells by the up regulating of Bax and down regulation of Bcl-2, Survivn, AIF and SOD-2 gene expressions in time and dose dependent manner. Followed by sub G1 accumulation and the G0/G1 phase arrest, ROS production, and mitochondrial transmembrane potential (ΔΨm). Finally, the active anti-proliferations agents such as luteolin and luteolin-7-O-glucuronide were identified from EAF by NMR. This work suggests that EAF could be a functional food to treat the chronic diseases.

Platiniferous Tetra-Auricupride: A Case Study from the Bolshoy Khailyk Placer Deposit, Western Sayans, Russia

Tetra-auricupride, ideally AuCu, represents the only species showing the coexistence of Au with an elevated level of Pt, as in the case of a detrital grain studied structurally for the first time, from an ophiolite-associated placer at Bolshoy Khailyk, western Sayans, Russia. We infer that tetra-auricupride can incorporate as much as ~30 mol. % of a “PtCu” component, apparently without significant modification of the unit cell. The unit-cell parameters of platiniferous tetra-auricupride are: a 2.790(1) Å, c 3.641(4) Å, with c/a = 1.305, which are close to those reported for ordered AuCu(I) in the system Au–Cu, and close also to the cell parameters of tetraferroplatinum (PtFe), which both appear to crystallize in the same space group, P4/mmm. These intermetallic compounds and natural alloys are thus isostructural. The closeness of their structures presumably allows Pt to replace Au atoms so readily. The high extent of Cu + Au enrichment is considered to be a reflection of geochemical evolution and buildup in levels of the incompatible Cu and Au with subordinate Pt in a remaining volume of melt at low levels of fO 2 and fS 2 in the system.

Highly sensitive and accurate optical thermometer through Er doped tellurite glasses

Highlights: • Upconversion mechanisms of transparent Er{sup 3+} doped tellurite glasses are discussed. • Sensitivity of the thermometer based on the FIR technique increases monotonously with the decreasing doping concentration. • Highly sensitive and accurate optical thermometer is achieved using low doping sample as the probe and high power laser as the pumping source. - Abstract: We investigate the thermal effects of Er{sup 3+} doped tellurite glasses on the upconversion properties. It is found that the sensitivity of the thermometer based on the FIR technique increases monotonously with the decreasing doping concentration. However, low doping leads to weak upconversion intensity and further decrease the sensing accuracy, which makes it difficult to achieve the high sensitivity and accuracy simultaneously. Influences of the irradiating laser power on the sensing behaviors were studied in the purpose of enhancing the signal intensity. It is found that the heating effect of incident laser (up to hundreds of mW) can be neglected, indicating the feasibility of using high laser power and low doping sample to produce both high sensitivity and high accuracy. In addition, this paper respectively discusses the mechanisms of the upconversion processes and the concentration dependent sensitivity.

Liebermannite, KAlSi3 O8 , a new shock-metamorphic, high-pressure mineral from the Zagami Martian meteorite

© The Meteoritical Society, 2017. In this paper, we discuss the occurrence of liebermannite (IMA 2013-128), KAlSi3O8, a new, shock-generated, high-pressure tetragonal hollandite-type structure silicate mineral, in the Zagami basaltic shergottite meteorite. Liebermannite crystallizes in space group I4/m with Z = 2, cell dimensions of a = 9.15 ± 0.14 (1σ) Å, c = 2.74 ± 0.13 Å, and a cell volume of 229 ± 19 Å3 (for the type material), as revealed by synchrotron diffraction. In Zagami, liebermannite likely formed via solid-state transformation of primary igneous K-feldspar during an impact event that achieved pressures of ~20 GPa or more. The mineral name is in honor of Robert C. Liebermann, a high-pressure mineral physicist at Stony Brook University, New York, USA.
