Engineering Methanogenic Microbiomes to Redirect Flux to Biomass

In this study, we present a method for acquiring and characterizing novel microbial consortia that regulates methanogens and methanotrophs through selective cultivation and metagenomic analysis of indigenous microorganisms in the environment. In addition, we present the work performed as part of this project to model the pathways that act as limiting factors in microbial methane metabolism based on a carbon cycle model. In this report, we describe the methods for selective cultivation of methane-metabolism-related microorganisms from environmental samples, the method for monitoring their methane consumption performance, and the method and results for verifying their functions using quantitative PCR and metagenomics techniques. The microbial consortia containing methanotrophs were obtained through selective cultivation and molecular biological verification, and their methane consumption performance was evaluated. In addition, the potential of the existence of bacteriophages interacting with methane metabolism-related microorganisms was identified through metagenomic sequencing.

Crop protection in algae by exogenous terpene expression

The present disclosure relates to terpene synthases capable of producing terpenoids. In one instance, a transformed organism can include terpene synthases or vectors encoding such synthases. One method of employing such synthases and organisms includes protecting an algal culture, in which the produced terpenoid can act as a biocide.

Production of fusel lactates via biocatalysis

Microbial enzymes are used for esterification of biomass-derived substrates for production of industrially valuable esters. E. coli was used as an efficient platform for recombinant synthesis of fusel lactates such as the green solvent ethyl lactate.

Engineering RuBisCo for food safety

A method of improving a ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBisCo) to have a higher protein score is disclosed. The method includes the steps of: making a modified RbcL of the RuBisCo, by, on an RbcL unit of the RuBisCo, either substituting Met for Leu, Phe, Val, or Ile or combinations thereof; substituting Lys for Arg, Thr, or His or combinations thereof; or both of these substitutions. The modified RbcL consequently modifies the RuBisCo and is added to a biomass host where it is stable for homologous recombination. Plastid and nucleus integration was observed. Example RbcL sequences are disclosed with the desirable substitutions. The improved RuBisCo can be used as an improved proteinaceous food source for humans and animals.

Alkyl dialkoxyalkanoates as bioderived, high cetane diesel fuels

A fuel for an internal combustion engine includes a C 5 to C 30 dialkoxyalkanoate corresponding to formula (I), wherein the R 1 group is —H or a —CH 3 group, the R 2 and R 2 ′ groups are alkyl groups independently selected to have 1 to 9 carbon atoms; and the R 3 group is selected to have 1 to 9 carbon atoms. The compounds described herein may be used as neat fuels or mixed fuels (with diesel, biodiesel, jet fuel, marine fuel or other fuel compounds) in autoignition or spark ignition engines, such as diesel engines, gasoline (spark ignition) engines, MCCI, Homogeneous Charge Compression Ignition (HCCI) engines, or more generally in Low-Temperature Gasoline Combustion (LTGC) engines (using gasoline-like fuels), that have the high-efficiency advantages of HCCI but can operate with some level of charge inhomogeneities. Methods of making these compounds are environmentally friendly and can be done in a continuous manner.
